    Study on Oviducts Embryo Transfer Technology in KM Mouse

    Objective To compare the effects of the different oviduct embryo transfer methods,the periods and numbers of embryo transfer.Methods The different numbers of 1-cell embryo,2-cell embryo and,8-cell embryo were respectively transfefred into the oviduct of pseudopregnancy KM mouse,and the embryos were transfer to the pavilion of the oviduct tranfer and the oviduct ampullar transfer,respectively.Result The pregnancy rate of 2-cell embryo transfer is significant higher than 1-cell and 8-cell treatment group(66.67% vs 25%,33.33%,P<0.05);Transfer of 25 2-cell embryos via oviduct tranfer showed a higher birth rate than 15 and 35 2-cell embryos(36% vs 16.67%,10.71%,P<0.05);The pregnancy rate and birth rate of oviduct ampullar incision transfer were significant higher than pavilion of the oviduct tranfer(50% vs 75%,38.9% vs 49.3%,P<0.05).Conclusion Transfer of 25 2-cell embryos via oviduct ampullar incision transfer can improve the success rate of mouse embryo transfer.

    Objective To study the effects of ethionine on lipid metabolism in mice,in order to provide a gist of decision-making for the time of drug administration for preventing fatty liver in mice.Methods Mice were divided to six groups.serum TG and TC or hepatic TG and TC were detected in normal group,and experimental group at the different time(6,12,24,36,48 h)after taking ethionine orally.Result Serum TG increased significantly during 6~48 h,and serum TC was increased significantly during 6~36 h after ethionine administration.Furthermore hepatic TG was increased significantly during 6~36 h,and hepatic TC was reduced significantly during 24~36 h after ethionine administration.Conclusion Ethionine-induced fatty liver in mice was stable during 6~36 h after administration.

    Objective To study the effect of Penyanjing granules on immunological function.Methods The effects of Penyanjing granuleson nonspecific cell and humoral immunity,phagocytic function of macrophage in abdominal cavity of mice with low immune function induced by cyclophosphamide,delayed hypersensitivity induced by DNCB,and production of hemolysin antibody in mice with low immune function induced by cyclophosphamide were observed.Result Penyanjing granules could obviously strengthen phagocytic function of macrophage in abdominal cavity of mice with low immune function induced by cyclophosphamide,and induce the delay hypersensitivity induced by DNCB.But Penyanjing granules had no effect on production of hemolysin antibody in mice with low immune function leaded by cyclophosphamide.Conclusion Penyanjing granules can strengthen function of non-pecific and cell immunity.

    Objective Through the special pharmacodynamics tracking test to determine the optimum extraction process.Methods The effect on mice hypnosis of pentobarbital sodium of subthreshold dosage and the hypnotic time experiment on mice by pentobarbital sodium in threshold dosage were used to evaluate the optimum process.Result The results showed that Anmian preparation No.4 drug group could enhance the number of asleep mice of pentobarbital sodium in subthreshold hypnotic dosage.Compared with the blank control group there was significant difference(P <0.05).Anmian preparation No.4 drug group also could shorten substantially the rats' sleep latent period and prolong obviously the rats' sleep time which induced by pentobarbital sodium at threshold dosage.Compared with the blank control group there was significant difference(P <0.05).Conclusion Through the observations of the 2 experiments,Anmian preparation No.4 drug group and the blank control group have significant difference.The optimum extraction process was as follows:The volatile oil of one of the drugs was extracted firstly,the residue with the other 2 drugs refluxing and extracting with water,filtering.The combined filtrate was concentrated to about 200 mL and then added the volatile oil.The Preparation made by this process could enhance the number of asleep mice.The Preparation also could shorten substantially the rats' sleep latent period and prolong the rats' sleeping time,suggesting that the process was the optimal process.

    Objective To isolate,culture and purify rat bone marrow mesenchymal stem cells(MSCs) and observe the effect of salvianolic acid B on vascular endothelial growth factor(VEGF),stem cells factor(SCF),the cardiac muscle specificity transcription factor NKX2.5 and GATA-4 expressing of MSCs in vitro.Methods MSCs were cultured in five groups of lower glucose DMEM media containing different concentration of Salvianolic acid B as follows:group of control(add nothing),four goups of Salvianolic acid B(the lower glucose DMEM media add 0.03、0.3、3、30 μg/mL Salvianolic acid B in turns).The expression of VEGF、SCF、NKX2.5 and GATA-4mRNA in the MSCs harvested at following 24 hour were analyzed with reverse transcription-polymerase chain reaction.Results The expression of VEGF、NKX2.5 and GATA-4mRNA from MSCs treated with Salvianolic acid B were increased significantly(Compared with the control group,P<0.01,P<0.05).Conclusion Salvianolic acid B could promote the secretion of VEGF and reinforce the expression of NKX2.5 and GATA-4 mRNA of MSCs.It is useful to establish the substantial foundation to the study on induction to different of MSCs.

    Objective To observe effects of Penyanjing granules on chronic pelvic infection of rats induced by Escherichia coli.Methids Using the chronic pelvic infection model rats induced by Escherichia coli were used,to observe the influence of Penyanjing granules on the model rats.Result The Penyanjing granules could improve chronic inflammatory disease of the blood stasis in rats,and significantly reduce the chronic inflammatory swelling of the uterus in E coli-induced animal model of pelvic inflammatory disease of rats,and reduce model the degree of endometrial lesions.Conclusion There exist remarkable therapeutic effects of Penyanjing granules on chronic pelvic infection of rats induced by Escherichia coli.

    Objective To establish an empirical method on the rabbit atherosclerotic plaque model by complex factors.Methods Japanese great ear albino rabbit were randomly divided into control group(8 animals) and experiment group(16 animals).Control group was fed with common animal diet,experiment group was fed with high fatty forage and injected with the bovine serum albumin fraction Ⅴ and their aortas were injured by balloon.After ten weeks,all the rabbits were sacrificed.Then,the serum MCP-1,TNF-α,IL-1 were measured,and the morphological characters of pathological changes of the carotids were observed,and thickening degree and area of the aorta endomembrane was measured.Result Compared with the normal control group,the serum MCP-1,TNF-α,IL-1,CRP,IT,IT/MT,IHI levels of experiment group stepped up significantly(P<0.05 or P<0.01) and rabbit atherosclerotic plaque with typical fibrous caps were formed.Besides,in model group there were correlation between VEGF mRNA and TNF-α、IHI、IT/MT(P<0.05).Conclusion The method of complex factors to establish model(high cholesterol diet,immunity harm and artery balloon injury),can be accurately controlled and easily repeated,and can induct rabbit atherosclerosis in a short time.

    Objective To investigate the cause of unusual pregnancy in rhesus macaques breeding colony.Methods Several factors relating to unusual pregnancy,such as infective pathogens(toxoplasma,Tox),environmental elements and other factors with the similar physiology and biological character in human,were analyzed.In this study,29 samples were collected from unusual pregnancy cases and Toxoplasma gondii DNA extracted from blood was examined,using normal delivery group and normal adult macaque group as controls.Results Statistical results showed that toxoplasma infective rate of the unusual pregnancy samples were 34.4%,significantly higher than those of the normal delivery group(12.5%,P<0.05) and the normal adult macaque group(5.0%,P<0.05).The unusual pregnancy rates were significantly different among macaques housed in distinct environments.Conclusion Toxoplasma infection,environment,maternal age and parity,are the factors relating to unusual pregnancy of rhesus macaques,which will be decreased according to our proposed measures.

    Objective To provide reference for laboratory animal production and management by analyzing of the recent three years test results on microbes and parasites of commonly used laboratory animals in Beijing area,which covered the conventional Beagle dogs,the conventional rabbits,the clean and over rats(strains of Wistar and SD) and mice(strains of KM and ICR).Methods The microbes and parasites of laboratory animal samples were tested according to the current national standards described in 《Laboratory animal—microbiological standards and monitoring》 and 《Laboratory animal—parasitic standards and monitoring》.Results The quality of conventional Beagle dogs was found to be improved gradually,but the exterior parasites had still been detected occasionally.The exterior parasites of conventional rabbits had been found universally.The test results on microbes and parasites of clean rats and mice were basically qualified,but there were positive samples of interior parasites.In addition,the amount of the SPF laboratory animals was very small that the needs of scientific research work were not fully met.Suggestions To improve the quality of the laboratory animals producers,our suggestions are following:Improving the managing level of the conventional animals and enhancing the preventive immunity to keep them away from the infectious and parasitic diseases.Enhancing the purification of the clean animals and improving the efficiency of the material disinfection to ensure the quality of the clean animals qualified and stable.Further improving the purification of laboratory animals and expanding the productive scale of the SPF laboratory animals to fully meet the needs of scientific research work.

    Objective From analysis of three year history of animal experiments in the Peking Union Medical College Hospital,an overview of experimental protocols,laws,and associated background information was found.Methods The animal research staff's educational background,associated departments and areas of research,as well as species and quantity of animals used were analyzed in hospital animal experiments.Result In Peking Union Medical College Hospital,animal experiment showed a steady increase in number every year,and many departments were involved in animal experiments.Conclusion In Peking Union Medical College Hospital,the research projects involving animal experiments are rapidly increasing,laboratory animal research in Teaching Hospital is playing a more and more important role.

    Objective Many SPF animal rooms were upgraded and reconstructed on the basis of the old ordinary rooms in our country,there are some deficiencies in the design of automatic monitoring system.In generally,the temperature,cleanliness,and the room pressure gradience are easily controlled.However,the humidity control is very hard,especially in the winter and rainy season.In addition,the monitoring system usually emphasizes on the monitoring function,but the control function is weak,and there is no electronic record reserved in the system.Based on these deficiencies,our laboratory animal room has been upgraded and the reconstructed,and performance of the new system is good.

    The animal laboratory of environmental barrier system in Zhengzhou University was built in 1998,and possessed automatic control of parameters,such as temperature,humidity,pressure.But there was not dehumidification function,and the facility was not in accordance with the current certification requirements of GLP.For this reasons,a variety of research and analysis on dehumidification mode were carried out in our center and finnally a system,that uses of small wheel dehumidifier for dehumidifying air supply without changing the original air-conditioning units was chosen.The performance of the system is good,not only in the technical indicators of the humidity control requirements,but also in reduction of investment and operating expenses.
