一种高效制备高特异性高亲和力单克隆抗体的工具——无抗原小鼠的培育

Abstract: objective In order to find a high effective tool to make high specificity and high affinity monoclonal antibody, improve scientifically and veracity of diagnosis. Method use isolator technique, feed germfree mice with antigen free diet (chemical diet, molecule weigh is not over 8 000 Dalton), when F1 wearing, transfer to another isolator, and continue to feed mice with chemical diet, in this way, the generation 4 is call antigenfree or antibody free mice. Results the concentration of sera IgG in antigenfree mice is very low that is 0 3mg/ml, it is only 5%-10% that is in conventional mice. Conclusion It indicated that this technique was grown up and the antigenfree mice clone was established successfully. The antigenfree mice will play an important role on making monoclonal antibodies.

摘要: 目的 为了寻找一种新的高效制备高特异性高亲和力单克隆抗体的工具 ,提高工作效率 ,增强诊断试剂科学性和准确性。方法 利用无菌隔离器技术 ,无抗原饲料 (由氨基酸、碳水化合物、维生素、必需矿物质、植物油等组成的分子量小于 80 0 0的化合物 )喂养无菌小鼠 ,将无菌小鼠培育成无抗原小鼠。结果 经ELISA方法检测该小鼠血清中IgG的含量 ,其含量是CV和GF小鼠的 5 % - 10 %。结论 无抗原技术已完全成熟 ,无抗原小鼠培育成功。由于该无抗原小鼠未接受过外源性抗原的刺激 ,可利用该动物制备高特异性、高亲和力的单克隆抗体
