Objective To clone,express and characterize the H5 N 1 avian influenza virus hemagglutinin(HA)and neuramidinase(NA)genes.Methods Expression vectors,pMET A/HA(49一I 587 bp)and pMET A/NA(121· 1 200 bp)were constructed and transferred into the pMADl 6 yeast.Recombinant HA and NA were expressed inducibly by methanol,and purified through Ni2+affinity chromatography column.Antigeneity of the recombinant proteins were determined using Western Blotting and ELISA.Results The recombinant genes can be highly expressed(more than 95%of total proteins)in P.methanolica.SDS—PAGE showed that the products[tle dimmers,and Western Blotting and ELISA showed that the recombinant proteins have good antigencity.Conclusion The HA and NA proteins of H5 N1 avian influenza virus can be suceessfully cloned and expressed in P. methanolica.

Objiective To establish the haploidentical bone marrow transplantation mouse model in nonmyeloablative conditioning,which will provide a platform for studies on inducing immune tolerance and promoting engraftment after transplantation.Methods One day before transplantation,the receptions CB6F1 female mice were exposed to a dose of 450cGy total body irradiation(TBI).Then they were divided into two groups randomly,and each mouse of the experiment group wag transfused with 5×1 07 nucleated bone marrow cells from the donor C57BL/6 male mice.Recovery of receptions hemogarm,engraftment of the sex determining gene(SRY),chimerism of the donor cells,especially the CD3 ceHs,and signs of aGVHD after transplantation,were examined.Results All control mice survived and the hemogram almost recovered to normal level within thirty days after transplantation.Most control mice shown slight graft·versus-host disease.For the experiment group,the sex determining gene SRY can be detected in the peripheral blood from receptions.Chimerism of the donor cells at day 14,30 and 60 after transplantation wag 23.8%,49.9%,67.6%in lymphocytes,36.9%,53.2%,5 1.6%in monocytes。and 19.4%,54.4%,56.9%in granulocytes,.In CD3+cells,chimerism rates were 4.4%,21.2%,and 54.4%, respectively.No significant signs of graft·,versus-host disease were observed.Conclusion In the nonmyeloablative conditioning,single dose 450 cGy TBI one day before transplantation can induce immune tolerance of receptions and enable donor bone mgITOW cells engraftment.The chimerism of donor cells was low until the end of observation.

Objective To evaluate the effect of Belamcanda chinensis extract on mouse immune function.Methods Immunosuppressive mouse model induced by cyclophosphamide was employed to determine the carbon clearance index of reticuloendothelial cell(RES,mononuclear macrophage)after peritoneal injection of the Indian ink,and the effect of Belamcanda chinensis extract on the nonspecific immune function was examined.The serum hemolysin level was tested to survey the effect of Belamcanda chinensis extract on the humoral immunity.Results Belamcanda chinensis extract could enhance the phagocytosis of the mouse reticuloendothelial cells,and the producing of hemolysin.Conclusion Belamcanda chinensis extract significantly improves nonspecific and specific immunologic function in mice.

Objective To investigate the anti—inflammatory and immunological effects of Guben Zhike Pingchuan Granule.Methods The anti.inflammatory effect was studied on the acute and the chronic inflammation models induced by xylene and agar,The effect of Guben Zhike Pingchuan Granule on the humoral and the cellular immunity induced by chicken red blood cells and 2.4-dinitr-ochlorobenzene was investigated.Results Guben Zhike Pingchuan Granule significantly prevented the acute and the chronic inflammation,and modulates the humoral and the cellular immunity.Conclusion Guben Zhike Pingchuan Granule has anti-inflammatory and immunemodulatory effects.

Objective To explore the effects of pilose antler on pregnant and fetal mice.Methods Kunming mice were divided randomly into two groups.In group A,there were 24 males and 48 females,and they were caged together at the rate of 1:2(male:female).From the fifth to the fourteenth day after pregnant。females were administrated by oral gavage with pilose antler at three doses,3 mg·kg一1d一1.6 mg·kg一1d一1 and 12 mg·kg一1d一1.In group B,48 male mice administrated with pilose antler for 60d were caged together with females at the rate of 1 :2(male:female).The conception rate,body weight of the pregnant mice,stillbirth rate,rate of fetal absorption, placental weight,uterine and fetal mice weights,appearance of the fetal mice,as well as viscera and skeletons, were examined.Results Compared to the control group in the 18th day,body weight,stillbirth rate,birth rate of absorption,appearance of the fetal mice,visceral and skeletal malformations in the high dose group of group A were statistically high(P<0.05);with the increase of the dose,there is no statistical differences in the aspect of placental,uterine and fetal mice weights.In group B,middle dose group showed a significant increase in pregnancy rate(P<0.05);With increase of dose,stillbirth rate,birth rate of absorption,weight of placenta, uterine and fetal mice had no significant differences(P>0.05);there is no impact on malformation of the fbtal mice.Conclusion Administration of high·dose pilose antler during pregnancy can decrease the weight of pregnant mice,but increase the stillbirth·rate,absorption rate in births,and fetal malformation of appearance,viscera and skeletons.Middle-dose administration to males can raise conception and does not induce deformities in fetal mice. Abstract:The immunomodulatory effects of LingZe Mixture were examined.In this study,LingZe Mixture could obviously reduce the phagocytic ability of macrophage in abdominal cavity of mice,inhibit the delayed hypersensitivity reaction induced by DNCB,and reduce the$erum hemolysin.The results indicate that LingZe Mixture has good immunomodulatory effects on nonspecific,cellular and humoral immunity.

Objective To establish a hyperurieemia model in SD rats.Methods Different doses of adenine, adenine and yeast,adenine and ethambutol and oxonie acid were administrated to the SD rats by oral gavage。 Concentrations of serum uric acid,urine nitrogen and creatinine were measured,before and after treatments. Pathological examination of re.hal tissue were carried out.Results The serum uric acid of th,e oxonic acid treated groups was increased smoothly,steady and there was no significant renal damage at a long time.Conclusion The oxonic acid is suitable for preparation of hyperuricemia model in SD rats.

Objective To measure the biochemical parameters in blood of Insulin—resistance rat model induced by high.fat and high—carbohydrate diets.Methods Male SD rats were fed on standard laboratory chow,high—fat diet or high—carbohydrate diet respectively for 6 weeks.Serum glucose(GLU),insulin,triglyceride(TG),total cholesterol(TCH)and HDL—cholesterol were measured at fortnightly"interval.The insulin sensitivity was estimated by ISI index,ISI=Inl/(FPG×FINS).Results Compared with control after 6 weeks,ISI index significantly decreased in both high—fat and high·carbohydrate groups(P<0.05),and$ei'um insulin was significantly higher and HDL·cholesterol was significant lower in high—fat group(P<0.05).Serum GLU and insulin in high— carbohydrate group were higher than those in other groups.However there were mild difference in TG、GLU and body weight among the three groups.Conclusion Both hish—fat and high—carbohydrate diets can induce insulin· resistant in rats.High-fat feeding results in significantly hish serum insulin and low HDL·cholesterol,but hi【sh—

Using the pathologic technique,skin structures of Landraee pig and human were compared.According to our study,the skin of Landrace pigs and human is similar,both of them are composed of epidermis,dermis, subcutaneous tissue and cutaneous adnexa.However,compared with human,there are minor differences in pig adnexa:the hair follicle of the pig is more larger;most of sweat glands are apocrine sweat glands,and a few of them are eccrine sweat glands;sebaceous glands are less developed and fewer in number,and lipid droplet in cytoplasm is small.After breeding for several ten years in Xinjiang,nevertheless,the Landrace pigs are still the xenoskin providers for cure of large area deep bums,and the optimal animal models for wound healing, xenotransplant and immunological rejection researches.

0bjective To test and analyze the coagulation time of four kinds of coagulation factors in healthy, captive bred tree shrew and rhesus monkey,and to establish the reference values range of coagulation factors values in the two animals.Methods Prothrombin time(PT),activated partial thromboplastin time(APrr), fibrinogen time(FIB),and thrombin time(TY)of captive bred tree shrews and rhesus monkeys were tested by using the Sysmex CA-510 automated blood coagulation analyzer,then the data were analyzed in using the statistics analysis software SPSS v17.0.Results In Tree shrew and Rhesus monkey.rI-I’was respectively 19.27 s and 20.81 s;PT was respectively 17.34 S and 9.82 s;FIB was respectively 30.51 S and 18.73s;AP胛was respectively 27.88 8 and 33.56 s.The differences in PT and FIB were highly significant(P<0.01)between captive bred tree shrew and rhesus monkey;and the differences in TI"and APr】[-I.were also statistical significant(P<0.05)between the captive bred tree shrew and the rhesus monkey.Conclusion There were significant differences in coagulation time of the four kinds of coagulation factors between animals,but there had no differencees in the different gender of same species.

Objective To investigate the electrocardiogram(ECG)features of rhesus monkeys in consciousness. Methods Sixty healthy rhesus monkeys,2—3 years old,2~3.5 kg,and 30 each gender,were selected.ECG changes of the lead I,Ⅱ,11I,aVR,aVL,aVF in resting conscious monkeys were examined,and the average heaM rate,mean electrical axis,P wave,QRS complex,amplitude of T wave,PR interval,QT interval and so on,were calculated.Results and Conclusions Sinus rhythm was found in all the 60 rhesu8 monkeys.In most of the rhesus monkeys,the P wave was upright in H lead,S wave was absent in 1I、Ul、aVF leads,and there was no Q wave in aVR lead.The average voltage of R wave in female rhesus monkeys was significantly higher than that of the males (P<0.005).In rhesus monkeys,the P wave is sharp,the QRS is diversified,the QT interval K value is 0.298± 0.030,the ST segment locates at the isoelectric line,and the T wave is smooth in six leads and upright in the II lead.In conclusion,we established the basic ECG data of rhesus monkeys.

Abstract:Objective To develop the reference values for serum resistin in rhesus monkeys at various ages using ELISA,a rapid quantitative method.Methods The rhesus monkey were divided into three groups(young,adult and elder groups,10 each.group).Sera were collected and resistin.was detefinined using human—resistin ELISA quantitative detection kit.The OD450 values were analyzed b.y statistic methods。Results The OD450 vslue$ decreased with aging.The mean value was(6 510±1 730)pg/mL in young group,(5 880±l 025)pg/mL in aduh group and(1 760±920)pg/mL in elder group.The value of elder group was significant lower than that of the young group(P<0.0 1).Conclusion The human—resistin ELISA kit is a quantitative and rapid detection tool for resistin determination.’nle reference values of serum resistin in rhesus monkeys are developed.and they decreased with aging.

It is essential to establish a standard culture system for monoclonal Dugesia japonica strains in the lab, since Dugesia japonica has been considered as a potential model in cell differentiation and embryogenesis studies due to its super regenerative ability.In this study,Dugesia japonica was cut into fragments,which then were cultured and regenerated to monoclonal groups.The monoclonal strain was consisted of 5 groups and the size of each group is from 37 to 45.The current technique for culturing Dugesia japonica may provide an useful tool for subsequent genomic and proteomic studies.
