Objective To compare with the merits and demerits of intraperitoneal injection in mouse and vein injection in mouse to establish acute immune liver damage tail model by Bacillus Calmette-Guerin(BCG) and lipopolysaccharide(LPS).Methods To compare with different doses BCG of and different doses of LPS induced by intraperitoneal injection of immune mouse established acute liver injury model,and BCG 0.5 mg/only and LPS 7.5 μg/only combined tail vein injection induced Kunming mouse to established the mouse model of acute immune liver injury.To serum transaminase levels in mice and liver pathology as index of liver injury inspection criteria.Results "BCG+LPS" mice by intraperitoneal injection to built the acute immune liver damage model,ALT and AST are increased with the dose gradually increased in section:No.5 Group(BCG 0.5 mg/only+LPS 7.5 μg/only) and No.13 Group(BCG 1 mg/only+LPS 15 μg/ only) is relatively higher.Compared with the control group,ALT increased by about three times,AST is also 1~3 times higher basic;Both of them pathological liver tissue injury appeared with gradually increased doses of BCG and LPS. No.5 Group:most mice Ⅰ appeared Ⅰgrade or above grade Ⅰ liver cell damage;13 Group appeared Ⅱ level in most mice or above grade Ⅱ liver cell damage.But to compared with the tail vein injection modeling,modeling mice by intraperitoneal injection of liver cell injury expression of specific laboratory index of ALT and AST is not high,and liver pathology injury is also lighter.Conclusion Mouse tail vein injection of BCG and LPS established Acuat immune liver damage model is superior to the establishment of intraperitoneal injection of immune liver damage model.

Objective To establish normal range and mean values of blood biochemical parameters in SPF KM mice used in long-term toxicity test.Methods Blood samples were taken from SPF KM mice in different age gender and a number of blood biochemical parameters were measured using an antomatic biochemical and analyzer.Results There are significant difference among mice of different age on WBC、MCH、MCHC、PLT.There are significant difference among day old and sex on RBC、HGB、HCT、Glu、ALT、CHOL、TP、Alb、Glb、A/G、BUN、PO_4、Na、K、Cl、Ca、AG. Concolusion The physiological and biochemical data were significant difference among different age and gender in SPF KM Mice.The normal range and mean values of all data were obtained.This result could be used as normal physiological and biochemical value in SPF KM mice.

Objective To establish and evaluate a mice model with kidney-Yang deficiency syndrome caused by overfatigue and sexual overstrain.Methods Mice had been prepared with compulsive swimming and excessive sexual activity induced by Colldege effect for 4 weeks,then general conditions,body temperatures and tongue pictures of the mice were measured.Results Average body temperature of model group was decreased obviously when it was compared with control group while average body temperature of treatment group was much higher than that of the model group.The model group mice had crenated tongues.In addition,the model group mice had the mantel changes as follows: the papillae of the lamina propria was less in density,the phase frequency of basal layer cells remarkably decreased,the thickness of the lamina epithelium and corneum increased obviously.Comparing with the model group,the density of the papillae increased while the thickness of the lamina epithelium decreased in the treatment group.The phase frequency of basal layer cells tended to increase and the thickness of the corneum tended to decrease,but there was no statistical significance.Conclusion A new mice model for studying kidney-Yang deficiency syndrome caused by overfatigue and sexual overstrain based on classical theory of Traditional Chinese Medicine have been developed.The model is highly in accordance with the conception of kidney-Yang deficiency syndrome in Traditional Chinese Medicine theory.

Objective To set up the animal model of dental fluorosis in the lower incisors in Sprague-Dawley(SD)rats and investigate the changes of enamel surface color and amoblogine in rat incisor exposed to fluoride.Methods 105 male SD rats,average weight 200 g,were randomly divided into four groups.15 rats were with distilled water(control group).The other three group SD rats were rendered fluorotic by the administration of 25 mg/L,50 mg/L,100 mg/L sodium fluoride in their drinking water ad libitum.At the different time sites of 5,10,20,30,42 day,every rat was taken a photo of labial enamel surface of the lower incisor.Then some of the rats were sacrificed,manbibular incisors were removed,fixed,demineralized and sections prepared for H-E staining for light-microscopic morphometric observation.Results The normal color of enamel in rat incisor was yellow/yellow-orange.In this test,the fluorosis model was established successfully in incisors at 50 mg/L and 100 mg/L sodium fluoride groups.The enamel color in incisors were present white-chalk,straition and pitting,the ameloblasts arranged in multiple layers.The apperences of enamel fluorosis were ealier and more severity in 100 mg/L than 50 mg/L sodium fluoride group.The normal incisor color and the normal morphology of the ameloblasts were in the control group and the experimental group 25 mg/L NaF.Conclusion 50 mg/L or 100 mg/L NaF may reduced the dental fluorosis successfully in continuous growth incisors in SD rats.the enamel surface in fluorosed rats exhibited opacity,white-chalk,and discoloration of varying extent.No obvious changes in ameloblasts and enamel surface apperenced in control group and 25 mg/L NaF group.

Objective To study the difference of haematopoietic function between rats of different months-old age,especially the aged and young rats.Method Bone marrow haematopoietic ancestral cells were cultured in the monolayer soft agar of the lung cell culturing conditioned medium containing 15% colony stimulating factor(GM-CSF.).The cell colonies of karyocytes per 4×10~5 were calculated.Result There were 92.5±23.8 colonies in the young rats of 2.5 month-old,but 23.1±9.6 colonies in the aging rats of 24 month-old.As the GM-CFU of the 2.5month-old rats was considered as 100%,it was 37.0% of the 12month rats,35.7%of the 18 month old rats,and 27.2 of the 24month old rats.Conclusion The haematopoietic function of rats decreased as the age was increased.

Objective To establish the partial physiology index,haematology index and biochemistry index.Methods SD rats of 1-month-old,2-month-old,3-month-old,4-month-old,6-month-old,7month-old,9-month-old,12-month-old were slected,each group containing 10 male and 10 female,and to measure the index of physiology haematology and biochemistry of rats.Result The tendency of the weigh increase of male rats was more obvious than that of the female.The coefficients of visceras decreased.Most biochemical values were influenced by age and sex of the rats.

Objective The variation of lipid peroxides(LPO),superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px) were tested in liver and kidney tissues of young and aged rats respectively;and to explore role of three enzymes in senescence process.Method There are ten twenty-one-day-SD rats and ten eighteen-month-SD rats.Liver and kidney tissues of rats were collected at 4 ℃;Thiobarbituric acid reaction was adopted to test LPO;Chemiluminescence used to test activity of SOD and activity of GSH-Px was tested through DTNB.Results Quantity of LPO in young rats' liver and kidney is significant lower compared to it in aged rats(P<0.01);Activity of SOD is markedly higher in young rat group than that in aged group(P<0.01);Activity of GSH-Px was obviously lower in young rat group contrast to aged group.Conclusion There were significant variation of LPO,SOD and GSH-Px in liver and kidney issues between young rats and aged rats.These variations are meaningful for study of senescence process.

Objective and Method The type and distribution of mucous cell stained with AB&PAS in the gut of Pingxiang red-transparent crucian carp were studied.Results The mucous cell in the gut of Pingxiang redtransparent crucian carp could be classified into 4 types: type I,pure red;type II,pure blue;type III,purplish-red;type Ⅳ,purplish-blue.The density of mucous cell presented an increasing trend from the anterior(1ow) to middle(high) and posterior(higher) segment.Conclusion The density of mucous cell in the gut was closely related to the functions of the different segment.

Objective The aim of the study were to investigate the influencing factors about the success rates of embryo transfer at diffferent periods and improve the successful rates of embryo transfer.Methods KM female mouse with superovulation treatment were mated with these normal 615 male mouse,fertilized eggs are obtained.Then these 1-cell embryo,2-cell embryo,morula and blastocyst were respectively transferred into the oviduct and the uterus of pseudopregnancy KM mouse.Results Inflowed 1-cell embryo,2-cell embryo,morula and blastocyst,pregnant rates of the KM mouse were respectively 36.36%,42.86%,10% and 27.27%,farrowing rates are respectively 46.58%,38.46%,58.33% and 60%.Conclusion 2-cell embryo transplantation in KM mouse can improve the success rate of mouse embryo transfer,uterus transplantion can improve the farrowing rates of mouse embryo transfer.

Objective To prepare IBV agglutination antigen via treating with phospholipase C.Methods IBV was inoculated into allantoic cavity of 9-day-old SPF chicken embryo.The eggs were incubated at 37℃ for 48 hours.Allantoic fluid was collected and centrifuged at high speed for concentration,following to individually treated with trypsin and clostridium perfringen culture(containing phospholipase C).Then performing routine agglutination test and agglutination inhibition test.Results IBV haemagglutination showed strong non-specificity by means of trypsin treatment.On the contrary,IBV haemagglutination expressed powerful specific with phospholipase C because the haemagglutination could be inhibited by specific antiserum to IBV,simultaneously could not be inhibited by antiserum to ILTV,NDV,Adeno-1 and 3,APIV,AIV,IBDV,HP,MS and MG.Conclusion IBV treated with phospholipase C could be used for haemagglutination inhibition test.

Objective To explore anesthesia method of cacodontiaes were exelcymosised and implantated again operation.To judgement anesthesia effect in beagle dog.Method Eight dogs were been operated of oral rehabilitation in electrocardio monitoring on anesthesia state.Heart rate,respiration frequency and temperature of the beagle dogs were measured by Naught traumatic electricity custody apparatus.Ease pain effect,breath path secretion and muscle laxity degree of the beagle dogs were also estimated.Result Heart rate of anesthesia state significant higher than normal condition at five minutes to forty minutes(P<0.01;P<0.05).Temperature of anesthesia state all significant lower than normal condition(P<0.01);Breathe rate of anesthesia state than normal condition none significant difference(P>0.05).Ease pain effect excellent;Respiratory passage secretion few;Oral cavity muscle relaxation fine.Conclusion The anesthesia method was convenient,security and anesthesia effect excellent.It can to ensure operative proceed smoothly on cacodontiaes were exelcymosised and implantated again operative in the beagle dog.

Objective To investigate the anaesthesia effect of intramuscular injection of sodium pentobarbital and diazepam combined with intravenous sodium pentobarbital on pignets.Methods Sixteen pignets were initially anesthetized by intramuscular injection of sodium pentobarbital and diazepam.The general anesthesia was maintained by an intravenous dose of sodium pentobarbital.Result Complete anesthesia was observed in all of the 16 pignets.The procedure resulted in complete relaxation of the pignet muscle,allowing experimental manipulation in vivo.All the anesthetized pignets survived after the operation.Conclusion Intramuscular injection of sodium pentobarbital and diazepam combined with intravenous sodium pentobarbital has a good anaesthesia effect in pignets.The procedure provides a useful tool for in vivo studies that demand a complete state of anesthesia.

Objective Eight trace elements(K,Na,Cl,Mg,Ca,P,Fe and Cu) were determined in serum of Macaca mulatta to compare the effect on trace elements in serum of Macaca mulatta lived in different surroundings.The results provided reference information for the selection of feed formulas and the diagnosis and control of nutrition metabolic diseases for artificial breeding Macaca mulatta.Methods The trace elements were detected by automatic biochemistry analyzer.Results There were a significant difference in the content of P,Mg,and Cu between A group(domesticate,raised in cage) and B(semi-natural,lived in park),C group(natural,lived in open air)(P<0.01),but A and B groups have no significant difference(P>0.05);there were a significant difference in the content of Ca between A and C groups(P<0.01),but it wasn't found between B group and A,C(P>0.05);and there were no significant difference in the K,Na,Cl and Fe in all three groups(P>0.05).Conclusion The content of K,Na in all groups were higher than those of normalhuman's upper limit,but the content of P was higher than those of normal human's upper limit only in A group and all the others was in normal range.Some information and test data in the paper are useful and reliable for the further study about trace elements in serum of Macaca mulatta.
