Objective To evaluate the effects of the Sufeirunchang Decoction on rat drug dependence constipation.Methods 30 Clean level SD rats were divided randomly into the normal control group(n=10),model group(n=10)and treatment group(n=10).Beside the control group,each group by limiting water but limitless diet contributes to cause the constipation.It is repeatedly treated by infusing stomach with rhubarb so as to cause medicine dependent constipation.The normal control group rats are filled the stomach with the distilled water.Treatment group tats are filled stomach with Sufeirunchang Decoction for observing the effect of Sufeirunchang Decoction to this disease model.Observation target:Excrement displacement /unit time,First grain of bowel movement discharges time,Barium sulfate advance rate in small intestine.The article compared the effect of Sufeirunchang Decoction around treatment with above targets.Results(1)each model group is worse than the normal control grouping ordinary circumstances excrement displacement is fewer and the nature is harder;intestinal tract transmission function is slow-down.The difference is significant(P<0.05).(2)The control group rats have not any abnormal performance.(3)The ordinary circumstances of model group rats is good after feeding the medicine;intestinal tract transmission function is close to normal control group;the difference is not significant(P>0.05).Conclusion The Sufeirunchang Decoction may improve the function of gastro-intestinal tract of rats;recover drug dependent constipation and promote stomach and intestines creeping motion.

Objective To observe the effects of JianWeiShu Chewable Tablets on the digestion and absorption function.Methods The Distance of KM mice Knk moving was used to evaluate the effect.The mice were randomly divided into treatment groups,control groups and the Difenoxin model control group.Treatment groups fed with 0.67 g/kg,1.34 g/kg and 2.01 g/kg dosage(as 10、20、30 times of population recommended amount respectively).The body weight,food intakes,food efficiency ratio and the secretion amount of Wistar rats' gastric juice and pepsin activity were used to evaLuate the effect.The rats were randomly divided into treatment groups and control groups.Treatment groups fed with 0.34 g/kg,0.67 g/kg and 1.34 g/kg dosage(as 5、10、20 times of population recommended amount respectively).JianWeiShu Chewable Tablets were fed through oral for 30 days.Results JianWeiShu Chewable Tablets could increase the rate of ink promoting,which was superior to that in the Difenoxin modeL controL group(P<0.05).The body weight,food intakes,food efficiency ratio and the secretion amount of gastric juice of Wistar rats compared with control group showed no obvious differences(p>0.05).High dosage group aLso couLd promote the secretion amount of gastric pepsin,pepsin activity,which was superior to that in the control group(\%p\%<0.05).Conclusion[WTBZ] JianWeiShu Chewable TabLets has the function of promoting the digestion and absorption function.

Objective Evaluate the security of Duogu oral liquid as heaith food.Methods According to ministry of Pubic Health《The technical standards for testing and assessment of health food》,to test the Duogu oral liquid by the acute toxicity test with oral administration on mice and mutagenicity test and thirty days feed tese on rats.Results Semi-lethal dose of mice with oral administration was more than 21.5 g/kg,The result indicated that the Duogu oral liquid has no toxicity.Thethirty days feeding test of rats indicated that the body weight was increasing;there were no significant difference of food exploit rate,organ/weight ratio,hematology and serum biochemical guideline quota of every dosage group compared with the control group(p<0.05).The pathological change related to experiment was not found by pathological examination.Conclusion Duogu oral liquid is a health food without toxicity and mutagenicity.

Objective To test the safety of Long-tai capsule as functional food.Methods Acute toxicity test,Bone marrow cell micronucleus test,Mice sperm abnormality test and 30 days feeding test were selected to study the toxicity of Long-tai capsule.Results In acute toxicity test,male and female KM mice were given with 20g/kg dose of Long-tai capsule two times within 24h,no toxic symptom were observed and no death.The results of bone marrow cell micronucleus test,mice sperm abnormality test were negative.In the 30 days feeding test,compared with control group,1.25%、2.50% and 5.00% contents of Long-tai capsule in the feed(as 25、50、100 times of population recommended amount respectively)showed no obvious toxic effects in clinical examination,blood test,biochemical test,organisms weight and pathological observation to Wistar rats.Conclusion Long-tai capsule is safe as a kind of functional food.

Objective To measure the indexes of biological characteristics of organ weight an organ coefficient of tree shrew.Methods 27 tree shrews(15females and 12 males)were chosen,and the body weight and 6 organs' weight were determined,then the organ coefficients were counted.Results Between female and male tree shrew,the brain weight showed significant difference(P<0.01)and the difference of heart,liver was obvious(P<0.05).The coefficient of brain and kidney had remarkably significant difference(P<0.05).Linear analysis showed that there were obvious linear relations between body mass and the weight of heart,liver,kidney in female tree shrew;there were linear relation between body weight and heart in male.Conclusion Differences existed in heart weight,liver weight,brain weight,brain coefficient,kidney coefficient between female and male tree shrew,and there had linear relationships between body weight and some main organs.

Objective The experiment make a pilot study about the technique of isolating pluripotent stem cells from primordial germ cells(PGCs).Methods Fetus was derived from pregnant WuZhiShan inbreeding pigs in 26~28 d.We got fetal germ ridge,then plated on STO cells.The culture medium is composed of DMEM,2 mmol/mL L-glutamine,0.1 mmol/mL non-essential amino acid,0.1 mmol/mL β-mercaptoethanol,100 IU/mL penicillin,100 ug/mL streptomycin and 15% FCS.The embryonic germ(EG)cells was cultured and passaged.Results and Conclusions The EG cells was passaged from F_0 to F_4 or F_5.The morphology of EG cells colonies are multiplicity.By means of Alkaline phosphatase histochemical staining,Expression of stage-specific embryonic antigen-1(SSEA-1)and expression of transcription factor Oct-4 were positive.We identified that these cells possess the characteristics of embryonic stem cells.

Objective To study the homology of MHC-DRB3.2 gene in human beings and some laboratory animals.Methods Cattle genomic DNA was used as the template to amplify DRB3.2 gene fragment by PCR and the products of PCR were sequenced.The DRB3.2 gene fragment of other laboratory animals,including human beings,macaque,sheep,goat and swine were down loaded from GenBank,and base compositions,homology and molecular evolution trees were analysis by DNAMAN software.Results DRB3.2 gene fragment in all those species mentioned above were 267 base pairs and the gene hasn't any deletion or insertion.There is no significant difference in the base contents of A,T,G,C and G+C among those animal species.As for certain special animal,the content of G was the highest and T was the lowest,the content of G+C was obviously higher than that of A+T.The homology between human beings and macaque,sheep,goat,cattle and swine were 91.8%,82.8%,82.4%,81.3% and 81.6%,respectively.Conclusion The nucleotide sequence homology of DRB3.2 gene shares a high sequence identity in different species;macaque plays a very important role in studies of human diseases,and it could not be replaced by other laboratory animals.DRB3.2 gene is an ideal genetic marker for organic evolution and phylogeny analysis.

Objective Monitoring infection of Lymphocytic Choriomeningitis Virus in mice and Golden hamster currently.Methods Using nested-PCR method in mice and Golden hamster to detect the presence of LCMV gene.At the same time to take the serum ELISA test,the results of the two methods were compared.Results[WTBZ] The PCR results showed existence of NP gene in spleen and kidney of mice.PCR positive rate is 2/148,ELISA is 5/148.Conclusion Combining of PCR and ELISA method can improve accuracy rate and prove currently exist in mice LCMV infection.

Objective To measurement normal range of the blood routine values and blood biochemical parameters of the transgenic EGFP mice.Methods Blood samples were measured from 5~6 weeks age male and 6~7 weeks age female using an automatic biochemical analyzer.Results Making a comparison of the male and female EGFP mice,There were very significant differences between the values of Hb and PLT(P<0.01),And the level of Mo and RDW were showed significant differences(P<0.05)in the blood routine values;The Alb and TG had very significant differences in the blood biochemical parameters.Conclusion There were some differences to some blood indicators between the male and female EGFP mice,but were in normal range.

Objective Establish one kind of simple and easy procedure to carry out the quality control to ELISA result.Methods Detect 3677 antibodies of 11 murine viruses to 578 serum of mouse on the basis of GB ELISA,carry out duplicate detection on the 242 positive serum in the initial detection after 56℃ 30 min water bath inactivation processing.Result Duplicate detection on the probable positive serum after inactivation processing.The normal antigen pore A numerus and special antigen pore A numerus obtained in the duplicate detection are obviously lower than that obtained in the initial detection respectively,with a remarkable difference(P<0.05);The special antigen pore A numerus in initial and duplicate detections are very different(P<0.05)from the normal antigen pore A numerus.The positive rates before and after serum deactivation are very different(P<0.01),6.58% and 2.86% respectively.There is no remarkable difference between the A numerus of positive control before and after inactivation(P>0.05).Conclusion The false positive rate is detected to be much lower after the inactivated processing of serum.

Objective To obtain the basic data of biological parameters of Beagle.Methods Routine methods and instrument are used to determine the biological parameters in Beagle using BECKMAN SYNCHRON CX5 PRO.ResultsThere were a few significant differences in Beagle between sexes(male and female Beagle)on parameters of CHOL、TG.Other indexes were no significant differences.ConclusionThere were no significant differences of some biological parameters in different sex and age.The all indexes were relatively stabilization.
