Abstract: Minipigs are ideal animals for diabetes mellitus ( DM) research for its similarities with human beings on metabolism. However,during the long-term selection as domestic animals,pigs have developed the features resisted to suffer from DM. We have screened 245 boars and sows of Bama minipigs with fasting blood glucose ( FBG) ,and a high FBG pregnant pig was found. 11 piglets ( 10 survived) were born from the pig and as founders to selecting breeding. After 2 generations selection with susceptibilities to diet-induced type 2 DM,the inducing period from 7 months shortened to 6 months. It is speculated that a new line of Bama minipgs susceptible to dietinduced DM will be developed in the future.

Abstract: Objective Effects and gender difference of high-fat diet on rat body weight and serum lipid indexes are discussed in this research. Moreover,some experimental date were provided in order to improve the specificity and reproducibility of hyperlipemia models. Method SD rats were randomly divided into 4 groups considering sexuality and diets ( normal diet or high-fat diet) . All rats were fed for 12 weeks with given diets. Effects of highfat diets on serum total cholesterol ( TC) ,triglyceride ( TG) ,free fatty acid ( FFA) ,aspartate aminotransferase ( AST) and alanine aninotransferase ( ALT) were investigated. Result High-fat diet increased the body weight and food intake for male rats,however,body weight and food intake were decreased after high-fat diet feeding in female nate. High-fat diet expanded the gender difference on body weight. The serum contents of TC,TG,FFA, HDL-C,LDL-C in female rats were significantly increased in high-fat diet group. The serum contents of TC,FFA, HDL-C,LDL-C in male rats were also obviously increased,however,the serum TG content remain stable. Serum lipid index increased more apparently in female rats than male rats. Serum contents of AST,ALT for both sex were increased after high-fat diet feeding for 12 weeks,however,these increasing was considered as safety and controllable. Conclusion High-fat diet feeding for 12 weeks can lead to abnormal of lipid metabolism in both male and female rats with significant gender differences. Typical hypercholesterolemia ( Ⅱa type) with high body weight are observed in male rats,however,mixed hyperlipidemia ( Ⅱb type) with low body weight are founded in female rats. Gender difference should be concerned and considered in animal modeling of hyperlipidemia and therapy treatment.

Abstract: Objective To investigate the process of inactivation effect and scavenging ability of different virus, provide effective method for inactivating virus as the virus infection in animal products processing. Method laboratory select Newcastle disease virus,Human reoviruslike agent,Pseudorabies virus,Porcine Parvovirus as indicator virus,add to the selected material. In the removal process of oxalic acid and hydrochloric acid,lesion hole in the 96 hole plate,calculate the initial and after treatment virus titer of virus,summarize the process of inactivation. Result After treated with 10% oxalic acid 1. 5h,the virus titer decreased significantly,but not completely inactivated; continue treat with 10% hydrochloric acid 1h,the virus titer decreased to zero,virus completely inactivated. Conclusion acid treatment process can effectively remove the virus after inactivated treatment,achieve good disinfection effect,to guarantee the safety of the product and offer reference method of virus clearance of animal origin.

Abstract: Objective To breed and identify HO-1 gene knock-out mice. Method The HO-1 gene knock-out heterozygote mice were bred in specific pathogen free environment and copulated with wild genotype mice. The Genomic DNA of the off-springs was extracted and the HO-1 gene was specifically amplified by polymerase chain reaction. Using dideoxy-sequencing method the genotypes of off-springs present as three types: wild genotype, heterozygote genotype and homozygote genotype. Heterozygote genotype mice were copulated for further breeding, while the homozygote genotype mice were used for research. Result Breeding and reproducing were both successful,and we obtained heterozygote genotype and homozygote genotype mice with HO-1 gene knockout. Conclusion Correct methods of breeding,reproducing and identifying can effectively obtain HO-1 gene knock-out homozygote mice from heterozygote mice.

Abstract: Objective To establish an experimental model for reversible atelectasis of minipig. Method Twelve male China Experimental Miniature Pig( CEMP) were divided into two groups randomly: atelectasis group( A group) and reversible atelectasis group( R group) . Ni-Ti alloys occluders were placed in the lower right main bronchus of all animals with a bronchofiberscope after anesthesia. The occluders were pulled out by bronchofiberscope after atelectasis confirmed after three days in R group. The development of atelectasis/reexpansion was studied by computerized tomography scanning every three days after occluders were placed in or pulled out. All of the group A animals were sacrificed on the third day after established atelectasis,and CEMP of group R were killed at two weeks. Result Typical pathology of atelectasis and computerized tomography imaging were observed in group A, and all changes alleviated obviously in group R,tended to be normal. Conclusion CEMP could be used as an ideal model for reversible atelectasis created by placing in /pulling out Ni-Ti alloys occluder in the lower right main bronchus by bronchofiberscope.

Abstract: Objective Discussion on the Miao medicine( Arisaema rhizomatum) half bad impact on the liver and kidney histology and function in mice,and discussion an the toxicity of thie Miao medicine on liver and kidney in mice. Method Observing the function and pathological changes of liver and kidney by given some dry and fresh Arisaema rhizomatum on mice,which were used respectively to one-off delivery seven days and continuous dosing for thirty days. Result There was no effect on kidney morphology changes of acute toxicity experiments of Arisaema rhizomatum in mice,but liver cell edema,fresh group was more severity than dry group compared with control group,subacute toxicity experiments show that the levels of ALT,AST and Urea were no significant difference of dose group,but the level of Scr was rose,and dry group has significant difference( P < 0. 05) ,fresh group has extremely significant difference ( P < 0. 01) . Conclusion There has a certain impact of liver morphology with using Arisaema rhizomatum in mice continuous,but there has no damage to its function; no effect on morphology of renal,but has a certain damage on its function

Abstract: Objective To establish experimental method for SD rat placenta extract and cell culture so as to lay foundation for the placental cell research. Method Push 20 days of gestation SD rats to death to extract rat placentas and culture cells were isolated from active placental tissue,adopt inverted microscope to observe cell morphology. Result Placentas can be obtained by dissecting 20 days of gestation SD rats,placental cells adhered under the cells culture. Cell morphology was polygonal or fusiform,cells had defined margins and abundant cytoplasm. Nucleus was circular and elliptical. Conclusion Experimental method is feasible in this experiment. This experiment provide specific experimental method for the research of placental cells,and lay foundation for the placental cell research.

Abstract: Objective To investigate the operation,personnel allocation,service content and management situation of laboratory animal facility in medical colleges and universities. Method We made a questionnaire and investigated 10 laboratory animal centers of medical colleges and universities in China. The content included school's size,facility planning and operation,personnel allocation and source of funding,service content and management situation. Result Animal facilities in 40% units could not meet demand. On the fund resource of facility operation,about 50% units were operating by appropriation and self-financing. Service content have transformed from traditional animal feeding to experiment assistant and commission. 80% units could provide pathological technical services and 70% units could produce transgenic animals. Personnel dealing with laboratory animals must get a certificate in all units. 80% units managed animal experiments through principal investigator team. Conclusion Service and management level of laboratory animal center in medical colleges and universities were good in general. Hardware facilities still need to be strengthened.

Abstract: Objective To observe the changes of serum biochemical indexes after blood samples stored at room temperature for various time. Method After rats were anesthetized,blood was collected via abdominal aorta. The blood sample of each rat were divided to three parts and placed at room temperature for the 30 min,2 h and 4 h respectively. The biochemical parameters were detected and the changes were observed. Result After blood samples were placed at room temperature for 2 h and 4 h,serum glucose ( GLU) and creatine kinase ( CK) were significantly different from those of samples stored for 30 min. Conclusion Storage of whole blood at room temperature for a long time affects values of serum CK and GLU.

Abstract: Although Postpartum Depression belonging to one of the types of depression,as a complex mood disorders,it’s different from simple depression. During the special period of postpartum ,maternal hormone levels, content of neurotransmitters and neuropeptides,function of related receptor fluctuate dramatically and these fluctuations may contribute to the etiology of PPD. Because of the complexity of the physiological and pathological particularity makes big difference on the study of animal models between this disease and simple depression. Abstract: Monoclonal antibody is hybridoma cell which fuses myeloma cell with specific lymphocyte B induced by certain antigen with the help of polyethylene glycol. The hybridoma cell can be serial passages and steadily secrete specific antibody. Abstract: This paper describes the study of the origin and history of salt-sensitive rats,and analyzes the impact of changes in different age-related factors on blood pressure in salt-sensitive rats,including the impact of genetic factors in female offspring during pregnancy ( 21 days) of high salt intake,and comparing lactation ( 3 weeks after birth) ,pups or adolescence early ( 3 ~ 5 weeks) ,adolescence ( 6 ~ 8 weeks) and mature ( over 8 weeks) and other changes in blood pressure salt-sensitive rats differences after high salt intake,contrasting drug intervention ( thiazide diuretics,candesartan and eplerenone) for blood pressure levels of salt-sensitive rats in different age stages,and discussing the impact of potassium and calcium intake on blood pressure sensitive to the different stages of salt.
