Detection of Staphylococcus aureus in Laboratory Animals by Dot Blotting Using Biotin-labeled nuc Gene Fragment as the Probe

To establish a sensitive dot blotting for detecting Staphylococcus aureus(S.aureus), a DNA fragment of 678 bps of the nuc gene was amplified from S.aureus genome DNA by Polymerase Chain Reaction(PCR) and labeled with biotin as the probe.The purified genomic DNAs of S.aureus,Streptococcus,Escherichia coli and Pasteurella were blotted onto hybridization membrane and hybridized under high stringency conditions.The Biotin\|labeled probe reacted positively with the S.aureus genomic DNAs,but not with that of the non\| S.aureus with a sensitivity of 1 pg.240 clinical samples taken randomly from laboratory mice and rats were submitted to detection by the dot blotting and previously established PCR with a positive detection rate of 2.9%(7/240) and an agreement of 100%.These data demonstrated that the established dot blotting was a quick,sensitive and specific method for detection of S.aureus in laboratory animals.

Objective\ To evaluate the therapeutic effects of Bleomycin A 6(BLMA 6) and cis\|platium Diamino\|Dichl\|Pride(DDP) on human esophageal adenosquamous carcioma and squamous carcinoma.Methed\ Two xenofrafts were treated with the same drug and every tumor xenograft was treated with both agents.Result\ Bleomycin A\-6 can reduce the growth of HEC\-2 and HEC\-6 and there is no apparent side effects in experimental animals.DDP only reduce the growth of HEC\-6.Conclusion\ Blemycin A\-6 is more effective than DDP in the treatment of human esophageal adenosquamous carcinoma and aquamous carcinoma.

Objective\ To study whether excess sexual activity have an effect on the male mouse and compare the effect of reproductive capacity of male mouse with different methods.Methods\ To compare LDH\|X of spermary,alive embryos of female mouse,activity and density of semen and amount of mutated sperms.It is used by excess sexual activity to establish the male mouse model with kidney yang deficiency,and the model compared with spleem dificiency and kidney yang deficiency by trepidation and hydrocortisone.Result\ The effect of reproductive capacity of male mouse established by excess sexual activity is evident.Conclusion Excess sexual activity can have an effect on reproductive of the male mouse model with kidney yang deficiency.

Choosing Smad3 gene konck\|out mice of the 6 month,hematological and blood biochemical parameters、avoirdupois、viscera weight of the Smad3 mice of 6 months were determinated.The results showed:There were a few significant differences in the mutated mice of MCH,CK,Tibil,GOT,LDH\|L and supraenal gland index,kidney avoirdupois of heterozygous.The parameters of WBC,HCT,MCHC,GLU,CHO,ALP,BUN,LDH\|L,CA and avoirdupois、heart、liver、kidney、thymus index were significant among three types through statistics.The parameters of WBC,HCT,CK,CRE,GPT,GOT,ALP,BUN,LDH\|L of homozygous mutants were higher than those of Wild\|types,but the parameters of MCHC,GLU,CHO,MG and CA were in reverse.

objective\ To compare and observe the granulocyte series in peripheral blood in SIVmac\|infected Rhesus monkey and Cynomolgus monkey .Method\ Examine the quantity and morphology of the WBC by routine method.Results\ The peripheral blood granulocyte in SIVmac\|infected Rhesus monkey and Cynomolgus monkey ,similar to those seen in human AIDS,showed a regular change on the quantity and morphology of the WBC.3 Rhesus monkeys and 4 Cynomolgus monkeys are incluted in this research.The WBC count started to decrease and abnormal lymphocytes increase two weeks after infected with SIV virus on Rhesus monkey and Cynomolgus monkey ,and then the value of WBC elevated continuously to a peak,abnormal lymphocytes remained increase;8 weeks later,the WBC and abnormal lymphocytes started to decrease,three Cynomolgus monkeys died from viremia in 8 weeks.The WBC decreased continuously in a remaining Cynomolgus monkey in 12 weeks.Conclusion\ This essay showed phenomena of WBC changes is similar to diagnostic criterion of human HIV/AIDS.Besides, Cynomolgus monkey is more sensitivity than Rhesus monkey in SIVmac infection.

Senescence\|accelerated mouse(SAM) strains, the murine models of accelerated senescence, are unique and appropriate models for gerontological studies because the SAMP strains have \!accelerated senescence\" phenotypes and each SAMP strain has a strain\|specific and aging\|associated disorder, while the SAMR strains are controls. In the first part of this paper, the history of the SAM development has been reviewed, which includes the circumstance of SAM development, the genetic background of SAM development and the pathobiological phenotypes in SAM strains. In the second part of this paper, the current status of the SAM used as murine models in gerontological studies was concisely summed up, including researches in nutrition metabolism and physiology, circumstance and immunology, behavior and cerebrology, senile amyloidosis, senile osteoporosis, senile lung, aging associated other diseases and the genetics research of those senile diseases. At last, the significance of SAM development was forecast.

Objective The S D rats are introduced into on our animals centre from Japan in 1994. They had been breed in barrier system. But the staphylococcus aureus are carryed on the S D rats. For this reason, the S D rats were not accord with the standard of SPF rats in China. We sucessfully cleansed the S D rats by cesarotomy in 1997, the SPF Wistar rat mothers took the place of S D rats mothers as murses. We successfully have cleansed 35 pregnant S D rats, 115 young animals have be obtained by cesarotomy. The staphyloccocus aureus are not carryed on the cleansed animals from 1997 year to 2001 year. The cleansed animals are accorded with the standard of SPF rats in China.

objective To observe whether the ketamine has any effect on the development of plasmodium in anopheles after the plasmodium infected monkey was given intra muscular injection of ketamine and then exposed to mosquitoes sucking to suck blood. Method The plasmodium infected monkey was exposed to mosquitoes to suck blood and after before being given injection of ketamin. Oocysts and sporozoits of plasmodium in mosquitoes were examined on day 9 and day 12 after mosquitoes sucking blood, respectively. Results The positive rates of oocysts and sporozoite infection of control groups were 100%, the same as those of the ketamine treated groups there was no significant difference ( p >0 1) between the control groups and the ketamine treated groups in the density of oocyst and the average size of oocyst and the average density of sporozoite infection.Conclusion After the plasmodium infected monkey was injected with ketamine and then exposed to mosquitoes to suck blood, there was no influence on the development of plasmodium in anopheles and the mosquitoe sucking blood could be improved and the experiment time could be shortened.

In order to determination of hazardous solid wastes,the acute toxicity on solid wastes were detected by using ten mice.The result indicated that the sample has no acute toxicity found.The results of screening test for acute toxicity can meet the environmental monitoring demand of hazardous solid wastes.
