Objective This study aimed to observe the histomorphological change of prostate induced by different sex hormone in castrated mice.Methods A total of 40 balb/c mice were randomly divided into 5 groups:normal control(Group N);castration group(Group C);3-estradiol treated group(Group E);testosterone treated group (Group T);B-estradiol and testosterone treated group(Group R).Four weeks later,serum androgen and estrogen levels were assayed by ELISA.The prostate indexes were calculated and histological change was examined by HE staining.Results In comparison with normal group,the prostate indexes were increased in groups treated with different sex hormone.which was most obvious in Group R(尸<0.05).Epithelial cell atrophy was noted in Group C by HE staining.Remarkable prostatic epithelial hyperplasia was observed in Group T but stromal hyperplasia is not obvious.The marked stromal hyperplasia was seen in Group E and prostatic epithelium was significantly reduced relative to normal prostate.Both Prostatic epithelial and stromal hyperplasia were observed in Group R.In addition,inflammatory infiltration around the vessels and acini was viewed in Group R.C:onclusion The components and degrees of prostatic hyperplasia were different with sex hormone status.The androgen mainly caused the prostatic epithelial hyperplasia hut not the.stromal hyperplasia and the obvious stromal muscle’s hyperplasia around the anci was seen after the estrogen treatment.The epithelial and strumsl hyperplasia associated with inflammatory infiltration were induced by the treatment with androgen and estrogen.

Objective In this study,pregnant mare serum gonadotropin(PMSG)and human chorionic gonadotrophin(hCG)were used to make treatments of superovulation with different strains and ages of mice, meanwhile to obtain a better way for superovulation,the dose of hormones(PMSG/hCG),mouse strains and weeks on the effect of superovulation were compared.Results (1)the hormone doses(with 10 IU)were better than those with 5 IU,but no significant difference wa$observed,(2)when the mice were injected with determined dosage of hormone,the effect of supemvulation with IC R mice and C57 BL/6J mice were significantly better than the BALB/c mice and FVB mice(尸<0.05),(3)meanwhile,the effect of mice with 8 weeks were better than those with 5 weeks without significance(P>0.05).Conclusion In this laboratory,using the hormone doses of lO IU PMSG 4-10 IU hCG to 8 weeks old ICR.C57BL/6J were relatively good effect of superovulation.

Abstract:Objective This paper will study the expression of NOS in the Hippocampus Neuron after stopping training 28 weeks by Using Immunohistochemical Technique.Methods Refer to the training scheme of Bedford, Establishing training model of SD rats.All rats in the normal cage live 28 weeks for observing the expression change of NOS in the Hippocampus Neuron.Results Aerobic training promotes NOS expression in CAl m'ea and CA3 area in hippocampus Neuron,Fatigue training restrain the NOS expression in CAI and CA3 ares of hippocampus Neuron After stopping training long—term.Conclusion Making an analysis,Aerobic training has taken good care of the Hippocampus Neuron of the aged rats.It would benefit postponing senility and promoting memory.Fatigue training has affected the function condition in the Neuron of Hippocampus of the aged rats.

Abstract:Objective The aim of this study was to evaluate the ureteral motility of rats in vivo and to determine the changes in density and morphology of interstitial cells of Cajal(ICC)一like ceils(ICC—LC)at the ureteropelvie junction(UPJ)of rats after artificial distal ureteral obstruction.Then,lhe correlationship was assessed between these two aspects.Methods Of a total of 35 rats,5 served a8 controls(C),5 underwent sham—operations(SH) and 25 were in the study(S)groups.The left ureters of the rats in the S groups were tied with 5-zero silk ligature approximately 3 mm proximal to the ureterovesical junction(U VJ).The ureteral motility of rats was measured by external pressure transducers.The UPJs were extracted initially in the C and SH groups,and 3,7,14,28,56 days after ligation of the distal ureter in the study groups(S1,S2,S3,S4 and S5,respectively).The sections stained by c-kit anticore were studied under a light microscope.Results Ureterohydronephrosis wilt8 observed in all the rats after the obstruction was established.The mean number of ureteral peristaltic rate was(1 9.6±4.037)/ min in C,(18.8±2.168)/min in S,(4.6±1.140)/min in S1,(8.0±2.345)/rain in S2,(5.4±1.673)/ min in S3,(5.4±1.517/min in s4,and(6.2 4-1.789)/min S5.There was a statistically significant decrease in ureteral peristaltic rate in the study groups compared to the C and SH groups.The mean number of ICC—LC was (7.20±2.29)/HP in C,(7.02±2.68)/HP in SH,(3.54±1.85)/HP in S1,(6.30 4-1.19)/HP in S2, (4.30 4-0.67)/HP in S3,(4.42±1.45)/HP in s4,and(4.70±1.09)/HP in S5.There w鹳a statistically significant decrease in ICC·LC density in the study groups,except S2,compared to the C and SH groups.No significant difference was detected in Cajal cell morphology between groups.Conclusion Based on the changes in density of ICC-LC at the UPJ after obstruction of the distal ureter.we suggest that ICC-LC have a close relationship with motility of the ureter.

Objective To establish a muhiparous rat cycloma8topathy model.Methods Twenty muhiparous female rats were divided into normal and model groups,and the latter were injected through muscular with oestradiol benzoate for ten times at a dose of 1.0 mg·kg~once every two days.From the last injection of oestradiol benzoate, luteosterone was given to rats for 5 days.In the whole procedure。rats were enclosed into fastening cages at night every two days for 1 5 times.After last injeetion of luteosterone,rats were sacrificed and galactophore were collected.Resul侮The number of gland alveolus increased more obviously than that of the normal groups,the volume of gland alveolus inflated.the acinar luminas expanded and the wall of gland alveolus was thinner and filled fully with basophilia secretion.All of those changes didn’t recover to normal levels in 3 weeks.Conclusion Muhiparous rat cycloma8topathy model can be established by hormones induction combined with fastening method. According to our study。this method is reliable。effective and simple.

Abstract:model of rats.Methods Subcutaneous injecting adrenalin hydrochloride twice,during the period,stimulating the rats with the iee water to induce the acute blood stasis model to survey the influence on the hemorheology.Results The low dose group(18 g crude drug/kg)、medium dose group(369 crude dms/kg)、higher dose group(72 g cmde drug/kg)of Renal Clearance Granule could significantly reduce the whole blood viscosity,plasma viscosity, erythroeyte electrophoresis time,the cell aggregation index,erythrocyte deformation index,erythroeyte rigidity index.Conclusion It was indicated that the Renal Clearance Granule could significantly improve abnormal change of hemorheology on actue blood stasis model of rats.

In May 2010 and Ocluber 2010 wc have taken 2I 9 and 230 rats from 10 and 14 units respeclivcly investigate the health status of experimental rats.All of these rats were killed by cutting off the cervical.We collected the heart,lung,liver,spleen,kidney,large intestine and small intestine,and then put these organs into10%paraformaldehyde solution for fixation.The samples were then observed with optical microscopy.The results show that the vast majority of experimental mice were pathologically diagnosed as healthy animals.Fatty liver and bronchial pneumonia are the two main lesions which affect the health of laboratory animals.The reason may be breeding and feeding environment and we can improve the management to ameliorate the health status of experimental rats.

Objective To observe the effect of chinese medicine preparation for improving the reproductive rate of laboratory Beagle dogs.Methods 24 female Beagle dog about l~2 years old were divided into a lab group and a control group randomly.They were fed different diets respectively:control group for foundation date grain,lab group for the foundation date grain and chinese medicine preparation.The experiment had mated repeatedly and recorded the estrus rate、the conception rate、the farrowing rate and the litter size of female Beagle dog.Results Compared with two groups,the estrus rate had risen from 66.67%to 100%;the conception rate had risen from 71.43%to 91.67%;the farrowing rate had risen from 80.00%to 90.91%;the litter size of female Beagle dog had risen from 6.80 to 10.18.Conclusion Chinese medicine preparation could improve the reproductive rate of laboratory Beagle dogs significantly.

Objective The aim of this study is to summarize the characters,limitation and technical main point of Bama and Guizhou mini—swines’coronary artery and evaluate the application of the two strains of swine in coronary stent model.Methods Nine Bama and 12 Guizhou mini—pigs were used,male and female.After general anesthesia,the animals were weighed and the right femoral artery was exposed.Then coronary angiography was performed and recorded.Coronary stent was chosen according to the diameter of left anterior descending and right coronary artery by visual observation and implanted by overexpansion(artery diameter:stent diameter=1:1.2). Coronary angiography was repeated to investigate the adherence of stent.The pigs were fed a standard laboratory chow after stinting until they were anesthetized again 26 weeks later.The style,curvature and diameter of coronary artery were detected by catheter doctor depending on the coronary angiography.Results All animals survived 26 weeks and had right—side dominance type coronary artery with curvature<30o.There was no statistic difference in the diameter of LAD between Bama and Guizhou mini·swine(P>0.05).The diameter of RCA in Bama was smaller than that in Guizhou(P<0.05).The diameter of stent was no statistic different either in LAD or RCA between Bama and Guizhou(P>0.05).The weight changing of Bama was bigger than that of Guizhou(P< 0.001).Conclusions In our study。the adult animM of both Bama and Guizhou was found with right—side dominance type just like human.The curvature of LAD or RCA is suitable to intervention.The diameter of stent is 2.75 mm or 3.0 mm can be implanted easily.Both Bama and Guizhou are fit for coronary stent model through a live and dynamic coronary angiography analysis。

Abstract:The bunyaviridae family comprises more than 300 viruses.Membership is usually based on antigenic interrelatedness or morphological similarity.Disease characterized by fever,headache,weakness,myalgia, pulmonary edema.The family is divided into 5 genera:1.Orthobunyavirus:Bunyamwera,La Cross,Tahyna virus, transmitted mainly by mosquitoes.2.Hantavirus:Hantaan virus。transmission does not require insects.3. Nairovirus:Dugbe virus infection of cattle in West Africa,transmitted by ticks.4.Phlebovirus:Sandfly fever,Rift valley fever。transmitted by sandflies.5.Tospovirus:Tomato spotted wilt virus,only infect plant and non·vertebrate. Man is not known to be a natural or reservoir for any of these viruses.Virions are 80~120 nm in diameter.5一10 nm projections visible on the surface.Genome consists of 3 pieces of negative stranded RNA.Virion has 2 surface glycoproteins G1 and G2,with HA and vires neutralization epitopes.Bunyaviridae is a family of negative stranded RNA viruses.Though generally found in arthropode or rodents,certain viruses in this family occasionally infect humans.Bunyaviridae are vector—borne viruses.With the exception of Hantaviruses transmission occurs via an arthropod vector.Hantaviruses are transmitted through contact with mice feces.Crimean·Congo hemorrhagic fever vims is associated with high levels of morbidity and mortality,consequently handling of these viruses most occurs with a biosafe Ievel 4 laboratory.Hantavirus or Hantavirus hemorrhagic fever,common in China,Korea, Scandinavia。Russia,and the American southwest,is associated with high fever,lung edema and pulmonary faihure.Mortality is around 55%of laboratory diagnosis of bunya virus infections.Virus isolation-intra-cranial inoculation of suckling mice is thought to be the most sensitive system available for virus isolation.However sensitive cell culture systems are available such as Vero,Vero E6,A549 and mosquito cells.Once isolated the virus can be types by neutralizing tests.Rapid diagnosis·antigen detection systems and the detection of specific IgM antibodies are becoming available as means of rapid diagnosis.A wide variety of serological techniques is available such as HI,CFT,IFA,NT and ELISAs.Treatment is mainly supportive.The drug ribavirin has encouraging results in vitro and systemic therapy with this agent should be considered in severe cases.Hyperimmune convalescent serum may be useful and those with a high titre of vires neutralizing antibodies should be selected. Abstract:Objective To explore and summary successful points for establish Langendorff heart perfusion models. Methods Through 1 6 healthy SD rats(200-4009 each),using HTK solution for eardioplegia and establishingstandard Langendorff heart perfusion models,and compare myocardial enzymes and hemodynamic with continuous perfusion and beating group.Results 1 case failed because of unskilled operation in extracting the heart;1 case failed because of inserting the perfusion—tube too deeply to damage the aortic valve;1 case failed because of lowly indoor-temperature;1 else failed because of excessive diameter of perfusion—tube:1 2 cases prepare successfully with the application of indoor heaters,perfusion needles with different diameters,self—made balloons and accelerating the operations.There’s no significant difference between cardiac resuscitation group and continuous heaa beat group in myocardial enzymes and hemodynamic.Conclusion After mastering the attentions above,isolated heart perfusion models can be established successfully and quickly.

Objective To study the reproduction method of experimental gingivitis and the objective indication for evaluation of the level of inflammation in rat.Methods The animal model of gingivitis was reproduced by suturing the neck of teeth and feeding of high sugar diet in rat.Local blood flow and crevicular fluid aspartate aminotransferase levels were evaluated by the 7th day after operation.Results The gingiva showed red and swelling and the superficial periodontal pocket formed in the rat of model group.The quantity and velocity of local blood flow increased in the model group.Cell number in blood flow significantly increased(P<0.05).The level of aspartate aminotransferase in crevicular fluid dramatically increased(P<0.00 1)in comparison of control group.Conclusion This is a reliable animal model of gingivitis.Local blood flow and crevicular fluid aspartate aminotransferase level are sensitive indication for the rat model of gingivitis.
