Objective To use a standard stock for potency testing of HB vaccine in China.Metheods Two inbred strains(DBA/1 and BALB/c) and two NIH closes-colonies of mice were typed in the H-2 region by used microcytotoxicity method and PCR.Groups of mice of the tested strains were immunized with the same hepatitis B vaccine,the titre of anti-HBsAg antibody was analysed by microplate,the ED_(50) was then estimated bu Karder method for each strain.Results Significant differences were found between potency estimates derived from assays using different strains of mice.Conclusion It is likely that the variation of immune response to HB vaccine in mice is correlative with the H-2 haplotype.In some special case,the heterozygosity in H-2 region found in MIH stock could influence the accuracy in such testing even a reference preparation of HB vaccine was used.Base on our experiment,to select a appropriate NIH stocks with the H-2q haplotype for potency testing of HB vaccine in China.

Objective To evaluate method of developing senescence by oral administration of D-galactose.Methods D-galactose was administered to NIH mice and Wistar rats in a daily dose of 70 mg/kg respectively by oral routine and subcutaneous injection to develop senility.Learning and memory retention abilities of the animals were measured by Morris water maze,and monoamine oxidase B(MAO-B) activity in mice brain,superoxide dismutase(SOD) activity malondialdehyde(MDA) level in the liver,and lipofuscin level in myocardium were determined by chemical methods.Results Gain in mice weight was affected by oral administration of D-galactose,and significant difference was not found between the group of oral administration of D-galactose and the control group(P>0.05).However,subcutaneous injection of D-galactose resulted in significant suppressed effect on gain in mice weight in the late stages,and significant differences were found as compared to the control group and the group of oral administration of D-galactose.Learning and memory retention abilities in mice and rats were significantly decreased,with a prolongation of time for swimming to the safe platform in water maze.Aniracetam could significantly anagonize D-galactose-induced degradation of learning and memory retention abilities in rats(P<0.01).MAO-B activity in mice brain and MDA level in the liver were elevated,SOD) activity in the liver was decreased,and lipofuscin level in myocardium was enhanced in the mice of the group of oral administration of D-galactose.The differences in the indexes between the group of oral administration of D-galactose and the control group were very significant(P<0.01),and the significant differences the group of oral administration of D-galactose and the group of subcutaneous injection of D-galactose(P>0.05).Conclusion Oral administration of D-galactose can induce senility and its effect is similar to that of subcutaneous injection of D-galactose.

Objective The objective of this study was to develop a rat orthotopic lung tumor model.Methods Forty female Wistar rats weighing 169 g±10 g were anesthetized intraperitoneally with pentobarbital,tracheotomized and intubated with an ultrafine catheter for inoculation with Walker's tumor cells(1×10~5~5×10~5).Pathological examination was performed and the tumor take rate was determined.Results 39 rats survived among the 40 operated animals.The tumor take rates were 60%,80%,90% and 100% for implants with 1×10~5~5×10~5 tumor cells respectively.Conclusion The present rat orthotopic lung tumor model was shown to be feasible: the take rate was high,surgical mortality was negligible and the procedure was simple to perform and easily reproduced.

Objective To study possible functions of the drug-dependent constipation by observing morphological distributions and quantitative changes of goblet cells and 5-HT cells of the drug-dependent constipation in rats.Methods To development model of drug-dependent constipation in rats and to use the immunohistochemistry SABC,common histological method and cell counting for observing the morphological distributions and quantitative changes of goblet cells,5-HT cell of intestine in rats.Results Compared with the normal control group,the number goblet cells and 5-HT of intestine of rats decreased and the immunohistochemical reaction duoenum deceased obviously in latter.Conclusion The morphological distributions and quantity of goblet cells,immune cells 5-HT has changed in intestinal of drug dependence constipation rat.It is suggested that the changes of morphological distribution and quantity of intestinal goblet cells,immune cells 5-HT may be an important factor of constipation drug dependence.

Objective To induce the oral immune tolerance in rats by oral administration of donor spleen cells and investigate the mechanisms of oral tolerance.Methods Skins of SD male rats were transplanted to the Wistar rats.The Wistar ratswere devided into three groups,each group with eight rats: group A,administrated with PBS,group B,administrated with 1×10~7 spleen cells of SD rats every day,group C,administrated with 5×10~7 spleen cells of SD rats every day.All the recipients were administrated 7 days and radiated with 450 Rad()~(60)Co before administration.The CD4+CD25+、CD8+CD28-Treg of the spleen and blood of the recipients were examed for three times:before administration、7 days and 14 days after administration.Results Survival time of allogeneic skin grafts in group A,B and C were 8.33±1.03,10.17±1.47,19.17±1.94 days,respectively.Compared with group A and B,the survival time of group C were prolong significantly(P<0.01).The DTH responses of group C were decreased significantly compared with group A and B.Conclusions Feeding donor spleen cells can prolonged the survival of skin allografts in rats.CD8+CD28-Treg may play a important role in the mechanism of oral immune tolerance.

Objective This article(study) was purposed to explore a method for rapid establishment of the rabbit atherosclerosis model within a short period of time and its evaluation indications.Methods The rabbit atherosclerosis model sould be prepared by the combination of feeding on hypercholesterol feed and immunological injury procedure.Before,4 and 8 weeks after the feeding on high fat diet,the following indexes should be examined:blood triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein(LDL-C),malonaldehyde(MDA),superoxide dismutase(SOD),nitrous oxide(NO),C-reactive protein(P<0.01). HE staining should be used to view the aterial morphological changes after 8 weeks.Results After feeding for 4 weeks,TG,TC,HDL,LDL,MDA,SOD,NO,and CRP all were increased statistically significantly(P<0.01).Typical atherosclerotic plaques can be observed by both naked eyes and light microscopy.Conclusion The above-mentioned methods can be used to establish a rabbit atherosclerosis model.

Objective To explore all kinds of reactions of animals which were implanted into the tendon suture.Methods The tendon suture and the absorbable catgut suture were implanted into Abdominal Wall of rabbits.Then the biocompatibilities were observed.Results Rabbits had no a fever and no changes in breath and pulsed after implantation.Classification of white blood cell and pathological examination are normally.Conclusion The tendon suture had good biocompatibility and it is an excellent absorbable suture.

Objective To develop a sensitive and specific double-antigen sandwich ELISA(dsELISA) kit for detection serum antibody of simian immunodeficiency Virus(SIV).Methods The envelope protein of human immunodeficiency Virus type 1(HIV-1) expressed in E.coli,was used as detective antigen in dsELISA kit.The dsELISA method was established by optimization reaction conditions and reagent selection.The sensitivity,specificity and reproducibility of the kit were also studied in this series A parallel detection of 400 monkey sera using dsELISA and a recognized standard kit(BioReliance kit,USA) was performed.The equivocal sera were confirmed by VRL lab.Results The kit has good specificity and reproducibility.The kit is stable well when storing at 4℃ for 8 months.More than 96% of the results were coincident with the results detected by BioReliance kit and dsELISA kit.Conclusion The kit developed in this series has several advantages compared with commercial kit,and can be applied in SIV antibody detection and research.

Objective Study of the effects of Two Anti-stressors on Rhesus Monkeys in heat-stressed .Methods Selected six health laboratory rhesus monkeys,and divided into group A,B,Nine hematological and nine blood biochemical were measured,A was added Vitamin C 300 mg/d and Vitamin E 50mg/d each in diets,B group was added mucaosu 0.2%,Two months later,Group A and B was bred under the condition of(32±1)℃,70% humidity for one hour,After that measured hematological and biochemical.Results Test cross-reference hematological was not significantly(P>0.05),After test,AST、ALP、LDH of group A were(101±23.64)U/L,(623.67±233.12)U/L,(1384.33±807.66)U/L and was not significant than before test(P>0.05);After test,TP、ALB of group B were(83.83±16.32)g/L,(38.53±0.57)g/L and was not significant than before test(P>0.05).Conclusion Effects of Vitamin C and Vitamin E were stood balance of AST、ALP、LDH in blood on Rhesus Monkeys in heat-stressed,Mucaosu were stood balance of TP、ALB in blood,finding two anti-stressors had effects on development anti-hotstress on rhesus monkeys.

Objective The comparison of embryonic implantation in order to improve the success of embryo transfer in mice.Methods The KM mice were dealed with PMSG and HCG to make them superovulation,afterwords were mated individually with the male mice.The fertilized eggs were collected from the plugged females.Embryos from the one-cell stage、two-cell stage、morula and blastula were transferred to pseudopregnant recipients.Results By transferring embryos from one-cell stage、two-cell stage、morula and blastula,the rates of pregnancy were 36.36%、42.86%、10%、27.27%,the birth rates were 46.58%、38.46%、58.33%、60%.Conclusion More embryos from the two-cell stage develop to term than those from the other stage.
