    The Establishment of the Multiplex PCR for Yersinia enterocolitica,Yersinia pseudotuberculosis,Shigella and Campylobacter jejuni

    Abstract: Objective To establish a simple,sensitive,rapid,and the specificity multiplex PCR method for Yersinia bacteria, Yersinia pseudotuberculosis, Shigella and Campylobacter jejuni from laboratory animals.Method According to the fox gene of enterocolitis Yersinia and inv of Yersinia pseudotuberculosis,ipaH gene of Shigella and gyrA gene of Campylobacter jejuni,we designed and screened a pair of specific primers. The annealing temperature and other conditions were optimized,then the specificity and sensitivity of The Multiplex PCR were analysis. At last,the multiplex PCR method was use to detect the samples from mouse and guinea pig. Result The multiplex PCR amplified the expected PCR product, namely Yersinia enterocolitis ( 511bp ) , Yersinia pseudotuberculosis ( 779 bp) ,Shigella ( 290 bp) and Campylobacter jejuni ( 156 bp) . The sensitivity of this method is 1 × 10 - 3 ng /μL ( Yersinia bacteria,Yersinia pseudotuberculosis,Shigella) and 1 × 10 - 2 ng /μL ( C. jejuni) . No specific positive was detected from the other samples. Conclusion The multiplex PCR method has good application and development prospects in detection of microorganisms in laboratory animals.
    Study on Mutagenic Effects of Pantene Anti-hair Shampoo by Ames Assay

    Abstract: Objective To study the mutagenic effects of Pantene anti-hair shampoo by Ames assay. Method The mutagenic activities of Pantene anti-hair shampoo were study in the TA series of typhoid salmonella bacillus strains TA97,TA98,TA100,TA102. Result In the presence and absence of S9 activator,the results of assay were negative at the concentrations within 1. 25 - 0. 156 mg /plate. Conclusion The mutagenicity of Pantene anti-hair shampoo were not observed in the assay.
    Determination of Blood Physiological and Serum Biochemical Values of C57 - ras Transgenetic Mice

    Abstract: Objective To determine the normal ranges of blood physiological and biochemical data in C57 - ras transgenetic mice and compare for variations of these data between the both genders and between 4 weeks mice and 8 weeks mice. Method The blood samples collected were tested for physiological values using MEK - 7222K Hematology Analyzer,Biochemical values using OLYMPUS AU400 auto-analyser. Result Sexual difference of 8 weeks C57 - ras was noted in biochemical values such as ALB、GLU and CHO( P < 0. 05) and physiological values such as WBC and LYM( P < 0. 05) . Age difference of 4 weeks and 8 weeks C57 - ras was noted in biochemical values such as ALT、TP、ALB、ALP、P、CHO and T. Bili( P < 0. 05) ,and physiological values such as WBC、RBC、HGB、HCT、MCV、MCHC、RDW、PLT、PCT、MPV、NEUTand LYM ( P < 0. 05 ) . Conclusion The normal physiological and biochemical data in C57 - ras transgenetic mice are affected by a variety of factors. Some of the data are significantly different between the both genders,and the majority of the data are significantly different between 4 weeks mice and 8 weeks mice.

    Abstract: Objective In embryo - fetal developmental toxicity test for SD rat,different doses of cyclophosphamide were given by subcutaneous injection to study the comparison of relativity between dose levels and clinical manifestation,establish a positive model and accumulate background data for laboratory. Method One hundred healthy pregnant SD rats were taken,randomly divided into solvent control group ( 7. 5 mg·kg - 1 Veh: sodium chloride injection) ,low cyclophosphamide dose group ( L: 7 mg·kg - 1 ) ,moderate cyclophosphamide dose group ( M: 10 mg·kg - 1 ) and high cyclophosphamide dose group ( H: 15 mg·kg - 1 ) . There was not less than 20 pregnant rats in each group. Subcutaneous injection was required and clinical manifestation was observed. Collected the body weight,food intake,litter weight,corpus luteum number,implantation number,the number of live births,the number of stillborn fetus and the number of absorptive fetus of pregnant rats in each group,and the sex ratio,body weight,body length,appearance abnormality,visceral abnormality and skeletal abnormality of fetal rats were also observed. Result Developmental toxicity were observed in all cyclophosphamide dose groups and dose - effect relationship was found obviously. Conclusion Developmental toxicity and obvious dose - effect relationship were observed in all cyclophosphamide dose groups by subcutaneous injection. A positive model of embryo - fetal developmental toxicity test can be set up by all three dose levels of cyclophosphamide.
    Pathological Diagnosis and Analysis of Canine Ear Tumors

    Abstract: Objective 32 cases of canine ear tumors were pathologically diagnosed and analyzed to explore the occurrence regularity. Method The canine ear tumors were obtained from China Agricultural University Animalhospital from 2009 to 2015 by surgical treatment,which were analyzed according to pathological diagnose the occurrence regularity of the location,tissue origin,age and breed. Result The occurrence rate of trichoblastoma and histiocytoma in canine ear were each accounted for 26. 7%. Canine ear tumors occurred in 0 - 4. 9 years old accounted for 20%,5 - 9. 9 years old accounted for 40%,more than 10 years old accounted for 40%. The Tumors all occurred in the external ear,but not in middle ear and inner ear. From the breed,the high incidence was 8 cases of Cocker Spaniel,other varieties were 2 - 3 cases. Conclusion Trichoblastoma and histiocytoma were the often occurred in the canine ear and there was an upward trend of incidence on canine ear tumors with age increase but not on histiocytoma. All the tumors were occurred in the external ear and often occurred in Cocker Spaniel.
    Application of H1N1 Influenza Virus Infection Mice Model for Immunology Research

    Abstract: Objective To find the immunological reactions post H1N1 influenza infection. Method Mice were infected with 0. 1LD50 A/PR/8 /34 ( H1N1) influenza viruses. Level of IL-5 /6 cytokines were tested by ELISA,and ratio of effective T cells were analyzed by flow cytometer. Result The virus loading in mice lung was peaked at 3dpi,and the level of IL-6 in serum was peaked on 5dpi with 122pg /mL. The level of IL-5 in BAFL was peaked at 7 dpi with 680pg /mL,and the ratio of CD4 + CD44 + CD62L-ratio T lymphocytes rised significantly. Conclusion C57BL /6 mice infected with PR8 virus is a good model to study immunology reaction of influenza virus infection.

    Abstract: The Lab-Animals testing is one of the laboratory accreditation field of the China National Accreditation Service for Conformity Assessment( CNAS) . The Lab-Animals as testing objects and subjects,which are widely used in food,medicine,health and epidemic prevention,biological safety,animal quarantine,agricultural and chemical industry,environmental protection and other disciplines. In order to standardize the consistency and effectiveness of the laboratory accreditation field,CNAS formulated the Guidance on the Application of Laboratory Competence Accreditation Criteria in the Field of Lab-Animal Testing ( CNAS-CL58) .     Abstract: Objective 2013—2015 proficiency testing ( PT ) were evaluated in order to find problems in laboratories,which can increase the testing capacity of laboratory animals’quality monitoring organizations.Method According to ISO/IEC 17043,the testing capacity was evaluated by statistics the results of qualitative experiment. Result Totally 174 laboratories participated in the PT. Totally 171 results were received,151 of which were satisfactory. The average satisfaction rate was 88. 3%. The respective satisfaction rate for nine items was 94. 1%,80. 0%,88. 9%,85. 0%,78. 6%,90. 0%,100. 0%,93. 3% and 80. 0%. Conclusion Proficiency testing can help laboratories to increase their testing capacity and strengthen the system construction of laboratory animals’quality monitoring.
    Pathological Monitoring Plays an Irreplaceable Role in Assessing the Quality of Laboratory Rodents

    Abstract: Objective Assess the status of laboratory rodents by pathological diagnosis for offering some advices in standardized breeding to ensure accurate results of scientific research. Method Our laboratory collected the heart, liver,spleen, lung,kidney, large intestine and small intestine of animals,and put these tissues into 10% paraformaldehyde solution for fixation,and then prepared into two different sections for optical microscopy observation including all paraffin specimens stained with H. E. and the frozen sections stained with Oil Red-O and PAS. Result Although the vast majority of laboratory rodents were microbiological,parasitological and genetical diagnosed as healthy animals every years. There still exists different degree of pathological damage in them. Liver cell swelling,slight lung lesion and protein cast of kidney were main lesions which might affect the health of laboratory rodents. Sometimes we also observed various lesions in heart,digestive tract and lymphoid organs.Conclusions These results further demonstrate that though the health levels of laboratory rodents meet the microbiological national standards,there is still a part of them in sub-health status. Pathological monitoring plays an irreplaceable role in assessing the quality of laboratory rodents. Relevant regulatory departments should make the national and local pathological standardized policies for laboratory animals as soon as possible to improve the quality monitoring system of the experimental animals.

    Abstract: Hair follicle stem cells ( HFSCs) normally differentiate into keratinocytes,sebocytes,and transient amplifying progenitor cells. HFSCs have the capacity of rapid proliferation,and thus can provide the basis for establishing a putative source of stem cells for cell therapy. The first section of this review describes the location,culture,and molecular markers of hair follicle stem cells in order to provide a powerful reference for the identification and isolation of cells. The second section of this review presents the discovery and molecular function of Noggin gene which regulates the proliferation and differentiation of HFSCs,and summarizes the advances in regulatory effect of Noggin on HFSCs through BMP signaling pathway.
