    Establishment of Mice Models of Staphy·lococcus aureus of Dairy Cows Mastiffs

    Objective Infect mouse mammary gland with Staphylococcus aureus(S.aureus)isolated and purified from clinical mastiffs milk samples in order to establish a mouse model of Staphylococcus aureus of dairy COWS mastitis.Methods 8~10 days postpartum far inbred ICR female mice were randomly divided into three groups, including the sterile saline control group,tow—dose—bacteria—attack group(2×1 02 CFU of Staphylococcus aureus) and high-dose—bacterial—attack group(5×106 CFU of Staphylococcus aureus).Staphylococcus aureus or sterile saline were injected into the fourth pair mammary gland through the nipple tube with non—invasive method.24 h after bacteria attack,milk of these rats was collected to do white blood cell count,and blood from eyes to do blood testing and flow cytometry;after the female mice were killed,taking the breast tissue to do homogenate bacterial count,and do biopsy.Results The high—dose group have a significant or very significant difference(P<0.05) with the control group whether in the histological observation and body temperature,or milk white blood cell count,breast homogenate count,some indicators in blood testing and flow cytometry.But compared the low—dose group with the control group,indicators have no significant differences except the blood CD4+/CD8+ratio was significantly different(P<0.05).Conclusion The distant strains of mice models of S.aureus mastitis was successfully established when the concentration of Staphylococcus aureus is 5×1 00 CFU.bacterial attack for 24 h。 and concentration of 2×102 CFU of bacteria is not suitable for the establishment of distant strains of mice models of S.aureus mastitis.
    Effect of Astragalus Injection on Immune,Aging and Plasma Lipid in Aging Mice Induced by D-galactose

    Objective To investigat the aging effects of astragalus injection on the mice induced by D—galactose. Methods The aging model mice were established by subcutaneous injection of D—galactosamine.The mice at the different groups were intraperitoneal injected by the different drugs.After 8 weeks,the mice were killed,and the parameters of malondialdehyde(MDA),superoxide dismutase(SOD),triglyceride(TG)and total cholesterol(TC) were observed,and the liver index,spleen index and thymus index were determined.Results Compared with the model group,the activity of SOD obviously increased(P<0.05)in astragalus group,and the content of MDA,TG and TC obviously increased also.Conclution Astragalus injection can increase the activities of antioxidant enzyme,increase the quality of immune organs,has the nice effects on aging.

    Objective To observe theAnti—inflammatory effects of Anti—virus tablets.Method Experiment of rat agar—induced granuloma,the abdominal capillary permeability of mice.After AA rat models were established,the apricot kernel’s antiarthritic effect on the changes of foot swelling、spleen indes、thymus index、tumor necrosis factor (TNF)and interleukin 1.Result Anti—virus tablets can reduce the abdominal capillary permeability:on agar granuloma caused significantly inhibited in rats;of Freund’s complete adjuvant induced significantly inhibited primary swelling,the thymus index increased spleen index decreased levels of interleukin 1.Conclusion Anti.villJs tablets have anti—inflammatory pharmacological effects.

    0bjeetive To examine the change of synaptic plasticity in VTA—NAc(Vental tegment area—Nucleu accumbens,VTA—NAc)circle in heroin-addicted rats.Methods Sprague-Dawley rats(180~220 g)were randomly divided into control group and heroin—treated group.Heroin were injected subcutaneously twice a day according to the principle of daily increasing dose to establish the model of heroin—dependence.Withdrawal was precipitated by administering the opioid antagonist naloxone to examine whether the heroin—addicted model was established.The control group was given equivalent volume of saline.For induction of LTP,high·frequency stimulations were applied to VTA and NAc,and population spike was recorded in vivo.Results 1)The withdrawal symptom is according with the scoring indications of the established heroin—addicted model of rats.2)The PS amplitude of LTP induced from NAc and VTA in heroin—addicted rats was significantly lower than control group(P<0.05)。Conclusion Heroin can influence synaptic plasticity in VTA—NAc circle in heroin—addicted rats.

     Objective This study was designed to investigate the anti—obesity mechanism of recombinant human ciliary neurotrophic factor(rhCNTF)in the Neonatally Monosodium Glutamate—Treated Obesity model rats and to provide the experiment data for the development of rhCNTF as a new drug.Methods MSG rat model was established in rats by treatment with monosodium glutamate in the neonatal stage.Treatment with rhCNTF at 30~ 300 Ixg·kg~s.c.once a day continuously for 10 or 33 days.The body weight and food intake were monitored every day.After final administration,rats were narcotized with choral hydrate,fat around genital was collected.Size of lipocytes was measured with the microscope.The density of jejunum villi was measured by means of electro— microscopic scan technique.Results The body weight and Lee’S index of MSG—rats significantly higher than that of normal rats,indicating that MSG—OB rats was established successfully.The short—term and long-term administrationof high and middle dose of rhCNTF resulted in a significant and dose—dependent reduction of size of lipocytes. Electromicroscopic scan displayed that the density of jejunum villi in MSG—rats receiving high and middle doses of rhCNTF on a long—term medication basis was markedly diminished,but not on a short-term use of rhCNTF seemed to be related to weakened intestinal absorption of nutritive substances as a result of decreased relative absorption area. Conclusion rhCNTF by SC route of administration has significant and dose--dependent anti--obesity effects in MSG-- rats.The anti-obesity effect of rhCNTF may be related to its anorectic property and its unfavorable effect on intestinal absorption of nutritive substances due to the reduced absorption area.

    Objective To evaluate the microbi0109ical quality of clean rat and mouse.Methods According to the national standard GB/T 14926-200 1,we collected samples from producing and breeding group in some related farms with a principle of simple random sampling,and detected the serum antibody level of main virus. Results The number of Sendai virus(SV)and hanta virus(HV)antibody positive were negative in 3 1 rats;and the number of positive mouse hepatitis virus(MHV)antibodies is 25,and Sendai virus(SV)1 0,Ectromelia(Ect) in 280 mice.Conclusion The virus monitoring and quality control for mouse of clean grade should be strengthened.

    0bjective To study the effect of the extraction temperature on the pharmacodynamic of Radix Bupleuri in decoction.Methods Through The experiments adopted the Lipopolysaccharide rat model of fever and the mouse pinna swelling induced by xylene,Then discussed the pharmacodynamic effect of Radix Bupleuri in decoction respectively at 60。C and I 00。C.Results First,Radix Bupleuriin decoction both have a certain antipyretic role at at 60。C and 100。C,Build the 4 h and 5 h temperature drop compared with model control group has significant difference(P<0.05),The effects are almost same as aspirin,and antipyretic role lasted stability,butthe antipyretic actions of Radix Bupleurisolution is poorer got at 1 00℃.Second.Radix Bupleuriin decoction made by two producing methods both have better antiinflammatory action.mouse pinna swelling group were extremely significant difference compared with the control group(P<0.01),the antiinflammatory action of Radix Bupleuriin decoction B extracted at 60℃are better than decoction A extracted at 100。C.but they are not significant. Conlusion The antipyretic role of Radix Bupleuri in decoction are better at 60。C,it is not significant effect of antiinflammatory action on extraction temperature.

    Objective Observation the reproductive features of wide original tree shrews.Exploring an optimum method that is economical,practical and on a large scale to raising tree shrews baby.Instead of artificial feeding which were reported previously.Method Provided suitable environment and conditions for feeding and management,entirely breast feeding and care by mother tree shrews.More data were collected and statistics from the pregnancy rate,each litter size,survival rate and observation the growth of young tree shrews.Results In 2010,354 tree shrews offspring have been successfully weaned from 1 20 couples and the survival rate of natural reproduction was 90.76 percent.Now breeding population had been established with 1 50 couples of wild original tree shrews and 40 couples of F1 generation,which have laid the foundation for reaching standard requirements.Conclusion This method was one of higher reproductive and survival rate,low cost and simple to operate,it should be promoted to scale produce the tree shrews as laboratory animals.

    Objective To set up a new rat grip and stomach gavage method.Methods Establishing a new rat side grip and stomach gavage method,Result 1 50 rats do intermediate biomarkers study,each rat stomach gavage one time every day,and lasts for six months.Till the end all rats live well,and finish the experiment favorable.     0haective To observe which concentration of pentobarbital sodium induces lower mortality when obtaining the same anesthetic effect in rabbits.Methods New Zealand rabbits were divided into 1%pentobarbital sodium group and 3%pentobarbital sodium group.The anesthetic effects and mortality were observed.Results There is no significant difference of anesthetic effects between the two groups.but 1%pentobarbital sodium group achieved a lower mortality(P<0.05).Conclusions We should replace 3%pentobarbital sodium with 1% pentobarbital sodium when anesthetizing rabbits because 1%pentobarbital sodium induces lower mortality and equal anesthetic effect compared with 3%pentobarbital sodium.
