    Ras /MAPK /ERK Promote Expression of CyclinD1 Collaborating with NF-κB in Hepatoma Cells

    Abstract: Objective To investigate the regulatory mechanisms contributing to over-expressed cyclinD1 in Hras12V induced hepatocellular carcinoma. Method The liver tumor tissues and peri-tumor tissues of 9-month-old male H-ras12V transgenic mice and normal liver tissues of male C57BL /6J mice were sampled. Part of each sample was fixed in 10% formalin and undergone paraffin-embedded tissue section,HE staining,and pathological analysis. Part of each sample was frozen in liquid nitrogen for extraction of total RNA and protein. miRNA expression was analyzed by High-Throughput Sequencing. The cyclin D1 and miR-5102 gene expression were examined by qRT-PCR. The protein levels of cyclin D1,p-ERK,ERK,and IκB were detected by Western blot. Result Compared with the normal liver tissues and peri-tumor tissues,the mRNA and protein expression levels of cyclin D1 in tumor tissues was significantly increased ( P < 0. 05) ; The protein and phosphorylation levels of signaling molecule ERK were significantly increased; And the protein level of inhibitor IκB were significantly reduced. The expression level of miR-5102 did not change significantly among the three sample groups. Conclusion In the process of H-ras12V-induced liver tumorigenesis,activation of MAPK /ERK and NF-κB signaling pathways contribute to the over-expression of cyclin D1.

    Abstract: Objective To study the inhibiting effect of glaucescent fissistigma root crystals in the nude mice with cervical adenocarcinoma; Method Using cervical adenocarcinoma inoculated to the nude mice and growing the solid tumor,subcutaneous transplantation the 3th the solid tumor to establish nude mice model. Take glaucescent fissistigma root crystals ( I,II) from gastric for experimental,0. 5 mL /one,take water saline as control group ,one time per day,30 days in a row. Measuring the weight and tumor size 2 times in a week,after 30 days of treatment to put to death in mice,anatomize the mice and weighted tumor,calculate the tumor-inhibition rate. Evaluating the tumor-inhibit effection by glaucescent fissistigma root crystal Ⅰ and crystal Ⅱ in nude mice with cervical adenocarcinoma; Result Glaucescent fissistigma root crystalⅠ and crystalⅡ have affection to nude mice with cervical adenocarcinoma,the anti-tumor rate are 88. 95% and 66. 71% ,respectively. The tumor suppression of crystalⅠ have significant difference ( P<0.01) ,crystalⅡ ( P<0.05) ; Conclusion Glaucescent fissistigma root crystalⅠ and crystalⅡ can inhibit nude mice with cervical adenocarcinoma to growth.
    The Effect of High Salt and High Protein Diet on Blood Pressure and the Pathological Changes of Kidney and Gastrointestinal in Dah1 /SS Rats

    Abstract: Objective To explore the effect of high salt and high protein diet on blood pressure and the pathology changes of kidney and gastrointestinal in Dah1 / SS rats. Method SPF female Dah1 / SS rats were respectively randomly divided into three groups: control,high-salt and high-protein group,and were respectively fed to common low-salt diet,high-salt diet witch contain 8% sodium chloride and high-protein diet with 2. 4% methionine in the individual ventilated cages ( IVC) . Systolic blood pressure ( SBP) and body weight of rats was measured every week. At 8 weeks,the kidney,esophagus,stomach ,small intestine ,rectum,and pancreas tissues were stained with HE to observe the pathological changes. Result The rat body weight of high-salt and high-protein group were significantly lower than the control group,but the SBP of high-salt group was significantly higher than control and high-protein group. Renal impairment was observed in high-salt and high-protein group. Besides,the pathological changes of kidney were observe only in high-salt group. Also,the pathological changes of esophagus,stomach, small intestine,rectum,and pancreas tissues were not found. Conclusion The salt-sensitive hypertension model was successfully made by Dah1 / SS rats fed with high salt diet,and the varying degrees of damage of kidney with high salt and high protein diet,but no damage to gastrointestinal and pancreas.
    The Effect of Renal Adventitia Artery Ablation on Blood Pressure in Spontaneously Hypertensive Rats

    Abstract: Objective To study the effect of renal adventitia arter ablation ( RAAA) on blood pressure in spontaneously hypertensive rats. Method Male 16 week old spontaneously hypertensive rats 25,were randomly divided into two groups: renal adventitia artery radiofrequency ablation group ( RAAA group,n = 13) and sham operation group ( n = 12 ) . Using radio frequency ablation device of bilateral renal artery adventitia for circumferential ablation. Result After 1 weeks,RAAA group postoperative blood pressure was ( 120. 4 ± 14. 6 ) mmHg,decrease to a normal level of blood pressure,blood pressure decrease of 44. 6 mmHg,compared with preoperative P = 0. 00,statistically significant; after 3 weeks,blood pressure was ( 127. 9 ± 13) mmHg,blood pressure decrease of 37. 1 mmHg,compared with 1 weeks before surgery,and P = 0. 00,there was statistical significance of blood pressure. Sham operation group with time increased,but there was no significant change in blood pressure. Heart rate decreased in RAAA group,compared with sham group,22. 7 times / min,P = 0. 141 decreased at 1 weeks after operation,no statistical significance; after 2 weeks,3 weeks to further reduce,reduce the rate of 41. 2 and 60 times respectively,compared with the preoperative,P = 0. 009,P = 0. 000,the difference was significant. Sham operation group heart rate was decreased,but no statistical significance between the two groups. In RAAA group,the diameter of the renal artery and renal artery blood flow velocity after 3 weeks compared with the preoperative,no statistical difference; Conclusion Renal adventitia artery ablation can effectively reduce the blood pressure and heart rate in SHR,,on renal artery diameter and blood flow velocity has no obvious influence.

    Abstract: Objective Established normal reference range of hematology and growth indicators about male and female SPF outbred population Mongolian gerbils and provided a basic data for the using of Mongolia gerbils. Method The body weight,routine blood test and blood biochemical indices in different days and weeks of male and female Mongolian gerbils were measured. Result Mongolian gerbils growth curve in different ages,normal reference range of different sex gerbils physiological index value of blood in different days and ages were detected. Conclusion Difference ages takes greater impact on the growth of gerbils weight,most physiological and biochemical index of blood are affected by age and gender,so in scientific research the effects of gender and age on the animal blood physiology and biochemistry indicators should be fully considered,when the Mongolian gerbils are using in the science research.
    Establishment of Rabbit liver VX2 Tumor Model via Percutaneous Transhepatic Approach under DSA and CT Guidance

    Abstract: Objective To evaluate the establishment method of rabbit liver VX2 tumor model by injecting tumor tissue mass into the liver parenchyma via percutaneous transhepatic approach under DSA and CT guidance. Method Forty New Zealand rabbits were divided into A and B groups randomly with 20 rabbits each. The VX2 tumor tissue mass was implanted into rabbits liver guided through percutaneous transhepatic route guided by DSA and CT in group A and through epigastrium incision method in group B. On the second and third weeks after implantation, MR examination was performed and the rabbits were sacrificed to observe the tumor in situ and ectopia. Result In group A,the success rate of the liver VX2 tumor model was 90% with mean operation time of ( 17. 3 ± 5. 2) min and there was no episode of ascites and death with the exception of ectopia implantation in one case. In group B, successful establishment rate was 85% with average operation time as long as ( 17. 3 ± 5. 2) min and ascites and death were observed in three and two cases respectively. The difference of operation time between two groups was statistically significant ( P < 0. 05) . Conclusion The method of liver VX2 tumor implantation guided by DSA and CT via percutaneous transhepatic approach is simple with higher success rate and shorter operation time for making liver carcinoma model of rabbits.
    Laboratory Investigation of Sevoflurane Anesthesia in Laparoscopic Surgery of Miniature Pig

    Abstract: Objective To observe the anesthetic effect of sevoflurane in laparoscopic surgery of miniature pig. Method 30 miniature pigs were anesthesiaed with lumianning Ⅱand ketamine mixture intramuscular injection. Mechanical ventilation was started after intubation. Sevoflurane inhaled and intermittent intravenous vecuronium were used to maintain anesthesia during ventilation. The anesthetic effect was observed and analyzed. Result The success rate of anesthesia was 100% . Conclusion Anesthesia effect was good,anesthesia depth was controlled easy,vital signs were stable,muscle relaxant effect was good and animals have waked up easy in laparoscopic surgery of miniature pig when the use of sevoflurane.
    Investigation of the Correlation of Coat Color,Genotypes of Coat Color and Sex with the Training Success Rate of Guide Dogs

    Abstract: Objective To investigate the effect of coat color,genotypes of coat color and sex of Labrador retriever and Golden retriever on the training success rate of guide dogs. Method In order to analysis the correlation of the coat color or the genotypes of coat color with the training success rate of guide dogs,the polymorphism loci of coat color gene MC1R ( c.C916T) and TYRP1 ( c.C991T) were detected by using PCR / sequencing method,and then the genotypes were decided,for 45 Labrador retriever and 16 Golden retriever. In addition,the correlation between the sex and the training success rate of guide dogs was also analyzed by exploring the Guide dogs in China ( Dalian) Guide dog training center. Result The genotype of 16 ( 14 male,2 female) Golden retrievers ( coat color: all gold) was all eeBB. The genotypes of 45 ( 22male,23 female ) Labrador retrievers ( coat color: 23yellow, 22black) were eeBB,eeBb,EEBb,EeBB,EeBb,respectively. The results of chi-square correlation degree analysis showed that the coat color,and the genotypes of coat color,of Labrador retriever and Golden retriever had no significant correlation with the training success rate of guide dogs ( P > 0. 05) . In addition,the sex of Labrador retriever and Golden retriever also had no significant correlation with the training success rate of guide dogs ( P > 0. 05) . Conclusion The coat color,genotypes of coat color and sex have no significant correlation with the training success rate of guide dogs,and can not be used as the effective index to predict the training success rate of guide dogs.
    Comparison of Physical Properties between Aspen Shaving and Two Corncob Beddings under Working Condition

    Abstract: Objective To provide basic data on laboratory rodent bedding choice under working condition. Method The mouse cages were filled with the equal volumes ( 500 ml) of aspen shaving and corncob beddings in two particle size ( big or small) ,respectively. In the working condition complying with the National Standard GB 14925 - 2010,the mass and the physical properties of the three beddings,volumetric absorbency,evaporation rate,heat preservation,and dust particle production,were examined. Result The mass of aspen shaving was significantly lower ( P < 0. 001) than those of the two corncob beddings. In contrast,corncob beddings had the significantly higher ( P < 0. 001) absorbency when compared to aspen shaving bedding,and the absorbency of the small particle corncob was lower ( P < 0. 001) than that of the big one. Evaporation rate was found faster ( P < 0. 001) for aspen shaving than for the two corncob beddings,and there was no difference between the two corncob beddings. The aspen shaving generated significantly more ( P < 0. 001) dust particles than the two corncob beddings. Conclusion The parameters of physical properties are correlated with the material and shape of the beddings. The advantage and disadvantage of each bedding should be fully estimated when a facility chooses bedding for laboratory rodent husbandry.
    Exploration on Vacuo Draw Blood Method from Submandibular Vein Plexus of Mongolian gerbils

    Abstract: Objective To explore on the technology and method of vacuo draw blood from submandibular vein plexus of Mongolian gerbils,and improve the experiment success rate of Mongolian gerbil. Method Forty Mongolian gerbils were selected in this study. The Mongolian gerbil submandibular vein plexus was punctured with intravenous infusion needle after anaesthetized. Result The vein blood of Mongolian gerbil was successfully obtained,and the volume of blood sample from individual gerbil reached 0.3 - 0.5 mL. Conclusion This draw blood method is a simple and safe technology to obtain a considerable amount of blood. And it can be repeated for several times with mild injury and high-quality blood samples.

    Abstract: Objective The some problems during the establishment and application of the Beagle dog telemetry model were investigated,and introduce the measures to prevent such problems. Method The problems during establishment and application of the Beagle dog telemetry model have been investigated using 32 Beagle dogs. The reasons of such clinical abnormalities as wound infection,suture removal by the animal and folliculitis have been analyzed. Correspondingly,certain advices on how to prevent and treat these clinical abnormalities have been proposed. Result The clinical abnormalities during the establishment and application of the Beagle dog telemetry model have been analyzed and solved. Conclusion The Beagle dog telemetry model was successfully established and valuable information was provided for the application and maintenance of the telemetry model in the safety pharmacology evaluation in our country.

    Abstract: Objective To establish the bacterial endotoxins determination method for the vincristine sulfate. Method According to Chinese Pharmacopeia 2010 edition Vol Ⅱ Appendix Ⅺ E,the interference test and bacterial endotoxin test of 3 batches of vincristine sulfate were carried out,using tachypleus amebocyte from 2 different companies. Result With dilution method,maximum non-disturb concentration of vincristine sulfate for bacterial endotoxins test was 0. 00104 mg /mL for Zhanjiang A&C Biological Ltd and Bokang Marine Biological Co Ltd. With pH modulation method,after adding 1 mol /mL sodium hydroxide to adjust pH of the sample to 7. 0, maximum non-disturb concentration of vincristine sulfate for bacterial endotoxins test was 0. 00417mg /mL. Conclusion The method is suitable for the detection of vincristine sulfate.

    Abstract: Tuberculosis ( TB) is a chronic wasting disease caused by Mycobacterium tuberculosis( Mtb) ,ranking the first death of a serious infectious disease by a single pathogen. TB has a long history,which can be traced back to the ancient society. For centuries,humans are extremely hard and bitter struggle with TB. To date,however,TB is still a serious public health problem world widely. Thus,there is urgent need to establish a suitable animal model for anti-TB vaccine and drug development. Nonhuman primates are evolutionarily close and similar to humans in terms of their characteristics of anatomy,physiology,immunology and biochemistry. Application of nonhuman primates as animal models for the study of the pathogenesis of TB,immune response and related vaccines and drugs is crucial,even irreplaceable. In this paper,we reviewed the recent advances in application of rhesus macaque and cynomolgus macaque models in research on TB prevention and treatment.

Transgenic animal is a kind of laboratory animal,and it is widely used as its unique advantages,but it also brought new problems on animal welfare ethics. The laboratory animal welfare legislation plays an important role in regulating human behavior,and promoting the rational use of animal. The EU animal welfare legislation and regulations were concrete,and kept constantly updated. The EU regulation has the maximum effect on the other country. Study on the advanced experiences of the European Union will help us on the perfection of relevant legislation and fill the deficiency of transgenic animal welfare ethics regulations in China.
