The bovine fetal otic fibroblast cells were isolated using tissue bulk attachment. The gender of fetus was identified using PCR. The fibroblast cells from female fetus were chosen for further morphologic observation,growth curve and chromosome analyzing. The results indicated that the cells had normal morphology,multiplication characteristics and chromosome number. The establishment of culture method and related biological characterization analysis of bovine fetal fibroblast provided not only a ready supply of somatic cells for the intraspecies and interspecies cloning of macaque but also fundamental conditions for somatic transgene and other related fields.

Objective To explore the isolation and culture of mouse embryonic fibroblasts ( MEF). Methods The MEF were isolated from the embryos of BALB /c mouse using in vitro culture system,the growth morphology of the MEF were observed,and the passage cell culture medium,embryonic gestational age,the trypsin concentration were screened. Results The MEF in vitro was a kind of adherent cell,the third,fourth,fifth cell proliferation with high purity and the most exuberant,adding serum M199 and DMEM can satisfy the growth of primary cells, the speed of proliferation had no significant differences in two cell culture medium; the optimal gestational age for isolation of mouse embryonic fibroblasts was from 11. 5 to 16. 5 day. The optimal time for digesting MEF was 8 ~ 11 min with 0. 1% of trypsin. Conclusion By selecting the factors which influence of isolation and culture laid the foundation for further study of the MEF.

To observe the development of testis in mice,the testis of total 17 groups of Kunming normal mice at their different developmental stages among 1 ~ 57 days after birth was examed by using the paraffin tissue section and HE staining. The result showed that only sertoli cells and primitive germ cells exhibited in seminiferous tubules of the early postnatal mice testis; B-type spermatogonia were observed at the 8th day after birth; Primary spermatocytes were observed at the 15th day; Secondary spermatocyte and little round spermatids appeared at the 23th day and then the quantity of round spermatids were gradually increased. At the 30th day,long-shaped sperm cells exhibited. At the 36th day,a large sum of sperms were observed in seminiferous tubules and lasted to whole subsequent stages. It means that via a process of continuously development and differentiation the primitive germ cells matured in about 36 days after birth. This study revealed more details on the development of mice testis and sperms.

Objective To establish a rat model of strangulated intestinal obstruction,which can present good time-dependent relation and unification between the intestinal incarceration time and the degree of intestinal necrosis. Methods By choosing adult healthy male Spraque-Dawley rats,an incarceration hernia model to imitate the strangulated intestinal obstruction was made. In the incarceration hernia model,a midline fascial defect was made,the length of the defect was set by the 80 percent of the diameter of incarceration bowel,and the length of incarceration bowel was 6 cm. All rats were divided into six groups by different intestinal incarceration time,from 1 hour to 6 hours step by 1 hour. With the sham operation group as the contral group,the grade of intestinal injury between the groups was compared by necrosis degrees under microscopy,and the statistic method was ritid analysis. Results Comparing with the control group,the different degree of intestinal necrosis was found,the longer intestinal incarceration time and the higher degree of intestinal necrosis. But with the same intestinal incarceration time,the degree of intestinal necrosis was similar. The mean ritid value of control group was 0. 038; the mean ritid value of each experimental group from 1 hour to 6 hours was 0. 248,0. 329,0. 464,0. 551,0. 765 and 0. 874,respectively. Using LSD test of analysis of variance,microscopic intestinal injury in each eaperimental group was significantly different from the control group(P < 0. 035); the significantly difference of intestinal injury was found between the experimental groups,which difference of intestinal incarceration time was 2 hours or more(P > 0. 09); no significantly difference of intestinal injury was found between the experiment groups,which difference of intestinal incarceration time was 1 hours,except for the 4 hours group and 5 hours group(P = 0. 01). Conclusion A rat model of strangulated intestinal obstruction has been established,which can present good time-dependent relation and unification between the intestinal incarceration time and the degree of intestinal injury.

Objective To study the action of DanlongCAPS for inhibiting thrombosis in Methods The inferior vena of the rat was ligated to make thrombosis in inferior vena,then the dry weight and humid weight of the internal clot was calculated; with the equipment of internal clot formation to observe and record the average blockage rate of the arteria carotis communis in 5 minutes of electric stimulation;the inhibiting effect of DanlongCAPS on adenosine diphosphate ( ADP ) induced by platelet aggregation was measured by platelet aggregation meter. Results DanlongCAPS could reduce the internal clot of the inferior vena,decrease the average blockage rate of the arteria carotis communis,and inhibite the platelet aggregation induced by adenosine diphosohate ( ADP) in the rabbit. Conclusion The results of the study confirm that the DanlongCAPS has the effect of inhibiting thrombosis.

Objective The effects of Heterogeneous Victoria A Acid Capsules on the treatment of the micro- acne on rabbit ear induced by oleic acid and Staphylococcus epidermidis. Methods Rabbit model of the formation of acne with spreading oleic acid to the ear,and subcutaneous injection of Staphylococcus epidermidis,was established,and then the model was treated with Heterogeneous victoria a acid capsules. Results The Heterogeneous Victoria A Acid Capsules markedly decreased sebaceous acid capsule diameter,and hair follicle size. Conclusion The Heterogeneous Victoria A Acid Capsules have a therapeutic effect to the micro-acne on rabbit ear.

Objective To observe the effect and safety of minipig dorsal paired expanded flap model. Methods 6 minipigs with weight 10 ~ 12 kg were chosen. And 6 symmetrical adjoining area (7 cm × 5 cm each) were drawn on the minipig's dorsum. Soft tissue expanders(100 mL) were buried in the left area,and the right area were control group. Results The survival rate in the expanded flap group was statistically different with the control group(P < 0. 05). The survival rate within the expanded flaps group was not statistically different ( P > 0. 05). Conclusion The minipig dorsal paired expanded flap model shows lower difference in expanded flap survival rate than control group. This is a suitable model for expanded flap survival research.

Thymus provides the complex microenvironment which is crucially required for T-cell development and establishing the immune self-tolerance of T cells. Thymic epithelial cells (TECs),which are composed of cortical and medullary TECs and arise from the third pharyngeal pouch,play an important role for this tightly regulated process of T cell development and maturation. Objective To observe the serum hormone changes in multiparous rats with cyclomastopathy model of combination of disease and syndrome. Methods 20 female multiparous rats were divided into normal group and model group. Estradiol benzoate was given to the rats in the model group by intramuscular injection (1. 0 mg /kg) for 10 times, one time every three days. Then, the Progesterone ( 5. 0 mg /kg ) was injected for 5 days continuously. In addition,the rats were bound during the night for 15 times in the model group. The day after the final injection,foods were banded for 12 hours. Then blood was taken through femoral artery for further test after Sumianxin anesthesia. Results In model rats group,high-leveled prolactin and estrodiol accompanied with low progesterone were found in blood serum. Conclusion High-leveled serum estradiol is the major characteristic of cyclomastopathy model of combination of disease and syndrome in multiparous rats.
