    The Measurement of Weight of Main Organs and Organ Coefficient in ROSA Genetically Engineered Mice

    Objective To measure the organ weight and organ coefficient of B_6;129-Gt(ROSA)26Sor mice.Methods 5~6 weeks male and 6~7 weeks female mice(15 in each group) were used in this experiment.The weight of various organs was measured using a Sartorius BP121S electronic balance.Results Between female and male mice,the body weight,the heart,kidney weight showed very significant difference(P<0.01);the liver,adrenal weight showed significant difference(P<0.05);the coefficient of liver,lung,kidney,brain,and adrenal showed very significant difference(P<0.01),but no significant difference was seen in other organ coefficient.Conclusion The weight of some main organs and organ coefficient in different gender ROSA mice are significantly different;the above data could be useful in the biomedical researches.

    The skeleton length of Wistar-Imamachi rat population 5~16 week ages was compared.The results show the skeleton is correlation with week ages including the total length of cranium,the length in base,the cranium height,the upper cheek tooth width,upper tooth arrange length,left upper arm bone length,the 3rd cervical vertebra length and the 1st lumbar vertebrae length,except upper cheek tooth width and upper tooth arrange length of male rats.

    Objective To observe the antilipemic effects of recombinant human ciliary neurotrophic factor(rhCNTF) in the Neonatally Monosodium Glutamate-Treated Obesity model rats.Methods MSG obese rat model was established in rats by treatment with monosodium glutamate in the neonatal stage.The increases in the Lee's index(body weight1/3/body length) and in retroperitoneal fat as well as decreases in spontaneous motor activity were recorded.Food consumption and associated hypothalamic low body length obesity(monosodium glutamate-treated obesity;MSG-OB) were observed as these rats grew.Treatment with rhCNTF at 30~300 μg·kg-1s.c.once a day was given continuously for 33 days.The body weight and food intake were monitored every day.After final administration,rats were narcotized with choral hydrate,then the body length and weight were measured to calculate Lee's index.Blood was sampled through abdominal aorta to meature the serum levels of glucose and lipids.Fat around kidney and genitals and in abdominal cavity was collected for calculation of fat index.Results The body weight and Lee's index of MSG-rats significantly higher than that of normal rats,indicating that MSG-OB rats was established successfully.The long-term administration of high and middle dose of rhCNTF resulted in a significant and dose-dependent reduction of Lee's index,fat index,serum triglyceride(TG) and tatal cholesterol(TC),and to profoundly increased high density lipoprotein-cholesterol(HDL-C) in comparison with obese saline-treated control.It also reduced a significantly the level of serum glucose hypoglycemic effect.Conclusion rhCNTF has significant and dose-dependen antilipemic effects in MSG-rats.Along with the antilipemic effect,rhCNTF also produces a significant hypoglycemic effect.

    Objective To study the influence of emotional stress to the ultrastructures of temporomandibular Disorder in rats,and to establish the TMD animal model.Methods 16 male Wistar rats were dividing into 2 groups randomly,one was of which experimental group,and the other was control group.The experimental group was subjected to make chronic unpredictable stress animal model.After 6 weeks,all of rats were killed and their condyles and articular discs were dissected and taken out completely,observing by scanning electron microscope.Result In experimental group,the ethology of rats changed significantly and under scanning electron microscope,the condyles in experimental group were found to have the surface gelatum cracked,disordered and broken collagen fibrils,and exposed deep layer collagen fibrils.The articular discs' surface gelatum were still integrated,but the gyri dispose were irregular.In control group,there were no such changes in condyles and articular discs.Conclusion The chronic unpredictable stress can induce the ultrastructural injuries of temporomandinbular joint in rats,which is consistent with the pathological changes of TMD,so we presumed it is feasible to induce the animal model of TMD.

    Objective To establish a Rat model of Type 2 diabetes induced by streptozotocin supplemented with 5% sucrose and high-fat diet,and to observe the therapeutic effects of metformin.Methods SD rats were injected peritoneally with streptozotocin(30 mg/kg) and 5% sucrose and high-fat diet were given to the rats for 68 days,and were engaged in glucose tolerance test.Blood samples were taken to detect glucose,blood lipid,sarum insulin and anti-oxidation level.Results Compared with the control group,the glucose,tolerance test after 2h glucose,glycated hemoglobin,serum insulin and insulin resistance index in high-fat feeds group were significantly higher(P<0.01).The insulin sensitivity index was significantly lower(P<0.01).TG,TC and free fatty acids(FFA) were significantly higher(P<0.05).The SOD activity decreased significantly(P<0.05).MDA was significantly increased(P<0.01).TC,FFA,glucose,glucose tolerance Anomaly,glycated hemoglobin,serum insulin and insulin resistance index,MDA In metformin-treated group were significantly lower(P<0.05).The insulin sensitivity index and SOD increased significantly(P<0.05).Conclusion 5% sugar water can improve the symptoms of anorexia,and combined with the low-dose streptozotocin and high-fat diet haS successfully induced a type 2 diabetes rat model with hyperlipidemia.Metformin has a hypoglycemic,lipid-lowering,anti-oxidation effects,and can improve insulin resistance,So it can be used as a positive therapeutic drugs to treat type 2 diabetes rat model with hyperlipidemia.

    Objective To identify the cause of mass disease in rabbit.Methods Pathological autopy,bacteria isolation and culture,biochemical identification,serum agglutination test,PCR detecting and drug sensitivity test of the isolated strain were conducted on diseased rabbit.Results Combined the results obtained from pathological autopy,bacteria isolation and culture,observation of culture characteristics and morphologic structure,biochemical testing,serum agglutination test and PCR detecting,the disease was diagnosed as Staphylocosis by Staphlococcus aureus.The results of drug sensitivity test showed that the isolated strain was sensitive to Penicillin,ampicillin and vancomycin.However,its resistence to ceftazidime and nitrofurantoin were produced.Conclusion The results of this study provided a basis for the identification of pathogen Staphlococcus aureus and diagnosis of Staphylocosis.

    Objective To discuss the anesthetic effect and its influencing factors of pentobarbitol sodium on dogs.Methods 540 dogs were anesthetized by 2.5%and 3% pentobarbital sodium intravenously in different week,the anesthetic effect and the acting time were recorded.Results Anesthetic effect were divided into good,moderate,bad,and death according to the reaction of dogs,which occurred in 418(77.4%),55(10>2%),47(8.7%),and 20(3.7%) respectively.Acting duration was less than 30 mins in 91.5% of dogs.Conclusion Anesthetic effect in laboratory dogs by using 2.5% pentobarbital sodium intravenously was safe and effective,and is more suitable when applying in large amount of dogs,Overdose of anesthesia could be reduced through standardize administration.
