Effect of Salvianolic Acid B on Cells Factor Expressing of Mesenchymal Stem Cells

丹酚酸B对骨髓间充质干细胞相关因子表达的影响

Abstract: Objective To isolate,culture and purify rat bone marrow mesenchymal stem cells(MSCs) and observe the effect of salvianolic acid B on vascular endothelial growth factor(VEGF),stem cells factor(SCF),the cardiac muscle specificity transcription factor NKX2.5 and GATA-4 expressing of MSCs in vitro.Methods MSCs were cultured in five groups of lower glucose DMEM media containing different concentration of Salvianolic acid B as follows:group of control(add nothing),four goups of Salvianolic acid B(the lower glucose DMEM media add 0.03、0.3、3、30 μg/mL Salvianolic acid B in turns).The expression of VEGF、SCF、NKX2.5 and GATA-4mRNA in the MSCs harvested at following 24 hour were analyzed with reverse transcription-polymerase chain reaction.Results The expression of VEGF、NKX2.5 and GATA-4mRNA from MSCs treated with Salvianolic acid B were increased significantly(Compared with the control group,P<0.01,P<0.05).Conclusion Salvianolic acid B could promote the secretion of VEGF and reinforce the expression of NKX2.5 and GATA-4 mRNA of MSCs.It is useful to establish the substantial foundation to the study on induction to different of MSCs.

摘要: 目的体外分离、培养和纯化大鼠骨髓间充质干细胞(MSCs),并研究中药单体丹酚酸B对MSCs血管内皮生长因子(VEGF)、干细胞因子(SCF)、心肌特异性转录因子NKX2.5和GATA-4表达的影响。方法分别用含0.03、0.3、3、30μg/mL丹酚酸B的低糖DMEM培养液培养MSCs,RT-PCR法检测培养24h后,观察VEGF、SCF、NKX2.5和GATA-4mRNA的表达。结果丹酚酸B可明显促进VEGF、NKX2.5和GATA-4mRNA的表达(与空白对照组比较,P<0.01,P<0.05)。结论丹酚酸B可促进MSCs的VEGF的分泌及NKX2.5、GATA-4的表达,为进一步深入探讨其诱导分化打下了坚实的基础。
