    The Comparison in Basically Biological Characteristics of Wuhan H_(22) Cell Lines and Its Subline H2D8

    With the single cell cloning technique, we cloned H2D8 subline from H 22 cell line stored by China Center for Type Culture Collection in Wuhan (Wuhan H 22 ). Then the comparative analysis of Wuhan H 22 and H2D8 about their dependence on serum when cultured in vitro , growth feature in vivo , pathomorphism and therapeutic feature to CTX were carried on. With the results, we found that there was no significant difference between H2D8 and Wuhan H 22 about the dependence on serum, growth feature in vivo and pathomorphism. When the mice bearing tumor were treated with CTX,the restrain neoplastic rates of H2D8 was higher than that of Wuhan H 22 . It means that H2D8 clonal cell line retained the characteristics of Wuhan H 22 cell lines about the dependence on serum when cultured in vitro , grouwth feature in vivo and pathomorphism. H2D8 has higher sensitivity on the chemotherapy feature to CTX than that of Wuhan H 22 .

    To establish the replantable models of tumon cell line S 180 and EAC using BALB/c mice respectively. The mices were randomly divided into 2 groups, with one group given therapeutic drug and the other as a control. After 10 days, the tumour weights and pathological changes were compared. There is very apparent difference of pathological changes between two groups. The tumour cells were found in the mices liver of the control groups. This proves that pathological evaluation shown a significant reference value.

    To analyze the causes for complications during surgical construction of rat penetrating keratoplasty model, to discuss how to prevent and manage these complications and to summarize experience and skills to increase rejection rate for this allografts model. In order to construct an orthotopic rat penetrating keratoplasty model, 34 allografts (SD→Wistar) were performed in the study group and 34 autografts (Wistar→Wistar)were used as controls. To increase rejection rate of allografts, the donor corneas were trephined eccentrically and the sutures were not removed on the recipients. No rejection was found on the autografts, while 100% rejection was identified in the allografts, with an average survival time at 10.8±2 1 days. Complications were encoutered in 10 eyes, which included iris prolapses, graft cleavage, hemorrhage in the anterior chamber, cataract and infection. Conclusion:MHC mismatched species, eccentrically removed donor corneas and kept sutures in the recipients contribute to the high rate of rejection. Gentle and skilled surgical manipulations, sharp instruments and pupil enlargement are of great importance to successful operations.

Results The ICR mouse is much better than KM and NIH mouse in producing diabetic animal model and s of blood sugar measured, and is the most suitable animal in this experiment.

    objective In order to find a high effective tool to make high specificity and high affinity monoclonal antibody, improve scientifically and veracity of diagnosis. Method use isolator technique, feed germfree mice with antigen free diet (chemical diet, molecule weigh is not over 8 000 Dalton), when F1 wearing, transfer to another isolator, and continue to feed mice with chemical diet, in this way, the generation 4 is call antigenfree or antibody free mice. Results the concentration of sera IgG in antigenfree mice is very low that is 0 3mg/ml, it is only 5%-10% that is in conventional mice. Conclusion It indicated that this technique was grown up and the antigenfree mice clone was established successfully. The antigenfree mice will play an important role on making monoclonal antibodies.
    Protective Effect of She-xiang-kai-qiao Granule on Learning and Memory Impairment in Mice

    objective To investigate the effect of she xiang kai qiao granule on mental disorder. Method ICR mice weighted 20-24g were divided into:the low ,the middle ,the high dosage(1 00,2 00,4 00g crude/kg),the model, the positive drug control and the negative control group. The drugs were administered (i.g) daily for 15 days, the effect of she xiang kai qiao granule was examined by the Y water maze test, the step down test and swimming in cold for mice. Results 1.In step down test, she xiang kai qiao granule was decreased the errors of respouse and prolonged the latent period, which showed a significant difference ( P<0 05,P<0 01,P <0 05)in every dosage group compared with model group. 2.She xiang kai qiao granule have a protective effect for the mice disorientation. From the 4th day, the number of errancy showed a significant difference( P <0 05) in high and middle dosoge group compared with model group. 3.She xiang kai qiao granule can improved on strength and endurance in some degree ( P <0 05).Conclusion She xiang kai qiao granule can improved mental disorder of mice significantly.

    objective To evaluate the safety of external applied Chinese traditional medicine“Ai zheng zhen tong san”. Method To make use of skin irritation test of rabbit, skin allergy test of guinea pig, skin acute toxicity test of rabbit, skin long term toxicity test of rabbit to carry out the evaluation. Results When“Ai zheng zhen tong san” was external applied under equivalent dose and concentration of clinical use, it doesn't induce allergic reaction in guinea pigs, but has a slightly irritation to rabbit skin. While external applied under high doses, it appeared slightly irritating to rabbit skin in cute toxicity test, and induced skin incrassation and connective tissue hyperplasia in corium layer in long term toxicity test, no systemic toxicity were observed. Conclusion “Ai zheng zhen tong san” is considered safety when it be used as suggesting clinic dose and concentration.

    It is our aim to establish model combining disease with syndrome of chronic atrophic gastritis(CAG) in rats more corresponding to symptoms of disease,syndrome and combining disease with syndrome in clinic.Design of methods how to establish the model:①To establish the model for a long time:We believe that “chronic” is not only simply a character of the disease,but a concept of dimension.It send the pathology of disease to three dimensions.Therefore we selected a fairly long time to establish all models of disease,syndrome and combining disease with syndrome in rats.Results have revealed that there were not hemorrhage,ulcer and dissipated in all gastric mucosa membrane of models,and exudation,necrosis is not the major pathology change in tissues of models.②Establishing the model of CAG:We considered that the precancerous lesion,even the carcinomatous change is only the result and termination of deficiency syndrome.Carcinogen is the especial matter of to cause deficiency syndrome.We did not use carcinogen to cause CAG,but general modeling factors as far as possible.We also pay attention to the change of entire body related to CAG as to estimated the model of combining disease with syndrome of CAG.③Establishing the models of syndrome:Methods of establishing the deficiency syndrome of spleen,stagnation syndrome of the liver,deficiency syndrome of the kidney are developed.According to the request of this experiment and the results of pretestings,we prolong the time to make model.④Establish the model of combining disease with syndrome:Deficiency syndrome of kidney was not taken as the major syndrome of CAG in clinic.According to reverse recapitulation law,we believe that the essence of CAG,especial precancerous lesion of gastric mucosa membrane,is deficiency syndrome of kidney.As far as possible we also pay more attention to integrating the modeling factors of disease with the modeling factors of syndrome,include integrating the modeling factors of CAG with of deficiency syndrome,and integrating the modeling factors of deficiency syndrome with of CAG.
