The Influence of Exogenous RNA Upon Phenotype and Protein Expression of Mouse Fibroblasts

Mouse fibroblasts were treated with different RNA and the influence of RNA upon their phenotypes were observed. It was showed that different RNA can all change phenotypes of fibroblasts. The changed phenotypes could be kept for 4 generation. Mouse fibroblasts treated by rat liver RNA were generated for 4 generations and SDS polyacrylamide gel electrophoresis were performed to analyse the influence of RNA upon protein expression. It was showed that there were changes in proteins between treated fibroblasts and their control.

A RT-PCR method for detecting canine distemper virus (CDV) was established with one pair of primer(P1、P2) designed and synthesized according to the sequence of the fusion(F) gene of CDV and reverse transcription template of the standard of CDV-ONP strain and was used to identify the isolate. The result showed that the method was specific and sensitive. 324bp gene sequence which is the same length with designation is amplified with one pair of primer from the reverse transcription template of CDV-ONP strain. The study has not only established a specific and sensitive RT-PCR method to detect CDV but also provided a basis for cloning , sequence analysis and expressing to the main domain of CDV F gene.

Hematological and blood biochemical index of Smad3 mice of the 70d old were determinated. The results showed: there were a few significant differences in mice between sexes(male and female mice). The parameters of the WBC and hemoblast of homozygous mutants were higher than those of heterozygous mutants wild-types. The parameters of CER、CK、UA、GOT、TP、ALP、LDH-L、MG、CA were significant among three types through statistics. The parameters of CK、UA、TG、ALP、LDH-L of homozygous mutants were higher than those of wild-types, but the parameters of GLU、P、MG、CA were in reverse.

Bovine frozen semen were thawed and capacitated in vitro by heparin and caffeine for 1h. The ultrastructrual changes of sperm after capacitation were observed under transmission electron microscope. The results were as follows (1) Both of the bovine sperm acrosome inner membrane and outer membrane were double layered; (2) There were various patterns of acrosome viscularization after sperm capacitation, such as fusion of plasma membrane and acrosome outer membrane, self fusion of the double layered acrosome outer membrane, invagination of acrosome outer membrane or extrudation and fusion of double layered acrosome inner membrane; furthermore several vesicularization patterns could be seen in the same sperm acrosome reaction; (3) After capacitation, the post part of the sperm head nucleous membrane without acrosome covered expanded and fused with expanded plasma membrane; (4) After capactiation, the tail membrane expanded, and the mainpiece of the plasma membrane was much more expanded than that of the midpiece part, which may be related with hyperactivation.

To study the effect of low or high protein diet on bone metabolism in male rats by bone histomorphometry. Eighteen 3-month-old Sprague-Dawley male rats were randomly divided into three groups:control group(pr20%)、low protein(pr8%) and high protein group(pr23%).Feeding purified diet for 90 days,all rats were killed.The poximal tibia(PTM) was harvested for histomorphometic analysis. LPD group had lost 57%( P <0.01)of bone cancellous compared with control group.Dynamic date in LPD group showed bone formation indices such as percent labeled perimeter(L.Pm%),mineral apposition rate(MAR) and bone formation rate(BFR/BV) increased without significance;HPD group had also lost 40%( P <0.05) of cancellous bone compared with control group, representing decreased bone turnover.Comparing with HPD group, indicates of LPD group all increased .The results indicated bone cancellous loss evidently、bone frame was weak、bone intension decreased in low protein dietary rats,and high protein dietory rats is also.But LPD restrain bone formation more strongly than HPD.

The analgesic effect of Sengui mixture was confirmed by the methods of light-hot,electric stimulation and chemical stimulation.This experiment showed that after taking Sengui mixture (5\,10\,20ml/kg)orally,the liminal value of rabbit ache caused by calorediance was increase evidently;the response of rabbit's masticatory movement caused by electric stimulating tooth was inhibited;the tic douloureux caused by peniciline G K was calmed down;the uterine spasm caused by oxytocin and prostaglandin E1 was calmed also down.

According to our national standard \[Experimental Animal Environment & Facilities\] GB/T 14925-94,a comprehensive monitoring and overall evaluation were conducted on the parts of the experimental center that were rebuilt with color plastic coated steel plate and epoxy resin.The results showed that,after the rebuilding,the facility quality for experimental animals has met all the requirements and demands set by our nation and achieved the objectives for the facility rebuilding:less investment for rebuilding,less cost for maintenance,with a rational layout and flow-direction science.The experience we acquired in the rebuilding is worth being recommended to other Research Institutes,Drug Plants and Experimental Animal Centers.

In order to evaluate safety of bacterial strains used in organic rubbish fermentation, acute toxicity test and bone marrow micronucleus test were done in mouse, and stimulation test was done on rabbit skin. The results indicated that no acute toxicity by ig, no mutagenesis and no stimulation on skin caused by these bacterial strains were founded in the animal experiments mentioned above.

Objective The purpose is to investigate the model of tracheal tube and the placed-depth of tracheal intubtion by rabbit's mouth. Methods\ Fifty rabbits were divided into four groups by weight, Group A (1.8-2.2kg), B(2.2-2.4kg), C(2.4-3.0kg), D(3.0-3.5kg).Tracheal tube was placed blindly into trache. Machine controlled ventilation was managed. Monitoring included airway pressure, uplifted-height of thorax, breathing of both lungs, et al. Postoperation, dissect rabbit's respiratory tract and measure its length and tracheal inside diameter. Result\ The tracheal tube model of each group is “3.0”,“3.0-3.5”,“3.5”,“4.0”. The placed-depth of each group is 11-12cm, 12cm, 13cm, 13-14cm. Conclusion\ To rabbits whose weight is ranged from 1.8kg to 3.5kg, The tracheal tube model is probably “[weight(kg)-2.2]+3” and the placed-depth is 12.5±1.5cm.

It was preferable method that feed for specific pathogen free (SPF) chickens was sterilized by common electric steam sterilizer in place of fluctuating vacuum steam sterilizer, whose invest price and utilizing cost were rather high. And this process required that the feed should be wetted with water prior to sterilization. The result demonstated that the modified measure could lower operating costs for SPF chicken in bio-clean house as well as could keep or increase the microbes eliminating quality.
