Objective To investigate the effects of intratumoral dendritic cells injection combined with radiotherapy on cytokines in the mice with renal carcinoma.Methods[WTBZ] BALB/c mice received renal cancer cell(RENCA) vaccination(subcutaneous injection of 2.5×10~6 RENCA cells),and then radiotherapy(7 Gy/time) and intratumoral injection of unpulsed dendritic cells at 1×10~6 cells/mouse/time were carried out.The mice were sacrificed on twenty-eighth day.Growth rate,weight and size of tumor mass were measured,and IL-2,IFN-γ,IL-4,IL-10,and TNF-α levels in splenocytes were determined by ELISA assays.Results Growth of tumor mass in the group of intratumoral dendritic cells injection combined with radiotherapy was significantly slower than that in other groups,and IL-2,IFN-γ and IL-4 levels in splenocytes in the group of intratumoral dendritic cells injection combined with radiotherapy was significantly higher than those in other groups.Conclusion Intratumoral dendritic cells injection combined with radiotherapy can inhibit growth tumor in the mice.

Objective To determine normal range of the blood routine values and blood biochemical parameters of the genetically engineered mice(B6;129-Gt(ROSA) 26Sor).Methods Blood routine examination was performed by the automatic hematology analyzer and blood biochemical parameters were determined by the automatic biochemical analyzer in male ROSA mice aged from 5 to 6 weeks and female ROSA mice aged from 6 to 7 weeks.Results The differences in granulocyte count,percentage of granulocyte,and mean corpuscular hemoglobin were significant(P<0.05) and the differences in redcell count,hematocrit,haemoglobin and platelet distribution width were very significant(P<0.01) between female and male ROSA mice.The difference in blood creatinine level was significant(P<0.05) and the differences in albumin and total cholesterol levels were very significant(P<0.01) between female and male ROSA mice.Conclusion There are definite differences in physiological and biochemical parameters between female and male ROSA mice.

Objective To observe the relationship between selecting and breeding of BALB/c mice,ascitic yield and titers of the antibodies during the preparation of monoclonal antibodies against Fl antigen of Y.pestis so as to provide reference for preparing vast monoclonal antibodies against Fl antigen of Y.pestis.Methods Ascites was induced in BALB/c mice of different age,sex and breeding conditions,and the titers of monoclonal antibody against Fl antigen of Y.pestis in ascites were detected by indirect ELISA.Results: The ascitic yield and titers of the antibodies in the male and well-nourished mice aged from 12 to16 weeks were higher than those in the control group.Conclusion: Selection of male BALB/c mice of appropriate age,intensifying nutrition and better care can produce an increase in ascitic yield and the titers of the antibody.

Objective To investigate antifatigue effect of Huili Capsule(回力胶囊 Capsule for Restoring Physical Strength) by swimming test in the mice.Methods Huili Capsule was orally administered to the mice at high,middle and low dose for 30 successive days.Loaded swimming was carried out in the mice.Loaded swimming duration was recorded,and glycogen level in liver,urea nitrogen level in serum,and blood lactic acid level were determined.Results As compared with the control group,loaded swimming duration was prolonged in the groups of high and middle dose.Blood lactic acid level after loaded swimming in the groups of high and middle dose was lower than that in the control group,urea nitrogen level in serum in the group of high dose was lower than that in the control group,and glycogen level in liver in group of high dose was higher than that in the control group.Conclusion Huili Capsule can exert antifatigue effect.

Objective To evaluate the security of bone nutrient as a health food.Methods The acute oral toxicity test and mutagenesis test for bone nutrient in mice and 30 days feeding test for bone nutrient in rats were carried out according to 《Technical Standards of Test and Assessment for Health Food》promulgated by Health Ministry of the People's Republic of China.Results Median lethal dose of orally administering bone nutrient in mice was more than(21.5 g/kg),which demonstrates that bone nutrient is nontoxic.Mutagenic test showed negative result,and 30 days feeding test in rats showed that the rats has gained weight and there were no significant differences in food exploitation rate,organ/weight ratio,hematology and serum biochemical indexes in the rats of each dosoge group as compared to the control group(P>0.05).The pathological changes related to the experiment were not found in the pathological examinations of main organs.Conclusion Bone nutrient is a nontoxic and non-mutagenic heath food.

Objective To develop heterotopic cardiac transplantation model by two methods in rats and compare their difference.Methods 144 SD rats were used in this series. Two methods were used to develop heterotopic cardiac transplantation model in rats.Results The heterotopic cardiac transplantation was done by suturing in 42 cases,and 33 cases were successful with a successful rate of 78.57%.The transplantation was done in 30 cases by cuffing,and 27 cases were successful with a successful rate of 90%.All the transplanted hearts could survive over 30 days.Conclusion The two methods of developing heterotopic cardiac transplantation model are very useful in the study of organ transplantation.But they have different advantage,so it is necessary to select an appropriate animal model according to the idiographic experiment.

Objective To explore the effectiveness of permanently breeding SPF rats in individually ventilated cage(IVC) system withing relative clean environment.Methods IVC barrier system located in class 100,000 clean laboratory was established.SPF SD rats aged 4 weeks were divided into two groups,and one group was fed in IVC berrier system and other group in traditional barrier system.Their hematological,general biological and part hemetaboly indexes were determined and histological examinations were carried out in each group.Results The growth and development indexes in IVC barrier system were little better than those in traditional protective system,and differences in growth and development indexes during fast growthing periods were significant between the two groups.The differences in hematological, biological and part hemetaboly indexes,and histological examinations were not significant between the two groups after breeding for seven months.Conclusion There were no significant differences in above indexes between the IVC and traditional barrier system and the experiment results from IVC barrier system should be reliable.

Objective To analyze single nucleotide polymorphism(SNPs) of peroxisome proliferative activated receptor-γ coactivator 1 gene exon 8 in three strains of Chinese native minipigs(Bama minipigs,Wuzhishan minipigs,and Nongda minipigs) to provide basic data for their use in the researches of diabetes mellitus and metabolic diseases.Methods Genomic DNA was extracted from the blood samples of the minipigs,and served as templates for target gene segment amplified and selected by polymerase chain reaction (PCR).The PCR products were sequenced and analyzed for documenting single-nucleotide polymorphisms(SNPs).Results Five SNPs were found in the target region,Ala354Val(450C→T),Gly363Gly(478C→T),Arg369Arg(494C→A),Leu570Leu(913G→A),Ser551Ser(1039A→G),but only the variation of 450C→T to cause an encoding change Ala354Val.Conclusion There are variations on the SNPs in PGC-1 gene exon 8 among the different strains and individuals of native minipigs.

Objective To improve the method of developing hyperlipidemia in rats.Methods The batch formula and preparation of lipid emulsion were improved,and the modified lipid emulsion was orally administered to the rats to induce hyperlipidemia.Afterwards,the total cholesterol(TC) and triglyceride(TG) levels in sera of two groups were determined by enzyme colorimetry.Results The lower mortality was found in the model group,and the differences in TC and TG levels were statistically significant between the model group and the normal control group.Conclusion The modified method of developing hyperlipidemia model has higher success rate,so it is workable and convenient.
