Abstract: Objective To establish streptozotocin( STZ) -induced type 1 diabetes mellitus model in Bama minipigs.Method 10 minipigs were divided into 3 groups,which were treated with normal saline( 2) ,120 mg /kg STZ( 2) and 150 mg /kg STZ( 6) respectively. The blood glucose was detected at 1 h interval during the first 24 h after treatment,and at one day interval during the following 30 days. The animals were sacrificed for pathological detection at the end of 30 day test. Result Transitorily high blood glucose was induced with 120 mg /kg STZ which the highest level reached 21. 3 mmol /L,and gradually decreased to normal after 17 days. Stable high blood glucose was induced with 150 mg /kg STZ which varied among 6. 1—35 mmol /L in 3 /6 animals,and 2 /3 animals induced transitorily high blood glucose like 120 mg /kg STZ treated animals,and 1 /6 animal showed normal blood glucose. Conclusion Mild diabetes mellitus was induced with 120 mg /kg STZ. Sever diabetes mellitus was induced with 150 mg /kg STZ with high variation among animals.

Abstract: Objective To investigate the mechanisms of treating OME with Ambroxol Hydrochloride tablets and Bromhexine Hydrochloride injection. Method The animal model of OME was made and was treated with Ambroxol Hydrochloride tablets ( AH) and Bromhexine Hydrochloride injection ( BH) . At the date of 7 and 14,the middle ear rinses and serum were collected. The levels of IFN-γ,IL-4,LTC-4,PGD2 and PAF were detected.Result Compared with normal saline group,the levels of IFN-γ,IL-4,LTC-4,PGD2 and PAF of the experimental group that treated with AH and BH were significantly decreased ( P < 0. 05) at the date of 7,but the differences of other parameters between the control group and the experimental group that treated with AH and BH were not significant. At the date of 14,compared with the control group,the differences of the levels of IFN-γ,IL-4,LTC-4,PGD2 and PAF of the experimental group that treated with AH and BH were not significant ( P >0. 05) . Conclusion The mechanism of action of Ambroxol Hydrochloride tablets and Bromhexine Hydrochloride injection is related to inhibiting type I allergy.

Abstract: Analysis and fitting of the growth curve was important when the regular pattern of growth and development was studied. The growth curve of 960 rats and mice of six species was analyzed with the help of the Bertanlanffy model. The results showed that Bertanlanffy model was more suitable for simulating animal growth( R2 > 0. 948) .The relative growth rate and the absolute growth rate of mice were in decline,and the relative growth rate of rats showed a downward trend,while the absolute growth rate reached a peak after the decline. Bertanlanffy model has been widely used in research for prediction of growth rate and genetic analysis of growth process,and can also provide the basic data for the standardization of laboratory animals.

Abstract: Objective To explore a simple establishment method of animal model of human meningioma. Method Under the strict experimental conditions,the fragments of human meningioma tissue were implanted subcutaneously into nude mice. The tumor growth curve was drawn and pathological examination was done to verify the pathological features of the transplanted tumor. Result The success rate of primary transplant was 78%,and the success rate of the expend transplant between nude mice was 100%. The morphology of all transplanted tumor was consistent with original human meningioma. Conclusion Implanting the fragments of human meningioma tissue subcutaneously into nude mice is an ideal method to produce meningioma animal model.

Abstract: Objective To observe the effect of Vitamin A on the islet fibrosis in type 2 diabetes mellitus ( T2DM) animal model of GK ( Goto-Kakizaki) rat. Method After administration of Vitamin A for 2 weeks; and the changes of islet fibrotic area were recorded. 5 μm serial paraffin sections of 4 months old GK rat pancreas were obtained and stained by H&E and Masson staining. Result Masson staining showed that the area of islet fibrosis staining in GK rats were significantly decreased after administration of Vitamin A compared with control group ( 44. 46 ± 16. 98 vs. 60. 22 ± 31. 47 μm2 ) . Conclusion Our study indicated that Vitamin A could significantly decrease islet fibrosis in GK rat.

Abstract: Objective To provide an efficient genetic diagnostics method for Labrador Retrievers,6 SNPs of 5 canine hip dysplasia ( CHD) related genes in genomic DNA of Labrador Retrievers were detected. Method 11 normal and 6 typical CHD Labrador Retrievers were selected by imaging diagnosis. The vein blood was sampled and the genomic DNA was isolated from white blood cells. The PCR/sequencing method was used to analyze the variants of 6 SNPs in 5 canine hip dysplasia ( CHD) related genes ( LAMA2 ( CFA1: 70997779) ,LRR1 ( CFA8: 29247021 ) ,SCR ( CFA24: 28944481 ) ,COL6A3 ( CFA25: 51031100, 51040259 ) , and FN1 ( CFA37: 25095511) ) . Furth more,the association between the SNP variants and CHD was analyzed by Chi-square Test.Result The variants of LAMA2 ( CFA1: 70997779 ) ,LRR1 ( CFA8: 29247021 ) ,SCR ( CFA24: 28944481 ) ,COL6A3 ( CFA25: 51031100) ,and FN1 ( CFA37: 25095511) SNP loci were not significantly associated to CHD ( P > 0. 05) . A/A variants in COL6A3( CFA25: 51040259) SNP locus was significantly associated to CHD ( P = 0. 007) . Conclusion A/A variants in COL6A3 ( CFA25: 51040259) SNP locus might be used as a practical genetic detection index for CHD diagnostics in Labrador Retrievers. However,further confirmation in large-sample is required.

Abstract: Objective To study the effect of low frequency electrical stimulation on hormone levels in organs of Chinese tree shrews after death. Method Chinese tree shrews were given low frequency electrical stimulation,0 h,3 h,6 h,12 h,18 h,24 h and 36 h and 72 h after death,thyroid,liver,spleen were taken,respectively,and levels of endothelin ( ET) ,atrial natriuretic factor ( ANF) ,thromboxane ( TX) were determined by RIA method; c-fos expression of midbrain ventral tegmental area ( VTA) of Chinese tree shrews at 0 h after the death was measured. Result After electrical stimulation,ET,ANF,TX level in body organs and c-fos expression in VTA of Chinese tree shrews were significantly increased than the control group, the contents were decreased with the extension of time after death. Conclusion Low frequency electrical stimulation can induce the synthesis and release of the hormone in organs and c-fos expression of brain tissue in Chinese tree shrews.

Abstract: Objective To determine and compare the basic parameters of blood,heart rate,carotid artery pressure,ventricular pressure and organ parameters of SPF( specific pathogen free) FMMU albino guinea-pig. Method The heart blood was collected and blood physiological and biochemical index were measured in a total of 30 SPF FMMU albino guinea-pigs of about 250—300 g weight,and blood gas was determined. Blood pressure in carotid artery was measured by arterial puncture. Ventricular pressure and heart rate were measured on the respiratory support condition. Result The differences of RBC、HGB、HCT、AST、AMY、CHOI、TCO2、HCO -3 、THbc、Na + in male and female SPF FMMU albino guinea-pig were significant( P < 0. 05 or P < 0. 01) . The differences of carotid diastolic blood pressure and mean arterial pressure,left ventricular diastolic pressure,right ventricular systolic pressure and mean pressure were also significant ( P < 0. 05 or P < 0. 01) . Finally,differences in both body weight and brain weight,and organ coefficient of pituitary,adrenal gland and left /right kidney were significantly different ( P < 0. 05 or P < 0. 01) . Conclusion There were differences of blood physiology and biochemistry,blood gas,carotid artery pressure,ventricular pressure and organ parameter in different gender of the SPF FMMU albino guinea-pig.

Abstract: Objective To discuss the impact of the rainy day,fog and PM 2. 5 in SPF chicken coop purification index,SPF chickens and its barrier environment maintenance management. Method Detemine air velocity and purification multiple indicators when using HEPA on the early and late,in the sunny day,rainy weather and fog weather conditions. Result We found it can produce HEPA“wet block”phenomenon in the air relative humidity ≥80%,at the same time,purification of air-conditioning box pressure can reach more than 1 000 Pa. it is different to haze and rain,HEPA damaged is irreversible of haze,but small damage of rain,after a rainy day HEPA ventilation can be part of the recovery. Conclusion People recognize the impact of the SPF coop barrier facilities and the influence of SPF. The “wet block”not directly to the detriment of SPF chickens,but caused a great negative impact on barrier facility SPF sheds management,alert us to the weather in the chicken house construction and management,especially the factors of haze.

Abstract: Objective To investigate operating performance and improving growth performance of surgical taildocking and rubber ring tail-docking of piglets. Method Two or four litters of piglets were randomly divided into two experimental groups and one normal control group. At approximately 1—3 days of age, piglets from experimental groups were tail docked using surgical tail-docking or rubber ring tail-docking respectively. All piglets were observed for tissue damage and pre-weaning daily gain. Result Compared with tail-docked piglets using surgical tail-docking, piglets using rubber ring tail-docking have been found to have fewer tail injuries.Furthermore,the growth performance of tail-docked piglets were increased significantly ( P < 0. 05) compared with normal control. Conclusion Tail docking of piglets using rubber ring was superior to surgical tail-docking in physical damage and operating performance. The growth performance of tail-docked piglets was remarkably improved.

Abstract: Autoclave sterilizer is commonly used in animal laboratory and extremely dangerous equipment. It takes most of the items in the animal room and laboratory sterilization work. When the pressure vessel accidents have occurred in recent years,the safety of pressure vessels should be paid great attention. Staff should have special equipment operating qualifications,while cultivate good quality of the profession. Equipment operation should pay attention to standardized operating procedure, and complete routine equipment maintenance carefully. The equipment should be periodically checked within the design working life. According to the regulation,pressure gauge of the autoclave sterilizer and the steam generator should be inspected every six months,safety valve of the autoclave sterilizer and the steam generator should be inspected every year.

Abstract: Objective To study the therapeutic effects of rats with intestinal obstruction induced by intraventricular injection of STZ in order to improve postoperative survival rate. Method STZ were given intraventricular injection to 60 rats and one week after injection 10 rats with intestinal obstruction were selected for evaluation of therapeutic effects of glycerine enema and radix et rhizoma Rhei water,at the same time the rates were supplied with soft food.Then the therapeutic effect was evaluated by the following parameters: food intake,fur color,mental health,emotional reactions,body temperature,stool and urine. Result During the therapeutic period 2 rats survived and 8 died ( autopsy results revealed the intermediate obstruction,usually with intestinal flatulence,perforation, necrosis. Conclusion The combined therapy with Chinese and Western medicine is simple and relatively successful ( especially the treatment started within 3—5 days after onset) .

Abstract: Objective To explore a practical fixation method of the unilateral indwelling needle inserted in the Sprague-Dawley rat tail vein covered with essential balm and record the indwelling time. Method 100 male SD rats were divided into two groups randomly: the control group and the improved group. All rats used the same disposable indwelling needle puncture and cannulation. Rats in both groups received the traditional fixation with medical rubberized fabric. In adition to the traditional fixation,the improved group covered with essential balm.The indwelling time was recorded. Result The indwelling time was ( 7. 05 ± 6. 34) h and ( 90. 36 ± 27. 42) h in the control and experimental groups,respectively( t = - 20. 929,P < 0. 01) . Conclusion Our new method is more safe and convenient than the traditional method prolonging the indwelling time effectively and providing a useful fixation method of indwelling needel inserted in the rat tail vein.
