    An Approach to Normal Range of Blood Routine and Biochemical Indexes of SPF Wistar Rats

    ObjectiveTo determine normal ranges of blood and biochemical indexes in laboratory rats for toxicological safety evaluation,and compare the influence of blood extraction by decapitation and by abdominal extraction after anaesthesia on biochemical indexes tests.Method Determine the normal range with 95% confidence level through normal distribution by processing statistics of all blood and biochemical indexes in 2~3 month-old SPF Wistar rats collected over recent 3 years by our laboratory;compare the differences of results from male and female rats and two extraction methods.blood indices.Result(1)There is no difference in WBC,RBC,HGB and PLT counts between males and females,but there is difference in WBC classification between them.(2)Biochemical indexes showed difference between males and females.In results of tests of blood by decapitation ALT,AST,TG in females are lower than in males,but their TP,Alb,TCH are higher than males.Therefore there is statistical significance(P<0.05).In results of tests of abdominal blood,ALT,GLU,TG in females are lower than in males and their AST,TP,Alb,BUN,TCH are higher than males and there is statistical significance(P<0.05).(3)Methods for blood extraction do have influence on biochemical test results.ALT,AST,TP,Alb,BUN,TCH,TG levels from tests of blood from decapitation are higher than from tests of blood from abdomen after pentobarbital sodium anaesthesia and there is statistical significance(P<0.01);GLU,CRE levels from test of blood through decapitation are lower than from the latter method and showed statistical significance(P<0.01).Conclusion The normal ranges of blood and biochemical indexes for Wistar rats were therefore established.

    ObjectiveTo obtain the reference value of normal hematological and biochemical indexes of 10 kinds of usual SPF grade rats and mice.MethodsThe some hematological and biochemical indexes were determined by semi-automatic biochemical analyzer and hemacytometer in 10 kinds of usual SPF grade rats and mice of different ages and sexes.ResultsThe some hematological and biochemical indexes of 10 kinds of usual SPF male and female rats and mice at four and eight weeks of age were obtained.Rather striking age-related and sex-related differences have been observed concerning some biochemical indexes in those rats and mice.The regular change of alkaline phosphatase was found in the rats and mice of different strains,ages,and sexes.ConciusionThere are rather striking age-related,sexrelated,and strain-related differences in some hematological and biochemical indexes of usual SPF grade rats and mice,which should be considered in the basal biological researches.

    ObjectiveTo delve into the changeable regulation of sex hormones in a sexual cycle in female Wistar rats and NIH mice.Methods The different periods of a sexual cycle in female Wistar rats and NIH mice were defined by vaginal smear examination,and FSH,LH,and E_2 levels in serum at different periods of a sexual cycle were determined by radioimmunoassay.Results In female Wistar rats and NIH mice,the difference in E_2 level of serum between the different periods was not significant.LH level was the highest in Wistar rats and NIH mice of superovulation,and the difference in LH level of serum between Wistar rats and NIH mice of superovulation and those at other different periods was very significant statistically(P<0.01).FSH level was the highest in Wistar rats and NIH mice of metestrus,and the difference in FSH level of serum between the metestrus and other different periods was very significant statistically (P<0.01).Conclusion The levels of FSH,LH,and E_2 in serum of female Wistar rats and NIH mice at the different periods of a sexual cycle are regularly changeable.

    ObjectiveTo carry out comparative studies on pharmacological effects five kinds of antifluorine Chinese herbal compound powder formulated according to the theories of the modification based on flavor and action of different Chinese herbs,and establishing of therapeutic principles based on syndrome differentiation in traditional Chinese medicine.Methods All the Wistar rats were randomly divided into 7 groups,namely,5 therapeutic groups with anti-fluorine Chinese herbal compound powder,negative control group,and positive group.The rats of the negative group drank freely normal tap water and the rats of the other groups drank the water containing 100 mg fluorine per liter.Five kinds of anti-fluorine Chinese herbal compound powder were orally administered to the rats of therapeutic groups at a dose of 0.1g/kg respectively.The dental fluorosis was observed and content of fluorine in bone,malondialdehyde levels and activities of total superoxide dismutase,catalase,and glutathione peroxidase in plasma,liver,and kidney were determined when finishing of the test.Results The fluorosis was found only in the rats of positive group.The anti-fluorine Chinese herbal compound powder could obviously decrease the content of fluorine in bone,inhibit significantly the lipid peroxidation and enhance the antioxidase activity in the rats.Conclusion The anti-fluorine Chinese herbal compound powder could depress the generating of fluorosis by inhibiting the lipid peroxidation and enhancing the antioxidase activity in the rats.

    ObjectiveTo compare the physical characters and animal adaptability of pine shavings,maple shavings,rice straw,and paper in order to find out the new-type bedding material that is more beneficial to growth,development,and reproduction of laboratory animals.Methods The tests for water absorbability,rising in temperature,dropping in temperature,and animal adaptability of pine shavings,maple shavings,rice straw,and paper were carried out in this series.Results The maple shaving was superior in source,price,and storage.Conclusion The maple shaving should be developed industrially.

    Objective To observe and analyse the effects of varied room temperature on the rabbit's body temperature.Methods The body temperatures of rabbits were taken according to the pyrogen test norm described in "Requirement for Biologicals of People's Republic of China(2000 Edition)".Results Within the certain range of room temperature,the passing ratio of the rabbit's temperature screening was increasing along with heightening of the room temperature.The passing ratio of the rabbit's temperature screening was the lowest(43%) at 20℃~21℃,while it was the highest(71%) at 24℃~25℃.At 22℃~25℃,the body temperature of more than 50% rabbits was at 38.5℃~39.0℃.Conclusion The change in room temperature could exert an effect on rabbit's temperature.Thus screening of rabbits should be performed according to their temperature before the pyrogen test.
    Observation on the Anesthetic Effects of Su Mian Xin Ⅱ and Sodium Pentobarbital on Experimental White Miniature Pigs

    Objective To observe the anesthetic effect of Su Mian Xin Ⅱ on experimental white miniature pigs.Methods30 adult white miniature pigs were randomly divided into group A,group B,and group C.Simple anaesthesia was performed by intramuscular injection of Su Mian Xin Ⅱ in group A,and combined anaesthesia was performed in group B and group C by intramuscular injections of Su Mian Xin Ⅱ and sodium pentobarbital based on different dosage proportion of the two drugs.The induction period of anaesthesia,maintain duration of anaesthesia,and analepsia period,and temperature,breathing rate,and heart rate during anaesthesia were observed and recorded in the three groups.ResultsIn group A,B,and C,the induction period of anaesthesia was 11.4±7.85 min,5.7±2.51 min,and 4.7±2.83 min respectively,maintain duration of anaesthesia was 122.7±42.82 min,104.4±27.68 min,and 122.8±19.59 min respectively,and analepsia period was 47.8±46.04 min,85.9±92.64,and 102.1±59.62 min respectively.The temperature of all the pigs from the three groups has been trending down from injection of the drug to analepsia.The breathing rate of all the pigs from the three groups has been trending upwards from 10 min to 90 min after injection of the drug,and then was decreased slightly during analepsia period.Heart rate of all the pigs from the three groups was gradually decreased from injection of the drug to analepsia.Conclusion Experimental white miniature pig may possess a stronger toleration to Su Mian Xin Ⅱ,and single application of Su Mian Xin Ⅱ or combined application of it and sodium pentobarbital could achieve more satisfactory anesthesia effect.

    ObjectiveTo observe the change of triglyceride,blood glucose and Uric acid at different time in normal Quail serum,and provide partial reference for study by quail as a kind of experimental animal.Methods To breed the normal male quails in compliance with conventional and then at different time to detect related biochemical indicators:determining blood sugar by glucose oxidase endpoint /GOD-PAP;determining uric acid by uricase-POD;determining triglyceride by endpoint GPO-PAP.Results The value of blood glucose,Uric acid and triglyceride is(7.09±3.54)mmol/L,(118.4±71.1) μmol/L and(0.79±0.16)mmol/L respectively.The maximul value is(13.53±1.03)mmol/L,(244.2± 96.7)μmol/L and(1.32±0.35)mmol/L respectively.The average value is(11.28±2.20)mmol/L,(176.66±70.11)μmol/L and(1.00±0.28) mmol/L respectively.Conclusion The result provide some reference for the change of blood glucose,Uric acid and triglyceride in normal Quail serum.

    ObjectiveTo study the effects of testosterone of physiological dose on glucose and lipids metabolism of male aging rats with diabetes.Methods The male aging SD rats were injected with alloxan through tail vein,and after 72 hours the rats with increase of fasting blood-glucose and abnormality of glucose tolerance test for 2 hours were selected and randomly divided into the control group and the treatment group.Testosterone propionate was intramuscularly administered into the rats of the treatment group at a dose of 2 mg/kg,two times every week.After treating for 35 days,fasting blood-glucose,blood-lipoids,and glucose tolerance were determined.Results After treating for 35 days,fasting blood-glucose,and blood-glucose of glucose tolerance test for 30,60,and 120 minutes in the treatment group were significantly lower than those in the control group.The triglyceride level in the treatment group was significantly lower than that in the control group,and the differences in total cholesterol,low density lipoprotein,and high density lipoprotein levels between the two groups were not significant.Conclusion Testosterone is able to produce a decrease in the fasting blood-glucose level and improve the glucose tolerance in the male aging rats with diabetes.

    ObjectiveTo investigate the complications caused by pentobarbital sodium anesthesia in Beagle dogs and their treatments.Methods Sixty-four Beagle dogs were used in this series.They were anesthetized by pentobarbital sodium and the anesthesia complications were observed.Results The anesthesia complications were found in seven Beagle dogs.Among them,six cases were cured and one was dead.Conclusion It is very important for lowering the dog's death rate to treat the complications caused by pentobarbital sodium anesthesia promptly and properly.

    ObjectiveTo establish the serological method for detection of porcine parvovirus(PPV).MethodsFirstly,the swine serum containing the antibodies against PPV was absorbed by the PK-15 to eliminate nonspecific reaction,next,the optimal reaction was determined by concentration gradient titration of the serum and the second reaction-antibodies,and then,the sensitivity among the reaction systems was compared.Results 1 TCID_50 could be acquired with IFA,however,only 100 TCID_50 could be acquired with the other two methods. ConclusionIFA method was more sensitive than the other two methods.

    ObjectiveTo test whether that we have builded cell laboratory operating instruction satisfy the requirement of cell culture by culturing mice primary histiocytes.Method Culture mice cells: osteoblast,cartilage cell,cardiac muscle cell,hepatic cell,lung cell,brain cell,marrow MES cell,embryo fibroblast and calculate primary culture survival rate,passage survival rate,freezing-revival survival rate.Results The primary culture survival rate of osteoblast,cartilage cell,cardiac muscle cell,hepatic cell,lung cell,brain cell,marrow MES cell,embryo fibroblast respectively: 89%,96%,100%,100%,100%,80%,100% and 90%;passage survival rate: 76%,93%,100%,92%,100%,66%,80%,92%;freezing-revival survival rate: 100%,100%,100%,100%,100%,100%,100% and 100%.Conclusion Through calculating primary culture survival rate,passage survival rate,freezing-revival survival rate of 8 kinds of mice histiocytes,it is prove that we can succeedly culture revival common primary cell by using cell laboratory operating instruction at the present stage.

    Objective To delve into feasibility of replacing Draize eye irritation test with Hela-MTT test(MTT being reduced in alive Hela-cells) in evaluations of cosmetics toxicity.Methods22 kinds of cosmetics were examined in this series by Hela-MTT test and Dralze eye irritation according to procedures and methods of eye irritation test in'Hygienic Standards of Cosmetic' promulgated by Ministry of Health,PRC in 1999(HSC-1999) and 2002(HSC-2002). The results obtained in Hela-MTT test were compared with those obtained in Draize eye irritation test.ResultsThe correlation coefficient between Hela-MTT test and Draize eye irritation test in HSC 1999 was-0.811,and that between Hela-MTT test and Draize eye irritation test in HSC-2002 was -0.639.ConclusionIt was suggested that Hela-MTT test could replace Draize eye irritation test in evaluations of cosmetics toxicity.
