Objective Introduce the reproduction of Parkinson disease animal model in C57BL/6 mice induced by MPTP and the behavior analysis.Methods The male animals of C57BL/6 mice were intrapenitoneally injected with MPTP(1-methy-4-phenyl-1,2,3,6-tetrahydropyrindine) at 40mg/kg per day for 7 days,and after injection the behavior tests were performed for each day.The behavior tests include pole test,traction test,swim test and Parkinsonism score.Then the animals were scarified at the seventh day for determining the contents of dopamine(DA) and protein in corpus striatum by HLPC-ECD and modified Lowry method.Results There are significant differences between PD models and controls in behavior tests.The content of dopamine(DA) in corpus striatum in PD models is significantly lower than the controls.Conclusion The method for reproduction of Parkinson disease animal model in C57BL/6 mice induced by MPTP and the related behavior analysis are simple,scientific and reliable.They are good methods for the pharmacological study in Parkinson disease treatment.

To study the effect of vinclozolin on the proliferation of cells,and the levels of lipid peroxide and the activities of antioxidative enzymes in mouse prostate cancer(RM-1) cells,3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) method was used for detecting the proliferation activity of RM-1 cell,and colorimetry method was used for determining the activities of antioxidative enzymes and the level of Maleic Dialdehyde(MDA).The results showed that the proliferation activity of RM-1 cell was significantly inhibited by 0.01 μmol /L,1 μmol /L and 100μmol /L vinclozolin in 24 and 48 h.100 μmol /L vinclozolin significantlyincreased the level of MDA in RM-1 cells in 48 h compared with control group.The activities of Superoxide Dismutase(SOD) and Glutathione peroxidase(GSH-PX) was increased with the increasing of vinclozolin concentration in 24,48 and 72 h.0.01 μmol /L,1 μmol /L and 100 μmol /L vinclozolin significantly inhibited the activity of CAT in 24 h compared with control group.The activity of CAT was increased with the increasing of vinclozolin concentration,and 100 μmol /L vinclozolin group was remarkably higher than those of the other three groups.The results suggested that vinclozolin inhibite the proliferation activity of RM-1 cell,while promoted the levels of MDA and the activities of antioxidative enzymes in the RM-1 cells.

Objective To establish a cigarette induced cough and sputum secretion mice model for effective evaluation of the effect of reduce the dangers of cigarettes with additives.Methods Mice were exposed to smoke through the nose-only device for one hour everyday,the cough latency time and cough frequency were tested by ammonia water method and the OD values of sputum secretions was tested by phenol red method.Results The cough time of model group mice significantly decreased,the cough frequency of model group mice increased,the OD values of sputum secretions of model group mice significantly increased.Conclusion It is successful to establish a cigarette induced cough and sputum secretion mice model.

Objective To investigate the influence of sulfur fumigated Dioscorea opposita Thunb on peroxidation and antioxidation in liver and kidney of rats.Methods Wistar rats were divided randomly into 4 groups,one control and three experimental groups administrated diets mixed with concentrated extractions from unfumigated,commercial and sulfur fumigated Dioscorea opposita Thunb respectively.The level of MDA and activities of SOD and GSH-Px 14 weeks after treatment were measured.Results(1) Dioscorea opposita Thunb enhanced the activity of antioxidation;(2) Level of oxidative stress is different in liver and kidney,and depended on the concentration of sulfur fumigated Dioscorea opposita Thunb extractions.(3) There were sexual differences in levels of oxidation damage by Sulfur fumigated Dioscorea opposita Thunb,so did in various organs.Conclusion It is possible to cause lipid peroxidation in liver and kidmey by long-term use of Sulfur fumigated Dioscorea opposita Thunb.

The chicken and the quail were used as experiment materials.The paper introduced the poultry’s chromosome samples technology which has been improved.The study explained experimental method and condition of the poultry’s chromosome samples by choose suitable experiment animals,improved the key steps of colchicine dosages,temperature and time on low-osmolar,centrifugal speeds.It was established to the poultry’s chromosome samples technology with strong practicality,clearer chromosome samples,stability result,lower cost,simple and feasible method.This will provide an experiment method to study on the karyotype and banding of chromosome for the undergraduate and postgraduate students of agriculture university.

Objective Isolate and cultivate Feline herpesvirus type 1 from clinical suspected cats.Methods The nasal and conjunctival secretion from the suspected cats were cultivated through 4 passages in FK-81 cells and collected,then identified by electron microscopy,hemocyte agglomeration test,physical and chemical character,the nucleic acid type assay,Polymerase Chain Reaction(PCR) amplification and genomic sequencsing.Results Virion was sphericity,and its diameter was approximately 160 nm with envelope.The isolated virus could agglutinate cat red blood cells,but it could not agglutinate red blood cells of pigs,rabbits,canines,mice and chooks.It was DNA virus and was sensitive to acid,heat,ether and trypsin.The isolated virus nucleotide was extracted and amplificated.it shared 100% homology with the sequence of the Feline herpesvirus type 1 in GenBank(ACCESSION:M26660).Conclusion The Feline herpesvirus type 1 was first isolated in China an dacoordinated with the foreign reports in morphology,hemagglutination,physical and chemical character and gene sequence.This finding will provide further research of Feline viral rhiontracheitis in pathology,molecular biology,diagnosis and vaccine.

Objective To determine and compare the hematological parameters of SPF and conventional New Zealand rabbits in weaning 7 day.Methods Hematological parameters of the SPF and conventional New Zealand rabbits were detected using an automatic hematology analyzer and clinical chemistry analyzer.Results For SPF rabbits,the WBC and Tp have had very significant difference between female and male(P < 0.01).The 6 indexes as RBC,GLU have significant differences(P < 0.05).For conventional rabbits,the only 3 indexes as MCHC,LY%,TBIL have significant differences(P < 0.05).Comparing SPF and conventional male rabbits,the 20 indexes as WBC,NE,ALP have very significant differences(P < 0.01),the 5 indexes as RBC,BA have significant differences(P < 0.05).Comparing SPF and conventional female rabbits,the 9 indexes as WBC,NE,ALP have very significant differences(P < 0.01),the 4 indexes as MCH,EO have significant differences(P < 0.05),other indexes had not significant difference(P > 0.05).Conclusion For New Zealand rabbits,the difference of the hematological parameters in same microbiological background is smaller.The Different microbiological background significantly affects the hematological parameters in New Zealand rabbits.

Objective To evaluate the efficacy,the physical disinfection method of ozone and UV was studied in the animal surgery laboratory,in which the main direction is organ transplantation of animal liver,heart and kidney.Methods In an animal clean room,where there is a day-night cycle of operation and rest,the efficacy of disinfection and sterilization was evaluated.Using culture utensils and surface microbe inspections,we compared the physical disinfection methods of ozone and UV to the chemical method of 2% Glutaral.Both sterilization processes were conducted during the night.Results Both methods achieved a normal level of sterilization after three times.However,the ozone is easy to isolate,safe to use and does not require washing.Conclusion Both Ozone and UV are physical factors.They are a good alternative to the chemical sterilization methods,as there are no waste products Also ozone is able to thoroughly permeate all parts of the equipment,including the hard to reach interior.As the needs of organ transplantation research increase,large numbers of equipment and materials need to be kept in the lab for long periods of time.Therefore it is important to establish an effective disinfection method.Our results suggest that at the correct dosage of ozone and UV,we can achieve good sterilization results in the animal surgery laboratory.

Objective To establish a nuclear colony of SPF IRM-2 mice for meeting the needs in scientific research,production and drug quality tests,the conventional IRM-2 mice were cleansed by cesarotomy.Methods The mice were cleansed by cesarean section,and the baby mice were reared and propagated in isolators.Results A SPF IRM-2 nuclear colony with 12 mice was established,and its growth and development of the colony is normal.It met the SPF animal standards indicating by two times of microbiological tests.Conclusion The nuclear colony of SPF IRM-2 mice was established successfully by cesarotomy.

Objective To observe the effect of some natural mineral water on oxidative damage of high-lipid rats.Methods Twenty four female rats were randomly divided into high-lipid control group(drink tap water) and high-lipid experimental group(drink natural mineral water) according to the level of TC.The serum in the two groups was taken for T-SOD activity and MDA content analysis after forty days.Results The MDA content in experimental group decreased markedly than that in control group.Conclusion Some natural mineral water could alleviate oxidative damage by free radicals to extent.
