Objective To establish embryo bank for Smad2 gene knock-out mice. Methods The OPS vitrification was used to cryopreservation of morula and blastocyst embryos of the Smad2 gene knock-out mice.We compare the results of superovulatory treatment,recovery rate and survival rate in different groups.Results The average numbers of embryo production of Smad2~(+/-) and Smad2~(+/+) were 14.13 and 24.60 respectively after superovulatory treatment.The recovery rates of the two group embryos were 90.16% and 91.67% respectively.The rates of development were 73.08% and 77.05% in vitro respectively after thawing.The results indicated that the numbers of superovulatory of Smad2~(+/-) were lower than the ones of Smad2~(+/+),the rates of recovery and the rate of survival were no different between the two group mouse after thawing.We have cryopreserved 1256 embryos of the Smad2 gene knock-out mice by OPS vitrification.Conclusion We establish the embryo bank for the Smad2 gene knock-out mice.

Objective Evaluating the effect on mice's non-specificity immune function of Bifidobacteria BB1.Methods The mouse was injected with cyclophosphamide in abdominal cavity.After setting up the immunodeficiency mice model(IDMM),macrophage phagocytosis test was done.Results Intragastricing medium and high dose of Lactobacillus BB1 could improve the phagocytosis function of macrophage. Conclusions The non-specificity immune function of IDMM was improved by intragastricing Bifidobacteria BB1.

Objective To observe the correlation between body weight and main organ weight of curly rats.Methods To dissect and get the main organs of curly rats and SD rats,weighed with BS210S electric scales,and assayed data with t test.Results Obtaining data on correlation between body weight and main organ weight.The weight of the spleens and thymuses in female curly rats was much lower than that of female SD rats(P<0.05,P<0.01) at 90th day.For males,the weight of livers at 28th day and of spleens at 60th day was much higher in SD rats than that of curly rats(P<0.05).For males,the coefficients of spleen at 28th and 90th days and of thymus at 28th day in cicinnus rats were lower than that of SD rats(P<0.05).Conclusion The mutant gene has a little effect on the organ weight and coefficient of rat.

Objective To compare the anesthetic effect of pentobarbitol sodium made by different factory on rats and rabbits.Methods Pentobarbitol sodium from chemical reagent factory of Foshan,Shanghai Xingzhi chemical reagent factory, Shanghai chemical reagent factory of Chinese medicine corporation(abbreviated as P_(fo),P_(shang),P_(zhong) individually) were made into solution,which were administrated to rats by celiac injection at the dose of 45 mg/kg and rabbits by vein injection at the dose of 30 mg/kg.Each kind of drug was administrated to 20 rats and 20 rabbits with half male and half female individually.The acting time,acting duration and the general state of anesthesia were recorded.Results The acting time(minute) of P_(fo),P_(shang),P_(zhong) on rats was 3.8,6.6 and 4.5 individually,acting duration was 152,107 and 126,the general state of anesthesia was P_(fo) and P_(zhong) were more ideal than P_(shang),The acting time(minute) of P_(fo), P_(shang),P_(zhong) on rabbits was 0,0~1and 0 individually,acting duration was 148,89 and 138,the general state of anesthesia was P_(fo) and P_(zhong) were more ideal than P_(shang).Conclusion Anesthetic effect of pentobarbitol sodium from different factory on rats and rabbits is P_(fo) were more ideal than P_(zhong),P_(zhong) were more ideal than P_(shang).

Objective Investigating bacterium in wild and domesticated Mongolian gerbil to establish Beijing local standard for control of bacterium in Mongolian gerbil.Method Wild Mongolian gerbil from Yinchuan city and Huhehot city and domesticated gerbil breeding in barrier system were euthanasied and anatomized.And then the bacterial flora of the cecum,trachea and larynx were isolated and cultivated by blood agar.After 18~24 h 37℃ cultivature,the bacterial colonies on the agar were classified and separated by singly colony.These singly colonies were identified by VITEK-32-auto bacterium analysis system.Result The wild gerbil from Yinchuan city and Hohehot city have 12 and 8 species,respectively.But the domesticated gerbil only has 7 species.Conclusion The bacterium isolated from wild Mongolian gerbil from Yinchuan city and Hohehot city have great differences in quantity,species and infection rate compared with laboratory gerbil from barrier system.These factors should be considered in establishing Beijing local standard for control of bacterium in Mongolian gerbil.

Objective To isolate and identify Corynebacterium kutscheri from rat,and to provide some useful reference for microbiological monitoring in laboratory animal.Method Pathogenic changes of rat was examined.Bacteria cultivation,biological trial,serum agglutination test and animal experiment were carried out.Results Histological examination of C.kutscheri infected rat demonstrated the organism within lung and liver tissues.Isolation and identification of C.kutscheri were performed from laboratory rat which was killed by C.kutscheri infection during a long-term toxicologic study in Beijing.Conclusion The long-term toxicologic study decreased laboratory rat resistance in stressed,which evoked inapparent infection and cause C.kutscheri infected rat dead.We should recognize the importance of C.kutscheri as a potential pathogen in such state.

Objective To identify the antigenic and evolution to study the mutation of CPV and lay the foundation for the identification and treatment of this disease.Methods A pair of primers were designed and synthesized based on VP2 gene of Canine parvovirus(CPV).The genomic DNA samples of CPV strain isolated from culture solution ware extracted and used as templates for PCR to clone the VP2 gene.The sequences of the DNA fragment were aligned and analyzed by DNAstar.Results 15 were CPV-VP-2,including 10 of CPV-2a and 5 of CPV-2b;1 was FPV-VP-2.Conclusions Compared with standard sequence,homogeneities were above 98%,they had not formed obvious branch.

Objective Besides the embryonic quality,we hope to explore the better technique and method of mouse embryonic transferred,to increase the yield of embryonic transfer.Methods Embryos were collected from both the natural and superovulated estrous mouse of pregnant 3.5 days.Using microscope to collect the embryos that were coincidence in the embryonic development and age,and transferred them to mouse uterus of natural estrous 2.5 days.Result As compared with,superovulation could increase embryonic quantity,however there is no better than natural estrous in embryonic quality.In twelve receptors of natural estrous,three receptors were pregnant and produced twelve mice.Conclusion The factors of affecting embryonic transfer are the donor and receptor,multiple ovulation,embryonic quality score,the number of embryo transferred,the technique and method of embryo transferred.
