    Radiation-induced Mice Leucopenia Dependent on Strain and Sex

    Objective To develop a mice leucopenia model induced by()~(60)Co gamma-ray.Methods ICR and C57BL/6J mice were subjected to single systemic()~(60)Co gamma-ray radiation a dose of 50 Gy,and peripheral white blood cell(WBC) count was taken for both radiated and non-radiated mice.Results Significantly elevated WBC count after radiation was observed in both strains.The WBC count in normal ICR mice was very significantly lower than that in C57BL/6J mice,and the WBC count in male C57BL/6J mice three days post radiation was significantly higher than the same count in female C57BL/6J mice.Conclusion()~(60)Co gamma-ray radiation can result in leucopenia and its severity is greatly dependent on mice strain and sex.

    Objective To develop atherosclerotic model of apolipoprotein-E(ApoE) knockout mice.Methods Six male 8-week-old ApoE-knockout mice were fed with atherogenic diet for 8 weeks,and 6 male 8-week-old C57BL/6J mice were used as controls.The mice were sacrificed,and blood was drawn for determination of ssubjected to paraffin sections stained with hematoxylin and eosine to examine atherosclerotic lesions in aortic root.Results Serum TG,TC,and LDL-C levels in ApoE-knockout mice were significantly higher than those in C57BL/6J mice,whereas the reverse was true for serum HDL-C level.Lipid plaques of atherosclerosis were seen in the sections of aortic root of the ApoE-knockout mice.Conclusion Atherosclerotic plaques have already generated in 16-week-old ApoE-knockout mice,which may be ideal model for research on cardiovascular diseases.

    Objective To investigate pathological lesions of liver induced by cyclophosphamide in BALB/c mice.Methods Cyclophosphamide was intraperitoneally administered to BALB/c mice in a dose 15 mg/kg once daily for 8 days.Eight days later, the mice were sacrificed and their livers were subjected to paraffin sections stained with hematoxylin and eosine to examine hepatic lesions under light microscopy.Results Some pathological lesions such as focal necrosis and interstitial inflammatory cell infiltration and so on were found in the liver tissue.Conclusion Cyclophosphamide can cause the considerable damage to the liver of BALB/c mice.

    Objective To evaluate the anticancer activity of some Chinese herbs combined with the trace element selenium.Method HepA or Erlich cancer cells injected to BALB/c mice to make solid or ascites cancer.To investigate the tumor growth curve,animal survival duration,the tumor inhibitory rate.Results (1)Tumor growth of the treated group was remarkably inhibited The inhibitory rate of HepA and Ehrlich ascites tumor was 66%~78%(P<0.01) and 72%~81%(P<0.001) respectively.(2) The survival duration of treated groups was longer than that of control group.The life span was also remarkably prolongated,being about 1.7 times than that of control group.Conclusion Trace element Se combined with some Chinese herbs has remarkabl anticancer activity.

    Objective To observe and compare effects of three different illumination durations on growth,development and reproduction of BALB/c nude mice.Methods Newborn BALB/c nude mice were fed under three different illumination durations,and their growth and development were observed.Mating and delivery intervals,delivery frequency,and number born were observed in female BALB/c nude mice,and rate of offspring survival was recorded.Some blood biochemical indexes were determined by LX20 semi-automatic analyzer.Results The differences in growth and development of newborn BALB/c nude mice were not significant among three different illumination durations(P>0.05).The average rate of offspring survival in the group of middle illumination duration was significantly higher than that in the group of long illumination duration(P<0.05).The differences in ALT,TB and SUA between the group of middle illumination duration and the group of long illumination duration were significant statistically(P<0.05). Conclusion The middle and short illumination durations are more suitable to growth,development and reproduction of BALB/c nude mice than long illumination duration.
    Infusion of Alloreactive Lymphocytes Induced in vitro Can Prolong The Survival of Skin Allograft in Rats

    Objective To investigate whether infusion of alloreactive lymphocytes induced in vitro can prolong the survival of skin allograft in rats,so as to find out a new method to induce transplantation tolerance.Method Lymphocytes from Lewis rat were used as stimulating cells,lymphocytes from BN rat were stimulated by these stimulating cells and expanded in vitro;the proliferation of BN lymphocytes were detected by CFSE method.The proliferated BN lymphocytes were used as idiotype vaccine to vaccinate BN rats,then skin allograft from Lewis rats were transplanted to BN rats.Results Alloreactive lymphocytes proliferated vigorously in vitro;infusion of alloreactive lymphocytes induced in vitro can prolong the survival of skin allograft significantly in rats.Conclusion Infusion of alloreactive lymphocytes induced invitro may be a new pathway to induce donor specific transplantation tolerance.

    Objective To develop ELISA for detection of antibodies against monkey B virus using BVgD-Peptide as antigen.Methods Antigenicity of BVgD-Peptide antigenic determinant was analyzed by the Western-blot method.The ELISA method was established by optimization of reaction condition and reagent selection.The parallel detection of monkey sera was performed by using three different ELISA kits using complete BV,HSV-1 and BVgD-Peptide as antigens.The detection results obtained by ELISA kit developed in the study were compared with the results obtained by ELISA kit made in USA.Results Compared with ELISA using as complete BV and HSV-1 antigens,ELISA using BVgD-Peptide as antigen was less sensitive but more specific.85% of the detection results obtained by ELISA kit developed in the study were coincident with those obtained form Laboratory of America.Conclusion ELISA for detection of antibodies against monkey B virus using BVgD-Peptide as antigen has been developed,and this ELISA is more specific than ELSA using complete BV and HSV-1 as antigens.

    Objective To develop the model of repair of the whole facial nerve defect bridging by chemically acellularized facial nerve allograft so as to explore the feasibility of the whole nerve defect repair.Methods The facial nerve from the rabbit was chemically treated by Triton X-100 and sodium deoxycholate to get rid of Shwann's cells and medullary sheath.A 2 cm left facial nerve defect was made in the rabbit and was bridged by chemically acellularized facial nerve allograft under surgical microscope.Results After 6 months,more satisfactory nerve regeneration were found in the defect,with no infection.Conclusion The model has good stability and repeatability and it is possible to repair the whole facial defect by chemically acellularized facial nerve allograft.

    Objective In order to establish an improved immunohistochemical staining method to detect avian influenza virus,how to eliminate the nonspecific reaction was explored in the report.Methods The antigen retrieval, incubating condition of primary antibody,and blocking reaction of endogenous biotin were optimized and the results of improved method and the unimproved one were compared by SABC detection system.Results Satisfactory staining effect was obtained by antigen retrieval with microwave oven,primary antibody being incubated at 37℃ for 1.5~2.0 hours,and endogenous biotin being blocked by 20% egg white solution for 20 minutes.Compared with common method,the improved one was able to eliminate the background interference effectively,and clear positive expression could be observed.Conclusion The improved method described was specificity enough and could be applied in preclinical medicine research and clinical detection for avian influenza virus.
