Abstract: Some apparatuses such as heart,liver,spleen,lung,kidney,large intestine and small intestine were drawn from laboratory rodents sampling from 16 units of Beijing in May 2012,and were prepared into serial paraffin sections stained with HE and the frozen sections stained with Oil Red O and PAS for microscopic analysis. The results showed that most of the laboratory rodents remained in good condition,and liver fatty degeneration,slight lung lesion,heart granular degeneration and protein cast of kidney were revealed in a few laboratory rodents. We have the impression that these lesions may be associated with feedstuff,feeding environment and transportation stress,and may be improved by strengthening the feeding management.

Abstract: Objective To clone of the large tumor antigen( T-ag-C) gene of simian virus 40 from kidney culture of Macaca monkey and analyze its sequence. Method The 441 bp DNA segment were obtained by PCR from both kidney culture of Macaca monkey and culture of SV40 strain 776 using primers T-ag-C gene of simian virus 40,and they were cloned into the vector PMD18-T for sequencing. The T-ag-C gene from kidney culture of Macaca monkey and culture of SV40 strain 776 were identified by comparison with other simian viruses 40 from gene bank and other reference. Result The sequence comparison with the T-ag-C gene of SV40 Macaca-RMK. seq showed nucleotide homologies of 97. 31% with SV40-Macaca-PBMC. seq,96. 33% with gene bank SV40-GI-9628421. seq, 97. 55% with SV40-strain-776. seq. Conclusion Sequencing DNA of gene is a powerful tool for classification as well as molecular epidemiology of simian virus 40 .

Abstract: Objective To establish transgenic mice brain-specifically expressing insulin-like growth factor -1 ( Insulin-Like Growth Factors,IGF-1 ) . Method Sperm was used for gene transduction as a carrier,postnatal mice were detected by PCR assay to establish transgenic mouse lines. Western blot and immunofluorescence were applied to detect the IGF-1 expression level in transgenic mouse subgranular zone( SGZ) and the amount of reporter gene EGFP positive cells. Result Three positive mice were obtained,2 of them could be inherited stably,and established transgenic mouse lines. The IGF-1expression level in transgenic mouse SGZ was higher than that in the normal mice,meanwhile,EGFP could be expressed in the same area. Conclusion IGF-1 transgenic mice were prepared successfully.

Abstract: Objective To observe and compare the liver and spleen metastasis of transplanted human ovarian cancer,gastric cancer and colon cancer in nude mice. Method The human ovarian cancer,gastric cancer and colon cancer were transplanted to the nude mice. The growth rate of the solid tumors,the survival time,and the pathological changes in the liver and spleen metastasis of three transplanted carcinomas were observed. Results The rates of tumor formation and the liver metastasis of xenografted human ovarian cancer,gastric cancer and colon cancer in nude mice were 100% . The spleen metastasis rate of human ovarian cancer was 100% ,that of gastric cancer was 62. 5% ,and that of colon cancer was 75% . The tumor growth rate of solid tumor of human ovarian cancer in nude mice was higher than that of solid tumor of gastric cancer and colon cancer,and spleen metastasis of human ovarian cancer was rapider with a short survival time. There were differences in the pathological changes in the liver and spleen metastasis of three transplanted carcinomas. Conclusion The models of liver and spleen metastasis of transplanted human ovarian cancer,gastric cancer and colon cancer in nude mice can reproduce clinical processes of the liver and spleen metastasis of three transplanted carcinomas,which makes for the studies on biological mechanism of liver and spleen metastasis of human ovarian cancer,gastric cancer and colon cancer and anti-metastasis

Abstract: Objective To Compare four methods of developing hyperlipidemia in Mongolian gerbils. Method The gerbils in the experimental groups were fed respectively with four high-fat fodders with varying amounts of cholesterol,lard and cholate. The gerbils in the control group were fed with normal fodder. Serum levels of triglyceride ( TG ) ,total cholesterol ( TC ) , low-density lipoprotein cholesterol ( LDL-C ) and high-density lipoprotein cholesterol ( HDL-C) in the gerbils in all the groups were measured at the 0th,30th,60th day. Animals were sacrificed and the morphological changes of the liver were examined by histopathology. Result Serum levels of TC,LDL-C and HDL-C in gerbils of the experimental group were significantly higher than those in gerbils of the control group during the 30th,6 0th days,with a remarkable increase in serum TC level in particular, and increase of serum LDL-C were obviously bigger than the increase of serum HDL-C. There have been varying degrees of fatty liver in the experimental groups. Conclusion Four high-fat fodders can all cause hyperlipidemia in the gerbils.

Abstract: Objective To observe the physical index properties of different experimental rabbits in the SPF breeding environment. Method The 10 SPF Japanese rabbits and 10 SPF New Zealand laboratory rabbits were adopted in this triall. The blood samples were obtained from the heart of rabbits. Result There were differences in some indexes for SPF Japanese rabbits( P < 0. 05) ,as well as SPF New Zealand laboratory rabbits. Conclusion There was obvious difference in physical index properties between the different strains of SPF laboratory rabbits, which is instructive and meaningful to choose stable laboratory animal and improve the advance of animal experiments.

Abstract: Objective To investigate conditions of complexation and dissociation in separating and purifying albumin and specific immunoglobulin from ordinary dog plasma and hyperimmune serum with rivanol method. Method The plasma and rivanol solution were mixed in the volume ratio of one to one and pH value of the mixture was adjusted with 2% Na2CO3 to make the dog plasma and the rivanol full complexation. The gamma globulin was obtained by adding rivanol solution to the supernatant for secondary complexation and dissociating with dissociation dissociation, and the albumin was obtained from the precipitation solution,by dissociating with dissociation dissociation. The concentration and purity of the supernatant and the two dissociation solutions were determined respectively. Result The optimum condition for the albumin complexation was pH 8. 5,at room temperature for 2 h,and 2. 3% rivanol,optimum condition for gamma globulin. complexation was pH 6. 0,at room temperature for 2 h,and 2. 3% rivanol. The optimum condition for the albumin,dissociation was 0. 6% sodium chloride,0. 25% sodium caprylate and pH 1. 37,and optimum condition for the the gamma globulin dissociation was 0. 85% sodium chloride,1. 3% glycine,and pH 6. 8. Conclusion The dog albumin and gamma globulin could be successfully separated and purified with rivanol method.

Abstract: Objective To determine and compare the levels of the certain harmful substances and microbiological levels in the different laboratory animal bedding materials come from the different producing areas and widely used in Beijing region,so as to provide references for constituting local standard of laboratory animal bedding. Method Physical characteristics,and chemical and microbiological pollution levels of different bedding materials were determined according to the methods in force. Result The levels of heavy metals,organophosphorous pesticides, organochlorine pesticides,and other drug residues in corn core and wood shaving bedding materials from different producing areas could meet the requirement of GB 14924. 2-2001. However,microbiological levels in all these bedding materials exceeded the limits to some degree. Conclusion These results can be used as references for constituting local standard of laboratory animal bedding.

Abstract: Objective To establish simple and practicable labelling method of identifying tree shrews with quick and restless behavior to realize scientific management for tree shrews population. Method 40 young tree shrews and 40 adult tree shrews of the F1 generation were selected randomly. Electronic chips with a size of 2. 12 mm × 12 mm and a weight of 0. 09 g,were implanted into subcutaneous part of the neck back through injecting. Observation of implantation site and periodic review were carried out. Result 90% of the chips were detected in young tree shrews and 95% of those were detected in adult tree shrews. Conclusion It is a simple,effective and lasting method to mark tree shrews with electronic chips

Abstract: Objective To investigate antitussive and antiasthmatic actions of Ding-chuan granule for providing theory evidence of its clinic use. Method Histamine and acetylcholine were used to induce tracheospasm in whole body and guinea pig's isolated trachea test,tussive models induced by SO2 in mice,cough model induced by citric acid in guinea pigs were used to obverse the antitussive and antiashmatic action of Ding-chuan granule. Result The result showed that Ding-chuan granule markedly reduced the contraction amplitude of isolated trachea induced by histamine and acetylcholine in the guinea pig and markedly prolonged the latent period of asthmatic attack induced by mixture of histamine-acetylcholine in guinea pig and decreased the frequency of cough induced by citric acid in guinea pigs and SO2 in mice. Conclussion Ding-chuan granule has the antitussive and antiasthmatic actions.

Abstract: Objective To obtain new pentobarbital sodium solution by replacing general method of preparation,and investigate it's anesthetic effect on rats compared with general pentobarbital sodium and chloral hydrate. Method 120 rats were used in this study,which were randomly separated into 3 groups. Each group rats were respectively treated by general pentobarbital sodium,modified pentobarbital sodium and chloral hydrate with intraperitoneal injection,then observed the general state of rats,the acting time and anesthetic time were recorded,which were analyzed by statistics software. Result There were no significantly difference in acting time and anesthetic time between three groups,the variation of acting time and anesthetic time in modified pentobarbital sodium treated group was less than other two groups,and the successfully anesthetic number at first injection was significantly more than other two groups. Conclusion The study showed that the modified pentobarbital sodium could provide the stable anesthetic effect,which function was better than general pentobarbital sodium and chloral hydrate.

Abstract: Animal models play a critical role in our understanding biomedicine sciences. The shortage of ideal animal models always limited our research in certain pathogen infection diseases such as HIV infection. Due to the species difference,some drugs and vaccines work well in certain animals but are not applicable for humans. Transplantation of fetal human thymus tissue,hematopoietic stem cells or immune cells into immune deficient mice will significantly establish human immunity. These humanized mouse models offered ideal models for studies on human immune system and immune-related diseases,and screening medicine and vaccines. The present paper will briefly introduce these humanized mouse models and their application.

Abstract: The skin is the largest body's organ,and also the first natural barrier to protect the body from the invasion of chemicals. The chemicals often cause the allergic contact dermatitis. Traditionally,the guinea pig sensitization test is often used to evaluate the allergenicity of chemicals. However,as this test has some disadvantages such as too long test cycle,and subjective and variable results,and collides with animal welfare and protection,an alternative test has become the inevitable trend of development. This article reviews the alternative tests in the study on skin allergy in the recent years.
