The Establishment of Exercise-induced Fatigue Model in Rats

After ten days loading-increasing swimming exercise,the rats showed that the co nditi on of fatigue, and on the 7th and 10th day, the body weight is decreased signif icantly(P<0.05,P<0.01).In the 12SG and 24SG, the CK are increased significantly(P<0. 0 1).In the 0SG and 12SG, the BU are increased significantly(P<0.05).Both t he behavior and chemical index showed that the rats are in the condition of fatigue.The model is suitable for the us e of the experiments of exercise-induced fatigue.

Objective To investigate the effect of “Hua-Yu-Qing-Fang-He-Ji”(HYQG) on hemorheology in Spontaneously Hypertensive Rats. Methods Study was performed on SHR.The hemorheology was determined. Results Middle dosed and hig h dosed HYQG groups could significantly decreased the whole blood viscosity and plasma viscos ity.in SHR.The two groups also could enhance the deformity ability,reduced the eryth rocyte aggregation. Conclusion HYQG has same protective effects on SHR by improving hemotheologic parameters.

Objective To summarize the experience of the reproduction and application of the experimental animal nude mice for 8 years in our laboratory. Methods Randomly drawing out 20 litters of BALB/c nude mice and 5 maternal nude mice.analyze t heir generational state. Results Averagely, one BALB/c nude mou se gives birth to 9.35 mice every time. Female nu/+ and male nu/nu crossbreeding, 55.0 per cent of offspring are nu/ nu homozygous mice,and 97.0 per cent of offspring survives after ablactation.From the first-born to forth-born,the fecundity is steady, the survival rate for 1 year is 100 per cent,and the longe st survival period is 752 days.Microbiological detection couldn’t find any pathogen or parasite whic h should be controlled. Conclusion Under the condition of 100?000 grade air filtrating and in 10?000 grade lamina r flowing cabinet,with strict aseptic manipulation,we bred qualified specific pathogen free(SPF) nude mice.

Objective To get rid of contamination of the transgenic mice. Methods Multi -parameters were got during the purification and analyzed by statistical measu res. Results ① The survival rate of li tter reached 83.3%,if the days of pregnant were correctly calculated. ② The weaning survival rate of litter got to 51.4% when CD-1 mice were used to nurse the litters. Conclusion The suitable conditions of caesarean operation are keys for increasing the survival rate and weaning rate of the litters.

Objective To study the effects of cyclophosphamide on some biological characteristics of l iver carcinoma cells. Method BALB/c mice were injected peritoneally . At 10-{th} day after treatment, the ascites cancer cells were taken, the mo r phologyic and flow cytometric analysis were carried out. Results We found that in cyclophosphamide group, many apoptotic bodies were observed and t he cancer cells in G2M phases were blocked, but the cancer cells in G1 phase dec reased significantly (P<0^05～P<0^01). Meanwhile, DNA index (DI) and proli feration index (PI) increased significantly (P<0.05～P<0.01). Conclu sion Cyclophosphamxide could induce Apoptosis of cancer cells; and also i nterfere the cell cycle of the cancer cells, which may be of value in the cancer treatment.=

Objective To observe the effect of two different whole nutrition forage on guinea pig whi ch may cause hair-lose and weight increase. Methods Those forage contained different content of trams or sheep grass. There were 20%,30% and 40% different content of rice trams or sheep grass. We used them to feed guinea -pig, then observed the situation on hair-lose and weight increase. Re sults After feeding one hundred and five days, the phenomena of guinea-pig hair-lose became lower. More grass content may bring weight increas e and heal the hair-lose fast. Conclusion Guinea-pig should b e fed with some scale of grass powder. The results indicated that sheep grass is superior to rice straw.

We simulated the air distribution at the statuses of empty, transverse and verti cal arrangement of animal cage in SPF animal room. We also analyzed the idiograp hic characters of those kinds of air distribution and compared the good and bad among them. The author simulated and analyzed the relation between the air distr ibution and contamination distribution in the animal room. They reached the conc lusion and gave some advises about how to optimize the air distribution in the l aboratory animal room.

The main progresses on transgenic mouse in last several years in Biotech Institute of Hubei Academy of Agriculture Sciences have been summarized. 4874 eggs were microinjected with 8 genes, such as POMT-PGH, hDAF, pBHSA, pSHSA, PT-HBV1.3, Bcl-xL, hCD59. 556 baby mouse are obtained. The average integration rate was 19.4%. The average transgenic efficiency was 2.2%. Bi-gene transgenic mouse a re o btained by mixture gene injection. The integration rate was 22.2%. The transgeni c mouse of expressing foreign protein was 50% to 100%. Author studied super-ovu lation and several factors of effecting transgenic efficiency. They studied the heredity rule of foreign gene on transgenic mouse via continuous mating and diag nosing of five generations on Bcl-xL transgenic mouse with non-transgenic mous e. The result showed that one of four original transgenic mice could stably inhe rit foreign gene to their offspring. Some transgenic mice don't have the heredit y stability. It is necessary to select original transgenic mice for establishing a line of transgenic mice.

Objective To study the failure cause during established the model of focal cerebral ischem ia in rat.Methods The healthy Wistar rat was obtained,the middle ce rebral artery occlusion (MCAO)was induced by the suture occlusion technique,to d eterminate whether the model is successful by the grade of Neurologic deficit an d the TTC stained.Results The successful rate in summer is better t han in winter and spring.(%P%<0.05).the weight 270～280 g is better than the o thers (P<0.05).the filament length <1.8 cm is better than >1.8 cm (P< 0.0 5).no significant differenceis found between the femaleand the male (P>0.05 ).Conclusion The best condition of established MCAO is rat weight 270 ～280 g,filament length <1.8 cm and in summer.

The technical parameters of the SPF chicken room were checked to con firm whether an overhaul was needed. The steps included cleaning up the room, shutting off the machines, ster ilization, replacing the strainer, mending the central air-condition and final sterilization.
