    Behavioral Analyses of Learning and Memory Obstructed Animal Models Used Commonly in Pharmacological Study

    Objective To observe and analyse behavioral characteristics of the rats in four kinds of animal models of learning and memory disturbances(model of aged rat,the animal model of learning and memory disturbances caused by damaged basal forebrain,caused by injection of scopolamine,and caused by ligation of bilateral carotids) used commonly in pharmacological study.Methods The learning and memory ability of the rats,spatial cognitive ability of the rats, and excitability and emotion of the rats in new circumstance were observed by water maze and open field tests in the four model groups,control group of young rats,and control group of pseudo-operation.Results The learning and memory ability of the aged rats was significantly decreased and they became more nervous in the new circumstance,with normal spatial cognitive ability and excitability.The learning and memory ability and spatial cognitive ability of the rats of damaged basal forebrain were markedly decreased,but their excitability and emotion in the new circumstance were normal.In the model group of injection of scopolamine,the learning and memory ability and spatial cognitive ability of the rats were decreased and their excitability in the new circumstance was enhanced.In the model group of ligation of bilateral carotids,the learning and memory ability and spatial cognitive ability of the rats were markedly decreased,but their excitability and emotion in the new circumstance were normal.Conclusion These results could come up with background information that would be beneficial to better uses of the four kinds of animal model of learning and memory disturbances in the pharmacological studies.
    Developing Carotid Atheroselerosis by High Fatty Forage and Balloon Injury in Rabbits

    Objective To delve into the method of developing animal model of carotid atherosclerosis.Methods 29 rabbits were randomly divided into three groups.12 rabbits in the first group were fed with high fatty forage and their carotids were injured by balloon,9 rabbits in the second group were fed with high fatty forage,and 8 rabbits in the third group were fed with normal forage.After eight weeks,all the rabbits were sacrificed,the morphological characters of pathological changes of the carotids were observed and thickening degree of the carotid endomembrane was measured.Results The typical pathological changes of atherosclerosis,such as thickening of the carotid endomembrane,transmigration and proliferation of smooth muscle cells,lipidosis,and formations of elastic fibers and collagen stroma,were found in 11 rabbits of the first group and 1 rabbit was dead in the first group,and no carotid occlusion was found in the first group.The pathological changes of mild atherosclerosis were seen in the second group.Conclusion The treatment of high fatty forage and balloon injury was able to develop animal model of carotid atherosclerosis in rabbit.
    The Effect of Different Lightingregimen on Peripheral Blood Parameters of BALB/c-nu Nude Mice

    Objective To study the effect on peripheral blood parameters of different lightingregimens in BALA/c nude mice.Method The BALB/c nude mice were divided into three groups:(16 h bright 8 h dark),normal illumination group(12 h bright 12 h dark),and short illumination group(8 h bright 16 h dark).The animals were maintained in three different lightingregiments,to produce second generation to 70 days old,then the peripheral blood parameters were measured,including 15 indexes altogether.Result In the long illumination group and the short illumination group,compared with the normal illumination time group,the hemoglobin determination(HGB) and the red blood cell pressure accumulates(HCT) were both lower.In the long illumination group,compared with the normal illumination group,MTD and the GRA quantity were higher.The short illumination group compared with the normal illumination group,GRA% were lower and LYM% were higher,and PLT were higher.Conclusion The different lightingregiment can create the certain disturbance to the mouse blood corpuscle,normal illumination creates the disturbance is smaller,and suits mouse maintenance.

    Objective To delve into the relationship of blood-lipid to serum bilirubin in ApoE gene-knockout mice.Methods Male ApoE gene-knockout mice of twelve-month-old were selected in this study,and male C57BL/6J mice of twelve-month-old were used as controls.The levels of TG,TC,LDL-C,ApoB,T-BIL,and D-BIL in serum were determined in these mice.Results The serum levels of TG TC,LDL-C,ApoB in ApoE gene-knockout mice were significantly higher than those in C57BL/6J mice(P<0.01),and the serum level of D-BIL in ApoE gene-knockout mice was significantly lower than that in C57BL/6J mice(P<0.01).Conclusion Blood-lipid of the mice with atherosclerosis was correlated with serum bilirubin.

    Objective To measure and analyze the main organ weight and coefficients of SD rats in a barrier system and individually ventilated cages.Methods Total 60 SD rats were used in this study,30 in each group.The rats were raised in a barrier system and IVC for 120 days.The main organ weight and coefficient for heart,liver,spleen,lung,kidney;adrenal,brian testis uterus,ovary were then measured using an electronic scales.Result between female SD rats,spleen weight showed significant difference(P<0.01),all organ coefficients were no significant differences(P<0.05);Between male SD rats,all organ weight were no significant difference(P<0.05),kidney coefficient showed significant differences(P<0.05);between female and male,in a barrier systems and IVC,all organ weight showed very significant differences(P<0.01),between female and male in IVC,no differences were found in the liver and kidney coefficients(P<0.05);between female and male in close system,the organ coefficient of the heart,spleen,lung,adrenal and brain showed significant the difference(P<0.05).Conclusion The organ weight and coefficient of SD rats in a barrier system and IVC were not different significantly.

    Objective To compare the effect of different anesthetics in rats.Methods To divide SD,Wistar and Lewis rats into different groups and to anaesthetize them with different doses of different anesthetics.Result The effects of different anesthetics on rats are different.Conclusion It is necessary to choose appropriate anesthetic and dose for the purpose of experiments.

    Objective To investigate infections of microbes and parasites in domestic miniature pigs from Beijing,Chongqing,Yunnan,Guiyang,Guangxi,and Shanghai by detecting the antibodies against microbes and parasites.Methods The serum samples of miniature pigs from domestic different areas were collected,and 14 indexes of microbes and parasites were determined by serological methods. Results The infectious spectrum and rate of microbes and parasites were different among the different populations of miniature pigs.Conclusion It was suggested that the miniature pigs with low infectious rates of microbes and parasites should be selected in the seed selection,and the breeding condition should be improve and well epidemic prevention program should be established in the reproduction.

    Objective To observe the effects of different grafting materials(that is,complex material of the osteoblasts from self-periosteum and calcined bone coated with collagen,complex material of osteomorphogenetic protein and fetal bone,and complex material of decalcified bone matrix gelatin and red bone marrow) on repairing of bone defects in rabbits by pathology.Methods The model of bone defects of rabbit's radii on two sides were developed,three kinds of complex material were respectively transplanted to bone defects of rabbit's radii on right side and no complex material was transplanted to bone defects of rabbit's radii on left side as controls,and the effects of different grafting materials on repairing of bone defects were observed by pathology.Results A large number of osteoblasts were seen around calcined bone coated with collagen and a few osteoid tissues were found four weeks after complex material of the osteoblasts from self-periosteum and calcined bone coated with collagen was transplanted,trabeculation of bone was found eight weeks after it was transplanted,and lamellar bone,mature osteocytes,and marrow cavity were found sixteen weeks after it was transplanted.The other two complex materials also possessed osteogenic actions.Conclusion By comparison,the osteogenic action of complex material of the osteoblasts from self-periosteum and calcined bone coated with collagen was better than other two complex materials.

    Objective To establish a reliable chronic venous access in the freely moving rat.Methods Some modification have been made based on the former experience.Result: 70% of rats can finish the experience for a month,a few rats failed for surgery death,infection,catheter block.Conclusion The method applied in this study is a reliable venous access in the freely moving art.

    Objective By comparing the pessary and the microscopic smear for predicting the accuracy of pregnancy to improve rat model manufacture method.Method 300 Wistar rats were put together in cages for 12 h in which the femela/male ratios were 2∶1.At the same time two methods were used to examine the pregnant situation of rats.Results The masculine gender rate of the pessary method was 72.5%,the false masculine gender rate was 2.06% and the false negative rate was 5.23%.The masculine gender rate of the microscope smear method was 43.5%,the false masculine gender rate was 0,the false negative rate was 43.1%.Conclusion The accuracy rate of the pessary is higher than the microscope smear.
    Observation on the Effects of Combined Application of Compound Ivermectin and Tianziqiuchongfen in Expelling Coccidian of Altitude Pika

    Objective To seek the safe and effective method of expelling coccidian of altitude pika.Methods Compound ivermectin and Tianziqiuchongfen(powder for expelling coccidian) were orally administered to altitude pikas with coccidian infection rate of 87.09%.The effects of expelling coccidian were observed in altitude pika.Results After taking the drugs for a day,the positive rate of coccidian infection was decreased to 39.51%,after taking the drugs for 30 days that was decreased to 14.51%,and after taking the drugs for 45 days,no coccidian infection was found in these altitude pikas.Conclusion Combined of compound ivermectin and Tianziqiuchongfen could exert a satisfactory effect in expelling coccidian and treating coccidiosis in the altitude pikas.
    Studies on Reproductive Capacity of Nude Mice in IVC Cages at Conventional Facility

    Objective To observe reproductive capacity of BALB/C-nu in IVC cages at conventional facility.Methods Three pairs of BALB/C-nu stocks(the ratio of female to male was one to one.) were introduced and the BALB/C-nu stocks were mated in pairs,and the expanding population of blood relationship from offspring was reproduced(the ratio of female to male was three to one,or one to one) in IVC cages at conventional facility.Results The average number born from ten births in BALB/C-nu stocks was 8.6±1.02(nude mice: 1.82±0.63),the average number born from 42 births in the expanding population of blood relationship(the ratio of female to male being three to one) was 6.92±1.47(nude mice: 3.52±1.29),and the average number of nude mice born from 12 births in the expanding population of blood relationship(the ratio of female to male being one to one) was 4.25±1.36.The quality of the nude mice was in keeping with SPF standards.Conclusion The certified nude mice could be reproduced in IVC cages at conventional facility if only the rigorous indoor disinfection and sterile working on purification working stable were carried out.

    Objective To delve into methods of reducing the running cost of barrier facility for laboratory animal under satisfying the demands of environmental standard of the barrier facility for laboratory animal.Methods Two running models were adopted in this series.The air conditioning unit was turned for one hour and then was turned out for two hours when the outside temperature was between 30℃ and 35℃ in the first running model,and turned on for one hour and then turned out for one hour when the outside temperature was above 35℃ in the second running model.Result The running cost was decreased by the two running models.Conclusion It was obvious to cut down greatly expenses in running of barrier facility for laboratory animal according to the two running models.
