To improve the rate of fertilization,the productive capacity of C_(57)BL/6 mouse was studied and the results showed that it was important to characterize the estrus among sexual cycles.The quality of zygotes obtained by natural mating was much better than that by superovulation,and the mouse kitten by natural mating survived highly,while superovulation was still an effective auxiliary method to get more zygotes.The mice sexual cycles were influenced remarkably by light,temperature,noise,air humidity and nourishment.

Objective To study the effects of maca and american ginseng on relieving physical fatigue.Methods 4 experiments were carried out,in each of which 70 mice were used and they were divided into 7 groups: deionized water,0.04,0.4,1.2g/kg of maca,0.04,0.4,1.2g/kg of american ginseng,administrated to each group for 30 days,respectively.The four tests were used to determine swimming time with weight loading;serum urea instant after exercise;hepatic glycogen;and blood lactose before or instant after or 20 minutes after exercise.Results Compared with the control,0.4,1.2g/kg of maca could extend swimming time with weight loading in mice(P<0.05,P<0.01),0.4,1.2g/kg of american ginseng could extend swimming time with weight loading (P<0.05,P<0.01);maca had noeffects on serum urea(P>0.05),1.2g/kg of american ginseng could reduce serum urea after exercise(P<0.05);maca and american ginseng could increase hepatic glycogen;0.4,1.2g/kg of maca could decrease the fluctuation of blood lactose before and after exercise(P<0.01),american ginseng had no effects on it.Conclusion Both maca and american ginseng can relieve physical fatigue,but they have different effects on blood lactose and serum urea in mice.

Objective To observe alcohol-dispelling effect of BUZUIDAN on drunken mice,and explore the mechanism of action.Methods: ①Male BALB/c mice were divided into three groups randomly: normal saline group,KingDrink group and BUZUIDAN group.Each mouse was administered with Beijing spirit er guo tou by gastric perfusion for three times,0.06mL once at 10 minutes intervals,and the total volume was 0.18mL.Following each alcohol administration,0.06ml of normal saline,KingDrink and BUZUIDAN were respectively fed to each mouse of the corresponding group immediately by gastric perfusion.The time period were recorded for each mouse getting drunk after the first alcohol administration,as well as the number of drunken mice.②Repeating the experiments mentioned above,effects on plasma alcohol concentration in mice.Blood were collected and plasma was dissociated from each mouse in 50 minutes following the first alcohol administration,and then alcohol concentration was determined.Results: ① The number of the drunken animals was 2/10 both in BUZUIDAN group and in KingDrink group,which was obviously less than that in the normal saline group with 9/10 drunken animals.There is no significant difference in the mean time of animal getting drunk when the BUZUIDAN group and KingDrink group were compared to the normal saline group;②Plasma alcohol concentration of the mice in the BUZUIDAN group was lower than that in the normal saline group(P< 0.05)significantly.Conclusion: BUZUIDAN can effectively relieve drunkness in mouse,and the mechanism of effects was possibly associated with lowering plasma alcohol concentration.

Objective To probe into the outlines of application and reproduction of animal model of Alzheimer's disease caused by lesion of the basal forebrain cholinergic neuron and to introduce the development of comparative biology of the model.Methods The animal model was developed by injection of Ibotenic acid in nucleus basalis of Meynert,and the abilities of learning and memory were evaluated by water maze.Results This treatment could induce the decline in abilities of learning and memory in the operated rats,however,there was marked different between individuals with variation coefficient from 27.1% to 46.2%.Thus 95% confidence interval of the score in water maze was selected as a standard to filtrate the animal model.The eligibility rate was 50% to 70%,which indicated that it was important filtrate the animal model after the reproduction.Conclusion the developed animal model by lesion of basal forebrain cholinergic neuron could play an important role in studying Alzheimer's disease,but the attention should be paid to the methodological aspect in the practice.

Objective To establish the animal model of spleen deficiency(NUD) in Wistar rats,and study the therapeutic effects of TCM.Method 30 Wistar Rats were divided into blank contral group,model group and Chinese herb treatment group.The body weight,intestinal propulsion and plasma motilin were observed in 30 days.Result The body weight increased in the blank control group,intestinal propulsion was higher in the blank contral group,and plasma motilin was lower in this group.All results in Chinese herb treatment group were higher than those in the model group(P<0.01).Conclution The animal model of spleen deficiency can be established by Fanxieye.The therapeutic effects of Chinese herb is good.

Objective Kainic acid was used in inducing epilepsy model in gerbils.The results showed that the model could be established by intraperitioneal injection of four doses of KA(5,10,15,20mg/kg), and it also showed the latent period and the time of seizure was significantly reduced (P<0.01) and the dead rate was increasing significantly(P<0.01).It indicates that the dose of 5mg/kg is perfect in establishing epilepsy model in gerbils.

Objective To establish me model of nausea and vomiting in Guizhou miniature pig.Methods The animals were divided into three groups at random and given with Cisplatin(DDP) via i.p.once.The conditions of nausea and vomiting were observed within 8 hours after injection.Results During the experiment,the conditions of nausea and vomiting were not observed in the control animals.0nly 25% of the animals in the group given small dose of DDP((3 mg/kg)) had the behaviors and symptoms of nausea at 382 min after injection but without vomiting.Nausea and vomiting were observed in all the animals in the group given large amount of DDP(4 mg/kg),and the average time of nausea onset observed was 68.25 min,and the mean time of vomiting onset was 138 min.Conclusions Significant dose-dependent effect was observed in the Guizhou miniature pig with homogeneous response,which indicates that Guizhou miniature pig may serve as a new experimental animal for disease model establishment used in the studies of vomiting mechanism,anti-vomiting drug and acupuncture.

Objective To study the anesthetic effect and application of drugs "Su-Mlian-Xin Ⅱ" on Guizhou miniature piglets under surgical operation.Methods 30 Guizhou miniature piglets were randomly divided into two groups:Group A and Group B.Group A were anesthetized by muscle injection of drugs "Su-Mian-Xin Ⅱ"(0.15mL/kg),while Group B were anesthetized by muscle injection of drugs !Su-Mian-Xin Ⅱ"(0.1 mL/kg) and 3.5% sodium pentobarbital solution(0.5mL/kg) simultaneously.Both groups underwent operations such as cleft palate casting,organ resection,peritoneo-probing and debridement etc.Both groups were observed in respect of stage of anesthesia induction,stage of maintain,and stage of analepsia,in addition,the heat,respiration rate and heart rate variation of piglets.Results Group A and Group B were recorded respectively with stage of anesthesia induction(7.67±5.20)min vs(3.58±3.20) min,stage of maintain (40.17±4.96)min vs(60.50±17.37) min,and stage of analepsia(7.00±2.45) min vs(56.27±33.60)min.As for the heat,a change of going down and then up was shown in Group A while constant drop was shown in Group B as a whole.Respiration rate and heart rate remained stable in both groups. Conclusion Drugs "Su-Mian-Xin Ⅱ" have satisfactory effects on surgical operation for Guizhou miniature piglets pertaining to anesthesia,pain easing,pacifying and muscle relaxing.Drugs Su-Mian-Xin Ⅱ" have good effects on anesthetic effect when compounded with sodium pentobarbital.

Objective To establish and optimize an animal model for fatty change of liver with clinical characteristics.Methods Comparative analysis of two kinds of duck fatty change model of liver was established.The ducks of model I group were fed with high-dose high glucose diet for 28 days.The ducks of model Ⅱ group were fed with low-dose high glucose diet for 49 days.The index of body weight,ratio of liver weight/body weight,blood sugar serum grease,liver grease,liver pathology was observed.Results The body weight,ratio of liver weight/body weight,serum cholesterol,serum triglyceride and blood sugar of two model groups were higher than that of the control group.Pathological alteration of hepatocyte steatosis in liver was observed in both model group.The liver triglyceride of model Ⅱ group was higher than that of the control group.Conclusion Two methods can replicate the fatty change model of liver successfully,but the model I is better than the model Ⅱ.

Objective To investigate whether Evans blue can be adapted as a method for quantitating blood-retinal barrier breakdown.Methods The content of Evans blue in the retina of normal and 1 week diabetic Brown-Norway rats were detected by absorption value after intravenous injection.Results Retinal Evans blue leakage of eyes from 1 week diabetic rats was(23.41±4.47)ng/mg(n=12),which was much higher than(13.22±3.59)ng/mg(n=12) from normal ones(P<0.05).Conclusions The Evans blue technique can be modified to a sensitive and quantitative method for measuring blood-retinal barrier breakdown.

During the animal experiments,the author explored a new method of stomach-lavaging of rat,which needed cooperation of two people,92 rats can be finished per hour.During the 30 days of rat stomach-lavaging continuously(40/d),there were no unexpected death because of unsuitable lavage,and the rate of success was up to 100%.During adminstration,the most critical thing is how to fix the rat skillfully and effectively.One person fixed rat with two hands,additionally using metal shelf and rope to fix the head of the rat,and kept the head and body in line,which can assure tubatton to go on smoothly.
