Abstract:Objective To establish a PCR method for detection of Feline panleukopenia virus ( FPV) in cats. Method The primers were designed and synthesized according to the published FPV specific sequences of VP2 gene. PCR method was established,and the specificity,sensitivity and stability of this method were tested. Then the method was used to detect 33 clinical samples of cats. Result The developed PCR method was no cross reaction with Feline herpes virus 1 ( FHV-1 ),Feline coronavirus ( FeCV),Feline syncytium-forming virus (FeSFV),Feline immunodefiency virus(FIV). Its minimum detection limit (the lowest detection virus titer) was 5lgTCID50 /mL,the corresponding template DNA concentration was 4. 9 x 102 copies. FPV DNA,maintained at - 30 ℃ for 12 months,was still able to amplify the fragment of about 301 bp. The 33 cats were 21 positive ,and 11 negative for FPV detected by PCR. Conclusion The developed PCR method is good in specificity,sensitivity, stability,and can be used for detecting the FPV in cat.

Abstract:Objective To establish a novel rat spinal cord injury model so as to furnish a suitable animal model for clinical research of bilateral spinal cord injury at different planes. Method Forty-five Sprague-Dawley rats were randomly divided into bilateral spinal cord hemi-section group ( group A,n = 15),spinal cord transection group (group B,n = 15 ) and sham-operation group ( group C,n = 15 ). After operation,the activities,ingesta, excretion,wound infection,death and other complications of rats in different groups were observed. Besides,BBB locomotive function scores and inclined plane test were performed to evaluate rats’behavioral function. Result After operation,the activities and ingesta were decreased in group A. Meanwhile,53. 33% of rats suffered from the uroschesis and 40. 00% died in the course of 28 - days’evaluation. In group B,The activities and injesta were obviously decreased,66. 67% of rats suffered from the uroschesis and 60. 00% died in 4 weeks post-injury. In group C,whereas,rats’activities and ingsta did not decrease as well as no complications and death occurred. The mean scores of BBB and maximum angle of inclined plane in group A and B were significantly lower than that in group C (P < 0. 05),but these parameters had no remarkable difference between group A and group B. Conclusion The spinal cord injury model with bilateral hemi-sections at different levels possesses the distinctions of lower incidence of uroschesis and mortality,and has not significant difference compared with the traditional spinal cord trans-ection model in behavioral evaluation. Thus,it is a suitable model for clinical research on bilateral spinal cord injury at different levels.

Abstract: Objective To investigate the effects and underlying mechanisms of Apelin-13 injected by intarcerebroventricular on cerebral ischemia-reperfusion injury in middle cerebral artery occlusive (MCAO) rats. Method Focal cerebral ischemia in rats was established by 2 h occlusion of the middle cerebral artery and 24 h reperfusion. After 2h ischemia,Apelin-13 and NS were administrated intracerebroventricularly. The neurologic deficient scores were investigated according to Zea-Longa’s Standard. The infarct areas were assessed with software Imagepro Plus 6. 0 after TTC staining. The activities of inducible nitric oxide synthase ( iNOS) and glutathione peroxidase(GSH-PX)were determined by specific kit. And the levels of Bcl-2、Bax and Caspase-3 in central neutral system were measured by western blot. Result Apelin-13(1. 0 μg·kg - 1 ) could decrease the neurologic deficient score,reduce the infarct areas of the brain in CIRI rats,depressed the activity of iNOS,enhance the activity of GSH-PX,increase the expression of Bcl-2,and decrease the expression of Bax and Caspase-3. Conclusion Apelin-13 can protect the ischemia-reperfusion injury,the mechanism may be related to depressing the level of NO, getting rid of more ROS,adjusting the expression of apoptotic and anti-apoptotic proteins.

Abstract:Objective To observe the protective effects of Bifendate and Oleanolic acid on acute hepatic injury induced by ANIT in olive oil in mice. Method At first,Bifendate and Oleanolic acid were taken by mice and rats. And then 100 mg /kg and 80 mg /kg ANIT were injected into abdominal cavity of mice and rats to damage their liver,respectively. After 48 hours,their fasting serum levels of ALT,AST,TBIL,DBIL,GGT,ALP were measured. The animals were sacrificed and dissected,and the wet weight of their liver and spleen as well were measured. Finally the viscera index was calculated. Result Oleanolic acid significantly reduced the serum ALT, AST and TB in mice,but Bifendate had no liver protection capacity. Bifendate significantly reduced the serum ALT,AST and ALP in rat,but Oleanolic acid had no liver protection effect. Conclusion Oleanolic acid has liver protection effect on liver injury induced by ANIT in olive oil in mice,and Bifendate has liver protection effect on liver injury induced by ANIT in olive oil in rat.

Abstract:Objective To screen the infection of parasites and viruses,including Sendai virus ( SeV)、Mouse hepatitis virus (MHV)、Lymphocytic choriomeningitis virus ( LCMV),in 90 laboratory mice and 45 laboratory guinea pigs of Kunming area. Method The specimen of 90 mice and 45 guinea pigs were selected from 3 different company randomlies. According to the description of experimental monitor methods of national standard and the protocol of virus assay kit ,parasites and viruses (SeV and MHV of mice ,LCMV and SeV of guinea pigs) of 90 laboratory mice and 45 laboratory guinea pigs were investigated. Result Ectoparasite was detected from 3 animals in 90 mice,detection rate is 3. 3% . Endoparasite was detected from 20 animals in 90 mice,detection rate is 22. 2% . Ectoparasite was detected from 4 animals in 45 guinea pigs,detection rate is 8. 9% . Endoparasite was detected from 9 animals in 45 guinea pigs ,detection rate is 20. 0% ,SeV of mouse and MHV were not detected, LCMV of guinea pigs and SeV of guinea pigs were not detected. Conclusion The situation of virus infection control is quite good,but both ectoparasite and endoparasite infection is relatively common in Kunming area. The works of quality control of laboratory animals should be improved in Kunming area. The quality monitoring of laboratory animals is very important in the biomedical research.

Abstract:Objective To analyze the effects of different types of endurance training on oxidative stress of myocardial tissue in Sprague-Dawley rats,and thereby to provide rational movement scheme to improve the myocardial antioxidant capacity. Method A total of 48 male SD rats (6 weeks) were enrolled in the study. The study consisted of control group (n = 12),low intensity exercise group (n = 12),moderate intensity exercise group (n = 12) and highly intensity exercise group ( n = 12). The endurance exercise training groups exercised for 6 weeks and the control group maintained their daily eating habits. superoxide dismutase (SOD) activity,glutathione peroxidase (GSH - Px) activity,malondialdehyde (MDA) content were measured after the 6 weeks experiment. Result After 6 weeks endurance exercise training intervention,compared with control group,the SOD activity were higher in moderate intensity exercise group (P < 0. 01),and significantly higher in highly intensity exercise group (P < 0. 01);the GSH - Px activity were higher in low and moderate intensity exercise group (P < 0. 05 or P < 0. 01),and significantly higher in highly intensity exercise group (P < 0. 01). Moreover,the levels of MDA significantly enhanced in highly intensity exercise group (P < 0. 01). Conclusion The low and moderate intensity exercise group were effective in improve the myocardial antioxidant capacity of rats,particularly in moderate intensity exercise group.

Abstract:Currently,except surgical treatment,we have not found a reliable method of treatment. CTLA - 4 and PD - 1 are two kinds of protein which are produced by T cells. They can puts the brakes on T cells,preventing them from launching full-out immune attacks. By using the monoclonal antibodies of these two protein,the blockerthe CTLA - 4 molecule can be blocked and set the immune system free to destroy cancer. In addition,a personalized treatment that involves genetically modifying a patient’s T cells can make them target tumor cells. It can be used in cancer treatment and is very potential.
