    BT,a bcnzo(a)pyrenc-induccd human bronchial carcinoma cell line,has been cstablished.To explore themechanism of benzo(a)pyrenc-induced carcinogenesis,p53 gene mutation in BT was examined. It was found that p53 proteinwas overexpres5ed immunohistochenucally within the nuclci. Abnormal clectrophoresis bands in cxon 7 of p53 gene were de-tected by nonisotopic PCR-SSCP,suggesting a gene mutation. DNA scqucncing showed a transversion of CGG to CTT atcodon 248, which catiscs the changc of the encoded amino acid from arginine to lcucin. The results suggcsted that the p53gene mutation might be involved in the benzo(a)pvene-induccd carcinogcnesis.

    his paper reports the cytogenctie comparative studies on human ovarian cancer cell line(HO-8910)establishedby ourself and human ovarian cancer of high metastasis transplatcd in nude micc(NSMO)by using this cell line.Results rcvcalcd that two cell lines were a hyper-diploid karyotype,and no singnificant differencc was observed betwcenHO-8910 and VSMO. The modal number of chromosome was 54.G-banding karyotypic analysis showed that chromosomeloss was often seen on 1.3.15 and 22 chromosome;chromosomc increasc was often observed on 10.16.19and 20 chromo-some,X single chromosome loss was seen on some karyotypes. At the same time four marker chromosomes were found as aresult of the abnormality of the 1.3. 13.14 and 15 chromosome. Various characteristics is that in HO-8910 karyotype besidcsthis four marker chromosomes observed frcquently,some other abnormal chromosomes were also seen. These four markerchromosomes possibly were cytogcnctic charactcristics of Hurnan ovarian cancer of high metastasis It is valuable to furtherstudy ctiology and carcinogesis mechanism of ovarian cancer.

    We have investigated the growth of human nasopharyngeal carcinoma(NPC)cells in nude mice three days af-ter the intragastric infusion of the mixed extract of 5 types of wild LINGZHI.The maimals had been inoculated with cellstaken from human poorly diffcrentiated nasopharyngcal squamous cpitheliocarcinoma ccll line. The number of cells used in in-orulation was 2×106 for cach animal.The dosc of LINGZHl was 9g/kg/d as comparcd to that of normal salinc,20ml/kg/d,in the control group intragastric infusion were given consccutively for 20days in both groups. The room,in which nude micewere grown,was sterilized with a temperature of 25℃ and a relative humidity of 50~ 70%。 After regular observations,theanimals were killed and weighed.The weights of he tumors were also noted down,the tumor inhibitory rate calculatcd andsecretions of the lung and liver examined histologically. The results revealed that wild LINGZHI oral extract may have amarked effect on the tumor formation in human NPC cell Iine. As seen in the results of the three series of our tests,the tumorsuppression rate of LINGZHI was 38.5~44.2% with P<0.001. Body weights of nude mice in all experimental groups in-creased significantly to an extent approximating that of the control groups. The general state of mice in various groups ap-peared normal throghout the study with no toxic side reaction under our experimental condition.

    The subclone cells of nasopharyngeal carcinomas,NPC)CNE-2Z-H5,were inoculated into nude mice.Thecancer cells of lymph node and/or lung metastatic foci were transplanted into nude nuce again when the metastasis in nudemice were observed.Through 2-4cycles like above,the metastatie phcnotype and the expression of transforming growth fac-tor-α(TGFα,epidermal growth factor receptor(EGFR),platelet-derived growth factor receptor(PDGFR)were observed inevery passagc of nude mice. The results showed that the metastatic rates were the higher ,the matastatic paths were more u-niform.and the expression of TGFα,EGFR ,PDGFR were stronger in last passage than in first one,following the progressionof the clone cells of NPC. The results suggested that there are close relationships between metastatic ability and the prolifer-ative ability and strong expression of some growth factor and its receptor in progression of clone cclls from NPC in nude

    The activitics of 0.02%and 0.04% homc-madc minocyclin ahainst M. leprae in the nude mice were cvaluat-cd by the kinetic method,and compared with those of 0.02% imported minocyclin and 0.02%rifampin. The nude mice weretreated for 105 days,The results revcalcd that the activity of home-made minocyclin against M. leprac is similar to that of im-ported minocyclin,and the larger the dosage of the drug is,the stronger the activity of the drug shows. During the early peri-od of stopping treatment,0.04% minocyclin showed the best anti-leprosy effect with the part of bactcriocidal activity.How-ever the activity of rifampin against M. leprae was more persistent and stablc than that of minocyclin when the period of stop-ping treatment is prolouged.

    The hairless mutant was found from ICR out bred stock.The ″nu″gene was retained with the method ofback-inter cross. Then the ICR mice in isolater were used as foster mother. Nucleus colong was cleaned by sterectomies.Microbiological monitoring achievcd to SPF criteria. The mice were bred in laminar flow cabinet after operation. Brecdingtrios of one heterozygous female and one homozygous nude male were used to set up a large breeding colony and named ICR/PMU-nu. At the same time,teste of imrnuno-biology Were made to mutant nude nuce Post-mortem examination of morethen ten nude mice between 3 and 10 weeks of age. Their thymies were abscnce. The lymphoeyte transformation test wasmade of the thymus,spleen and lymph nodes of seven nude mice. The stimulation inder is 0.71±0.50 of the thymas,L 60±0. 64 of the splecnand 0.85±0.13 of the lymph nodes,SI<2.The nude mice have no T cell.no response to ConA. The Smlgls not dlfferent from the control. ICR/PMU-nu nude mice showed a high human tumor transplantation retes,8/10(100%)for E-J,2/2(100%)for Bq23,2/2(100%)fur X19,2/2(100%)for PG,2/2(100%)for PC3M and a high human tumormetastasis. Hybroma cells of 2-5×106/0.4ml were injceted into abdomen cavity to produce McAb 45ml of ascitcs of micemonoelonal antibolies SD3.IL-5 and Ia were obtained.

    LTEP-a-2 human lung adenocarcinoma cells and CVE-2Z human nasopharygcal carcinoma cells were usedin prcsent experiment.Tumor cells were injected into different sites(including site of subeutaneous costal region and subcu-taneous footpad)and different ways(including inoculation of suspension of tumor cells and tumor fragment)into 24 scidrnice(severe combined immunodeficience mice)and 48 sitcs. Experiments were divided into two groups,named as group ofsuhcdtaneous inoclatuon into costal region(including 6 LTEP-a-2 tumor-bearing scid mice and 6 CNE-2Z tumor-bearing scidmice)and group of subcutancous inoculation into footpad(including 6 LTEP-a-2 tumor-bearing scid mice and 6 CVE-2Z tu-mor bearing scid mic).Two transplantable sites in cach mouse were chosen for this study,i.e the suspcnsion of tumorcclls was implanted into one site,and the tumor fragments were implanted into opposed site in same mouse:All tumor-bcar-ing scid mice were sacrificed on 68th day.In the group of subcutancous inoculation into costal rcgion,CNE-2Z tumor couldgive rise to growth(100%)with progrcssive invasion.and lung mctastasis which occurred in 50%(3/6).LTEP-a-2 tumorwere grown in the form of nodule with incomplete thinner fibrous capsule(100%).slight tumor invasions were noticed insome mice. metastasis was not observed In the group of subcutaneous inoculation into footpad,after inoculation of tumorfragment of CNE-2Z tumor cells into 6 scld mice tumor give rise to growth(100%)with progressive invasion and lymphaticnetastasis which occurred in 66%(4/6)of cascs,And after inoculation of suspcnsion of CNE-2Z tumor cells into 6 scidrnicetumor give rise to growth(50%)without invasion and mctastasis. In thc group of subcutancous inoculation LTEP-a-2 tumorcclls into footpad tumor growth was observed in the form of module with fibrous capsul(100%).and slight invatsion was pre-xented in the some cascs,metastasis was not observed In the solid tumors of some cases of CNE-2Z and LTEP-a-2 tumorsthere was an accompanying squarnous element.the mcchainsm was unclcar.
