Abstract: Objective To observe the correlation between the exterior signs of mice model induced by thyroxine and hyperactivity of fire due to yin deficiency syndrome to provide reference for constructing hyperactivity of fire due to yin deficiency syndrome animal model. Method 48 standard weight mice were randomly divided into control group and three model groups,12 each group, male and female in half. Model groups were managed with thyroid tablet suspension of different doses ( 40 mg / kg,80 mg / kg and 160 mg / kg) respectively, whereas the control group with double-distilled water. The body weight, food intake, water intake, body temperature, spontaneous activity and heart rate were measured dynamically. Result As compared with the control group, mice in the model groups manifested slowly increased body weight, enhanced food intake and water intake, elevated body temperature, quicken heart rate and frequent spontaneous activity, which could simulate the major clinical characteristics of hyperactivity of fire due to yin deficiency syndrome, such as emaciation, voracious appetite, thirsty for water, internal heat, palpitation, impatience,et al. Among which, the exterior signs of female mice in high-dose model group were extremely notable compared with the control group ( P < 0. 01 ) . Conclusion The exterior signs induced by thyroxine can simulate the major clinical characteristics of hyperactivity of fire due to yin deficiency syndrome, the correlation between the exterior signs of female mice model induced by thyroxine i. g. 160 mg / kg / d for consecutive 14 days and hyperactivity of fire due to yin deficiency syndrome is more significantly.

Abstract: Objective To investigate the change of reference value for body weight、mortality、hematology and serum biochemistry of SD rats in test method of combined chronic toxicity / carcinogenicity study. Method 135 male and 138 female specific pathogen free SD rat of 4 weeks were selected. We measured the body weights once a week between 4 ~ 17 weeks of age and once four week between 18 ~ 108 weeks of age. The indexes of hematology and biochemistry of SD rats were measured at the age of 8 weeks old、30 weeks old、56 weeks old、82 weeks old、108 weeks old and record the mortality rate every day. Result The body weight of male rats was much higher than female rats from 6 weeks old. The 82 ~ 108 weeks of age was the rapid growth of mortality in rats. The coefficient of variation of indexes of hematology and biochemistry of male and female become larger with increasing age. Gender has a significant impact on hematology and serum biochemistry in rats. But the hematology and serum biochemistry which has significant difference between female and male become less with increasing age. Conclusion During the safety evaluation tests, expecially combined chronic toxicity / carcinogenicity study, both of gender and age have a greater impact on the body weight、hematology and serum biochemistry. Male and female should be separated from the statistics and the animals background data should be used at the same weeks.

Abstract: Objective To study the microsatellite markers of early warning on the response of genotoxicity of phenol in red crucian carp C1HD. Method The red crucian carp C1HD were divided into 4 exposure groups and 1 control group and choose 5 microsatellite markers to study the effect of phenol on the microsatellite polymorphism after exposed to different concentration phenol for 8 d. Result The indexes were increased by the enrichment of phenol in red crucian carp C1HD,such as the gene heterozygosity ( H) ,gene polymorphism content ( PIC) ,and so on. In the 8th day,the gene heterozygosity ( H = 0. 13 ± 0. 19) reached the peak value after exposed to thephenol of 1 /2 LC50 And in the 6th day,the gene polymorphism content( PIC = 0. 42) reached the peak value after exposed to the phenol of 1 /8 LC50 . Conclusion The microsatellite polymorphism will be changed in the red crucian carp C1HD after phenol was enriched to a certain concentration. The microsatellite heterozygosity ( H) and polymorphism information content ( PIC) will be increased with the enrichment of phenol and the extension of exposure time.

Abstract: Objective To investigate the relative factors for experimental atherosclerotic plaque of rabbit induced by immunolesion and high fat diet. Method 30 Japanese white rabbits, intravenously injected bovine serum albumin ( 250mg / kg) ,feeding high fat diet after injection. 7 days later, injected again bovine serum albumin ( 250mg / kg) . Meanwhile, established purely high fat diet group and normal diet group as control,10 rabbits each group. 72 days later, got blood from central artery of rabbit ears to determine lipid level, inflammation factors, vasoactive substance, platelet aggregation and vascular endothelium, etc. and analyzed which related to the pathological change of atherosclerotic plaque or not with SAS statistic software. Result After 72 days, the aorta of both the test group and the purely high fat diet group formed plaque. The plaque of the test group is localized, which is similar to the human being. The lipid level and the content of inflammatory factors,e. g. ,interleukin - 6 ( IL - 6 ) , interleukin - 8 ( IL - 8 ) and high sensitivity-C reactive protein ( hs-CRP) are obviously higher than the control group; the content of vasoactive substance and thromboxane B2( TXB2) changed significantly than the control group ( P < 0. 05) . By the Pearson correlation coefficient statistics processing,And above factor into positive correlation to the plaque size induced by immunolesion and high fat diet; high density lipoprotein ( HDL-C,r = - 0. 58 ) ) , nitric oxide ( NO,r = - 0. 26) are negatively related with the plaque size, and the correlation coefficient of HDL-C is the greatest. Conclusion The rabbit atherosclerotic plaque induced by immunolesion and high fat diet formed in a short time with obvious pathological change, which is similar to that of human beings and closely related to lipid level, inflammation factor, platlet aggregation and endothelial factor, which further show it more reasonable for evaluation the atherosclerosis.
