Abstract: In order to obtain materials of eye-mutant mice for human eye related diseases research,generation zero ( G0) mice were mutagenized by N-ethyl-N-nitrosourea ( ENU) ,and G1 generation mice were bred and selected for eye mutant phenotype. All mice were proceeded heritabilitytest,clinical diagnosis and pathological observation respectively. A total of 2782 mice were bred and 65 eye mutants were screened,in which,three cases mice were inheritable,and congenital corneal opacity,nanophthalmos and iris malformation were its phenotypic characteristic respectively. Corneal opacity turbidity was fairly different,especially in terms of hyperplasia and new angiogenesis. The phenotypic traits of nanophthalmos mice was blepharophimosis,blepharosymphysis,and the eyeball was invisible ( severe,general inspection) ,but dysplastic nanophthalmos was visible ( pathological observation) . The phenotypic characteristic of iris malformation was found that the pupil was larger,obvious deviation from the central location,and no light reflection,irido-lens adhesions,collyriculum iridis,retina abnormality ea tl. Three cases of ENU-induced eye mutant mice were bred successfully and lay a foundation for human eye disease research.

Abstract: Objective In the process of making animal models with single Monilia albicans vaginitis,there were two cases of serious infection in mice,so the infection mechanism were studied deeply. Method Isolating fungi and bacteria from mice vagina with severe infection,the 15 fungal samples and 19 samples of bacteria were got after purifying for 3 generations. For fungal samples,we conducted chromogenic medium identification of Monilia albicans. For samples of bacteria,we conducted 16SrDNA extraction,PCR validation and PCR-DGGE analysis, besides we conducted sequence determination of different bacteria DNA samples. Result We isolated 1 kind of fungi and 4 kinds of pathogenic bacteria,it confirmed that the mouse vaginal severe infection is caused by the interaction of 1 kind of fungi and 4 kinds of bacteria. Conclusion The mouse vaginal severe infection model we found,is caused by the mixed infection of 1 kind of fungi: Monilia albican and 4 kinds of bacteria: Proteus vulgaris,Stenotrophomonas maltophili,Alcaligenes Castellani&Chalmers and Bacillus.

Abstract: Objective To observe the effect of a Uyghur medicine,Munziq Sapra,on the mouse model of psoriasis. Method 130 healthy Kunming mice were used in this study. The mouse model of psoriasis was established by applying 5% propranol hydro- chloride emulsion to the mouse ear. The tail and ear skin histology was examined using HE staining,and the serum levels of IL-10,IL-17,IL-23 were measured with ELISA. Result The mice of Munziq Sapra treated groups ( 2,4,8 g /kg) did not show obvious abnormalities of general condition and body weight changes ( P > 0. 05) . The 4 g /kg Munziq Sapra treated group showed decreased thymus index ( P < 0. 05) . The 2,4,and 8 g /kg Munziq Sapra treated groups showed no significant changes of the serum levels of IL-10,IL- 17,and IL-23 ( P > 0. 05) ,but showed promoted formation of epidermis granular layer in the mouse tail skin and improved histology of the ear skin. Conclusion The Uygur medicine Munziq Sapra shows therapeutic effect on experimental mouse models of psoriasis.

Abstract: Objective To inspect the behavioral differences and the retest reliability of methods in two closed group of Kunming mice ( KM) . Method The behavioral differences and the retest reliability of methods in two closed group KM mice were detected by ethological methods. Different closed groups were divided into A,B two groups of 20 male mice. The data of weight,coat hanger,inverted screen,sucrose preference,open field test and forced swimming test were recorded to inspect the behavioral differences and the retest reliability of methods by SPSS17. 0 software after 3d adaptive feeding. The same tests were repeated after 7 d. Result The items of weight,sucrose preference,the horizontal movement of open field test ,score,inverted screen were significantly different ( P < 0. 05) in the statistics at the first experiment. All items of weight,sucrose preference,inverted screen were significantly different ( P < 0. 05) in the statistics in the retest reliability experiment. Fecal pellets test was not credible ( reliability coefficient was negative) ,hanger test and open-field test in the vertical motion score were poor credibility,but other tests are credible ( weight of highly credible) . Conclusion The behavioral differences exist between A and B groups KM mice. The results of reliability ratings are weight > forced swimming test > sucrose preference > horizontal movement > score > inverted screen > coat hanger > vertical movement.

Abstract: Objective To investigate whether silver nitrate has the advantages over conventional antibiotics in the therapy effect of wound infected with S.aureus and P.aeruginosa in mice. Method The research was divided into three groups. Firstly animal models of trauma was established,and the wound surface was infected with S.aureus , P.aeruginosa and mixed infected with the two bacteria,respectively. Then each group was treated with different concentration of silver nitrate. Finally,scab and healing time were recorded and analyzed. Result To the bacterial infection of wound surface,silver nitrate played the same therapeutic effect with antibiotics,and was also effective for the treatment of drug resistance of P.aeruginosa. Conclusion In this study,we confirm the bactericidal effect and wide antimicrobial properties of silver nitrate in vivo. With the bactericidal and bacteriostatic effects of the silver particles and silver ions are re-realized in the modern medicine,the combination of silver nitrate and antibiotics may be an effective therapy strategy in clinical.

Abstract: Objective To study the effect of Dahuang Mudan Decoction ( RPDP) on inflammatory response and oxidative stress in acute pancreatitis( AP) model rats. Method Divided 90 SD rats randomly into three groups: the sham operation( SO) group ,the AP group,and the RPDP group. The models of AP were made by pressurize inject 3% sodium taurocholate into the bile duct where was 0.3mm away from the pancreatic duct with the speed of 0. 1ml /min. The rats in the RPDP group were lavaged with RPDP ( 40 g /kg) after made AP model. The rats in the SO group and the AP group were lavaged with partes aequales Sodium Chloride. Sacrifice the rats 4 h,8 h,12 h hours after establish the animal model,separated the serum and detected the content of IL-6,IL-β and TNF-α in the serum,cut pancreatic tissue in every time quantum and detected the content of MDA,MPO and SOD. Result Compared with the AP group,the serum content of IL-6,IL-β and TNF-α were significantly decrease in RPDP group( P < 0. 05) ,the content of MDA and MPO in pancreatic tissue in every time quantum were also decrease in RPDP group( P < 0. 05) ,while the content of SOD were significantly improve( P < 0. 05) . Conclusion Dahuang Mudan Decoction could repress the release of TNF-α,IL-6 and IL-β,could decrease the product of MDA,and had the effect of anti-oxidative damage,cleaning oxygen free radicals.

Abstract: Objective To compare the differences of three methods for DNA extraction from guinea pigs blood genomic DNA. Method Fresh anticoagulant NaI manual method,Blood clot manual method and DNA extraction kit method were used to extract blood genomic DNA from 20 guinea pigs. The integrity ,concentration ,purity and the effects of PCR amplification of obtained DNA were compared by Spectrophotometer,agarose gel electrophoresis, PCR amplification and other methods. Result The highest DNA integrity,concentration and purity could be achieved by employing fresh anticoagulant NaI manual method. The DNA concentration of blood clot manual method was approximate with the DNA concentration of fresh anticoagulant NaI manual method,but the purity was the lowest,and have some degree of breakage and degration. The DNA concentration obtained by DNA extraction kit was the lowest,integrity and purity were in the middle. There was no differences in PCR amplification results of the three methods. Conclusion The genomic DNA could be extracted by the three methods and could meet the requirements of subsequent PCR amplication,but each of them has its advantages and disadvantages,researchers could choose the suitable method according to specific conditions.

Abstract: Objective To explore the feasibility and reliability of application of anterior cruciate ligament and the medial and lateral meniscus resection of making animal model of OA. Method 20 adult male New Zealand rabbits ,on the right leg knee cut off the anterior cruciate ligament and excision of medial and lateral meniscus,just open the left hind limb joint capsule,no joint operation,observation of postoperative rabbit activities; In 2,6,10 weeks later,observation the left,right posterior articular cartilage pathology. After 2,6,10 weeks,the right leg knee cartilage as the experimental group,the left limb knee joint cartilage were as control group,comparison of two groups of cartilage lesions. Result 2,6,10 weeks later,from the experimental animal activity and gross specimen observation,rabbit right leg knee showed the typical characteristics of OA. Conclusion anterior cruciate ligament and the medial and lateral meniscus resection is an ideal method for a OA animal model making.

Abstract: Objective To discuss the suitable training bouts for Bama mini-pigs,and the treadmill exercise parameters of Bama mini-pigs. Method The exercise training last consecutive 4 weeks,once a day. Speed was progressively increased by 0. 3km /h or its multiples depending on the ability of the the Bama mini-pigs until they can not tolerance. The speed and duration and the changes of vital signs before and after the treadmill exercise were measured. The eight Bama mini-pigs used were 9 - month old,male and castrated,and with the mean body weight of ( 53. 46 ± 2. 52) kg. Result The 8 Bama mini-pigs could run smoothly after 1 week training. After the 3 - week process of skilled treadmill exercise,training bouts included a slow acceleration,a rapid acceleration,a rapid deceleration,and a slow deceleration,with the maximal speed of ( 3. 4—7. 5 ) km /h,the longest duration of 1 530 seconds,and the average duration of 735. 26 seconds. The ear temperature,crissum temperature,heart rate,respiratory rate and systolic pressure incresed significantly. while the respiratory rate was significantly increased after the 3 - week skilled treadmill exercise,the changes of the others of vital signs under the rest state were without statistical significance. Conclusion The training bouts of Bama mini-pigs included a slow acceleration,a rapid acceleration,a rapid deceleration,and a slow deceleration. Nevertheless, it needs further research whether the daily maximum speed and duration can reach the level of the foreign swines.
