    Impact of Skin Allograft Rejection on Thymocyte Subsets in Mice

    Abstract:Objective To investigate the impacts of allograft rejection on the development of thymocyte subsets in a mouse allogeneic skin transplantation model. Method Skin tissues from BALB / c mice were transplanted into C57BL /6 mice. The total cell number of thymocytes and their subsets in allo-skin-grafted recipients were analyzed by flow cytometry. Result In allo-skin transplantation group,the thymus weight was decreased,the quantities of thymocytes and their subsets including DN,DP,CD4SP and CD8SP cells were significantly decreased compared with the control mice. Conclusion The data showed that the acute rejection significantly impacts the development of T cells in the thymus.

    Abstract:Objective To study when and how much to dose KM mice while creating the cleft palate model induced by RA ( retinoic acid) . Method Divide randomly sixty KM mice which have been identified in pregnancy into four groups (A,B,C,and D),there were fifteen ones in each group. The mice of group A,B,C were tube-fed RA( retinoic acid) with 20 mg / kg、50 mg / kg and 80 mg / kg respectively at the time of GD10( gestation day 10), while the ones of group D tube-fed with 0. 20 mL corn oil at the same time. All the mice were allowed to reproduce naturally and then record the mortality of pregnant mice,stillbirth rate and incidence of cleft palate in the live fetuses. Data were compared between groups with χ2 test using SPSS16. 0 software package. Result The incidence of live fetuses with cleft palate in group B was 93. 59%,in group C which was 20. 00%,there was significant difference in the two groups (P < 0. 007),but the stillbirth rate and mortality of pregnant mice in group C were 40% and 76. 19%,which were higher than those in group B,the differences were statistically significant ( P < 0. 007) . In addition,the pregnant mice in remaining group A and D were without the occurrence of death and the fetal mice of these groups neither appear stillbirth nor cleft palate deformity. Conclusion Tube-feeding the KM mice with RA 50 mg / kg at GD 10 is an appropriate  method of setting up cleft palate model

    Abstract:Objective To explore the effect of electro-acupuncture on learning and memory behavior of five months APP / PS1 transgenic AD mice. Method 16 APP /PS1 transgenic positive male mice were randomly divided into the model group and the electro-acupuncture group,8 in each group. At the same time,there were 8 APP /PS1 transgenic negative male mice as the normal control group. The“baihui”and“yongquan”of electro-acupuncture group were acupunctured using dilatational wave for 15 min ,every other day at a time. By the same method ,the mice of control and model group were bounded by binder for 15 min. The all three groups were treatmented for five weeks. Finally the ability of learning and memory of all mice was tested by Morris water maze. Result Escape latency between three groups was statistically significant( P < 0. 05 ),escape latency between the model group and the electro-acupuncture group was statistically significant ( P < 0. 05 ) . Conclusion Electro-acupuncture can improve learning and memory ability of APP /PS1 transgenic AD mice.

    Abstract:Objective To compare and study three inspecting methods which test whether the mice were pregnant or not. Method Check and study the fertilization of the same mice group with three methods. The first method was to use vaginal suppository to identify the mice were pregnant or not; the second method was to use vaginal suppository and check mice’s weight; the third method was to check mice’s weight. Result The first method sees the sensitivity of the mice’s pregnancy was 0. 71 and the specificity was 0. 47. According to the second method,the sensitivity was 0. 71 and the specificity was 0. 96. And third method shows that the sensitivity was 0. 96 and the specificity was 0. 94. Conclusion For the methods detecting mouse gestation GD 10 and late state,observing change of weight is more reliable and practical than other methods in the process of inspecting pregnant mouse in the GD 10 period and late stage.

    Abstract:Objective Comparing with the common hyperlipidemia rat model,which was induced by exogenous high-fat feed,explored the blood lipid level and dynamic change trend of acute high blood triglycerides rat model, which was lavage by fructose. Method 48 SD rats were randomly divided into normal diet control group (C1,C2, C3),high fat diet group (M1),high fat chyle agent group (M2),fructose group (M3A,M3B,M4) . Blood was sampled from rat eyeball angular vein when M1 group was raised by 4 week,M2 group was lavage for 2 w,M3 group was at 0. 5 h after the last lavage,which M3A and M3B groups were established by different fructose doses and times of lavage. M4 group was before lavage,at the time of 8 h ,16 h,24 h,36 h after the last lavage respectively,Serum total cholesterol ( TC),triglyceride ( TG),high-density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol ( LDL-C) were tested. Result In high-fat diet and high fat chyle groups,TC increased significantly ( P < 0. 05 ) but TG was no significant difference. In fructose group ,TG increased significantly( P < 0. 05 ) whereas TC,HDL-C was no obviously difference,furthermore,the blood lipids level between M3A and M3B groups were no difference;In M4 group,TC,HLD -C level decreased as time went on,TG peaked at 8 h,decreased to base value at 36 h. Conclusion TG rapidly rised stability by fructose lavage , meanwhile,in fructose groups,TG increased obviously,it could achieve the same effect by different doses and times, TG lasts for 36 h.

    Abstract:Objective To investigate the effects of rutin on ROCK I protein in STZ-induced DCM. Method DCM model was induced by STZ. 24 male Waster rats were randomly divided into control group ( n = 6),STZ group ( n = 6),rutin + STZ group ( n = 6),Positive drug + STZ group ( n = 6 ) . The rats of the treatment groups were given rutin,DCM group and normal group were given 0. 9% sodium chloride. The index of rats after intervention of rutin morphology,collagen content and ROCK I protein were observed. Result After injection of STZ,collagen and ROCK I protein expression increased compared with normal control group( P < 0. 05) . After rutin treatment, its value reduced as compared with the model group. And HE staining showed that the degree of cardiac injury reduced after rutin treatmenr. Conclusion Rutin may regulate the expression of ROCK I and reduce myocardial injury. Specific mechanism needs further study.

    Abstract:Objective To investigate the effects of Hulan hypoglycemic sustained-release tablets( TGE) on glucose and lipid metabolism and islet function in diabetic animal model. Method The animal models of diabetes mellitus was established by intraperitoneal injection of streptoztocin ( STZ) 40 mg / kg. The rats were divided randomly into four groups:normal control group,diabetic group,western medicine group( repaglinide) and Hulan hypoglycemic sustained-release tablets group. After different administration for 4 weeks,the rats were killed and blood sample was collected for fasting blood glucose ( FBG ) ,Triglyceride ( TG ),total cholesterol ( CHO ),high-density lipoprotein cholesterol ( HDL-C) and low density lipoprotein cholesterol ( LDL-C) and fasting insulins ( FINS ) . Result Compared with other,the pancreas was fixed for intervention groups,TGE group had lower blood glucose(P < 0. 05) . Compared with normal control group,other group shown lower FINS(P < 0. 05) . Compared with diabetic group,western medicine group no difference for FINS(P > 0. 05),but the difference of FINS was observed between TGE group and diabetic control group ( P < 0. 05 ) . Conclusion Hulan hypoglycemic sustained-release tablets have a modulatory role in glucose and lipid metabolism and function of pancreatic islet in diabetic animal model.
    Correlation Analysis between AChE Activity Variation of Whole Blood and That of Brain Hippocampus in Nerve Damage Rats

    Abstract:Objective To study the correlation between AChE activity of whole blood and that of brain hippocampus in nerve damage rats. Method 120 male SD rats ( 80 - 100 g) were equally divided into 4 groups,including control group ( peanut oil),low dosage group (40 mg CP / kg body),middle dosage group (80 mg CP / kg body) and high dosage group ( 160 mg CP / kg body ) . Intragastric administration at 1 time a day were planed,the treatment lasted 28 days. General symptoms of rat were observed everyday. 10 rats of each group were selected at 14,28 day after treatment and 7 days after last treatment,respectively. Then blood was collected by tail abscise, activity of whole blood AChE was measured. Brain hippocampus of selected rats were separated,then AChE activity were measured by immune-histochemistry. Result Rats showed symptoms of nerve damage,AChE activity of whole blood and brain hippocampus were all decreased considerably(P ﹤ 0. 05),AChE activity of middle and high groups between whole blood and hippocampus was positive correlation(P ﹤0. 01) . Conclusion Rats showed symptoms of nerve damage after chronic exposure to CP,AChE activity between blood and hippocampus was positive correlation. AChE activity of whole blood has potential of becoming marker of nerve damage.

    Abstract:In order to measure survival,body weight and food intake of Wistar rats feeding for two years,tumor occurrence and survival of Wistar rats were observed for two years,and food intake and body weight were investigated weekly. The results showed mortality and survival time of male rats were higher than female rats, spontaneous tumor incidence of female rats were slightly higher than male rats,and multi-organ tumors were found in one animal. Compared with female rats,food intake and water intake of male rats had no apparent change trends. Body weight grew obviously in earlier and obtained peak later,and then declined slightly. Daily food consumption and body weight percentage declined earlier in the test,and slightly rebounded after the minimum value was reached.     Abstract:To identify the pathogen from diarrheic tree shrews which may provide an useful reference for preventing and treatment diseases in tree shrews ( Tupaia belangeri Chinensis) . 6 fresh feces were collected,and identified the dominant and suspicious bacteria by cultural characteristics,colony morphology,staining,biochemical testing and molecular biology. Antibiotic sensitivity of isolated strains were detected with 8 kinds of antibiotics. Most colonies were white-yellow with smooth and damp surface on nutrient agar plates,round with dark cores. Some of them spread black ripples and smelly odour. Bacterial character were brevibacterium with blunt,without spore and capsule,size were (0. 4 ~ 0. 6 μm) × (1. 0 ~ 3. 0 μm),gram negative and non acid fast bacteria. The isolated strain produced hydrogen sulfide. The reactions of ornithine ,urea etc were positive. Genetic similarity were 99% with the Proteus mirabilis from GenBank. All results showed the isolated was a strain of Proteus mirabilis. The isolated strains were sensitive to all tested antibiotics. The Proteus mirabilis isolated from diarrheic tree shrews is the pathogenic agent leading to diarrhea and death in lactation tree shrews and infant. It was be controlled by improving management and treatment.

    Abstract:Objective Investigation and analysis for the classification,characteristics and possible pathogenic to animals of the parasite infection in tupaia belangeri chinensis,to provided the scientific basis for protecting tree shrew population and the quality control of breeding tree shrew. Method Two hundred wild tree shrews were random selection,then we took living floating and depilating to detect the epizoic parasites,at the same time took faeces precipitation and faeces floating method to detect endoparasite,at last,diagnosed species of parasites under the platoscope and microscope. Result The epizoic parasites in wild tree shrew were respectively louse(20. 5% ), flea(5. 5% ),sarcoptid mite (3. 0% ),Ixodes granlalus(1. 0% ),Niptus bololeucus Faldermann (1% ),Thorictodes brevipennis(1% ),W. acranulat(1% ),Anobiidae Fleming(0. 5% ) ;The endoparasite were respectively wireworm (14% ),Wireworm egg ( 33% ),tapeworm ( 9% ),tapeworm egg ( 20% ),amoeba ( 2% ) . Conclusion Parasite species with infection tupaia belangeri chinensis were a great variety,should be recognised as serious,we could not be underestimated its impact on tree shrew population.
    The Improvement of Method of Intraperitoneal Injection in Rat Anesthetization

    Abstract:Objective To introduce a new method of intraperitoneal injection in rat anesthetization to decrease the anesthesia mortality. Method One hundred SD rats were divided into 2 groups: control group and experimental group. The animals were anesthetized by intermittent intraperitoneal injection in experimental group,while the control group anesthetized by single injection. Differences in the anesthetic onset timespan,duration and mortality were assessed between two groups. Result In control group,the rats with the onset time of 3. 8 ± 1. 4 minutes, duration 115. 5 ± 12. 8 minutes and anesthesia mortality was 12% ; in experimental group,onset time of 10. 9 ± 2. 5 minutes,duration 129. 3 ± 9. 8 minutes and zero anesthesia mortality. Conclusion Intermittent intraperitoneal injection in rat anesthetization could decrease the anesthesia mortality and improve the animal surgery success ratio.

    Abstract:Oxyrrhis marina is one of marine pelagic protozoa and widely distributed in the global coastal waters,and now became model marine planktons for laboratory marine biology experiments. The feeding characteristics of O. marina on C. marina in co-culture were analysed by continuous microscopic examination. The results showed that the feeding method of O. marina on C. marina is disturbance feeding,and this feeding method was reported for the first time in the world. The results provided experimental evidences for discussing succession mechanisms of red tides and coastal biological communities.

    Abstract:An old rhesus monkey developed diarrhea,marasmus as the main syndrome of acute disease. To study the pathological changes of digestive system in old rhesus monkey,stomach and duodenum was investigated by general observation and routine HE staining. It was found that varying degrees of pathological changes were appeared in digestive system of rhesus monkeys including a variety of inflammatory cell infiltration,goblet cells and mucus secretion reducing,degeneration or necrosis of the digestive gut mucosa,digestive gland atrophy. It was showed that this older monkeys was physiological senility.
