    Preliminary Study on Normalization of Developing Deficiency of qi of the Spleen in Animals——A Study on Changes of Part of Immunologic Functions

    Objective To delve into the changes of immunologic functions in normatively developing of deficiency of QI of the spleen in rats.Method\ Based on improving and perfecting of the previous method of developing deficiency of qi of the spleen and diagnostic criteria for deficiency of \%qi\% of the spleen,the changes in partial cytoimmunity,nonspecific immunity,and erythrocyte immunity during developing deficiency of \%qi\% of the spleen by the preliminarily normative method were observed in Waster rats and the results were compared with those of the other five representative methods of developing deficiency of \%qi\% of the spleen.In preliminarily normative method of developing deficiency of \%qi\% of the spleen, purging by using drugs of bitter in taste and cold in nature,and wasting \%qi\%,dispelling the stagnated \%qi\% and checking upward perverted flow of \%qi\% with the durgs were adopted based on screening of the rat's constitution of traditional Chinese medicine,and oral administration of decoction of Sheng Da Huang (生大黄 Radix et Rhizoma Rhei),oral administration of decoction of the durgs for wasting \%qi\% and dispelling the stagnated \%qi\%,controlled feeding, overfatigue,and hypodermic injection of reserpine were adopted respectively in the other five representative methods.Results\ According to the changes in wet weight and index of spleen,thickness of thymic cortex,diameter of lymph sheath surrounding central artery of spleen,proliferative function of splenic T\|lymphocyte,killing rate of splenic NK cell against YAC\|1cell,and phagocytic activity of splenic macrophage,deficiency of \%qi\% of the spleen caused by the preliminarily normative method was considerably superior in conformity with the diagnostic criteria for deficiency of \%qi\% of the spleen to deficiency of \%qi\% of the spleen caused by the other five representative methods,and the restoration of deficiency of \%qi\% of the spleen caused by the preliminarily normative method was slower.Conclusion\ The goal of the optimization and normalization in developing of deficiency of \%qi\% of the spleen was attained in the experiment.

    Objective To investigate the effects of chronic restrained stress for the viewpoints of the neuro endocrinol disorder and the regulation of the mediation of Compounded Prescribtion of Smoothing the Liver of the Traditional Chinese Medicine(TCM).Method A stressed model was built by chronic restrained stress for 3h per day. Evaluating the behavior changes, measuring the ratio of the adrenal gland and the thymus grand and the content of monoamine neurotransmitter, and abserving the effect of the Chai Hu Shu Gan San to discuss the mechanism of the Compounded Prescribtion of Smoothing the liver. Result The Chaihushugansan can alleviate the changes of the physical strength and the emotion, and mediate different monoamine neurotransmitter.Conclusion Chai Hu Shu Gan San can improve the dysfunction of chronic restrained stress situation in some degree.

    Objective\ To observe therapeutic effect of the Duoqi capsule.Method\ Duoqi capsule is composed of the blood of Jinling female Wuya and Calcium, the pharmacodynamics effect of Duoqi capsule on model mice after castration was observed. Results\ Number 1 capsule can improve remarkedly uterus weight of animal ( p <0 05) and skeletal density( p <0 05 and p <0 01). Number 2 capsule can improve the level of estradiol in blood ( p <0 05), and the capsule reduced the level of blood phosphorus. The mineral substance of bone was improved remarkedly ( p <0 05 and p <0 01). Conclusion\ Duoqi capsule can supply body with estrogen, maintain metabolism of bone, reduce osteopenia and prevent osteoporosis.

    Objective To study growth and development as well as reproductive capacity of RCS rats. Methods 46 rats (70—80days old) were divided into two groups and were mated to analyze the body weight gain and other reproductive capacity.Results Average litters per cote were 7.9, and the weaning time, the death rate of litters in group 2 is 10% lower than that of litters in group 1. Conclusions The study supplied a basic data for researching on ophthalmology.
    Changes of Bone Metabolism in Rats of 4 and 12 Week after Ovariectomized

    ObjectiveTo observe changes of bone metabolism in rats of 4 and 12 week after ovariectomized (OVX).Methods 28 female Sprege Dawley rats (4 month age)were randomly divided into four groups:①Sham OVX 4 weeks group(5 month age);②OVX 4 week group;③Sham OVX 12 weeks group(7month age);④OVX 12 weeks group. The changes of bone metabolism with quantitative bone histomophometry were observed. Results\ In comparison with sham OVX 4 weeks group, the OVX 4 weeks group's Trabecular area percent (%Tb.Ar) decreased, and all the parameters that reflect bone resorption and formation increased. In comparison with Sham OVX 12 weeks group, the parameter of OVX 12 weeks group show same change with OVX 4 weeks group compared with Sham OVX 4 weeks group. %Tb.Ar of OVX 12 weeks group didn't show significant change compared with OVX 4 weeks group.Conclusion\ ①The activity of bone formation and resorption increased and bone volume decreased in OVX 4 and 12 weeks rats.②OVX 4 week group,the speed of bone lost is most fast in the first 4 weeks. ③Compared with 5 month age rats, the activity of bone formation and resorption of 7 monthp age rats decreased, regardless of sham OVX or OVX group.

    Objective To observe the effect of extracted liquid of alcohol in cortex mori on glucose and serum lipid in diabetic rats. Methods The diabetic rats were pured into stomach with extracted Liquid of alcohol in cortex mori(2g/d) to measure the levels of glucose and lipid after 30 days. Result In the experimental group,the level of glucose was decreased from(20 4±0 6)mmol/L to(12 0±0 7)mmol/L, with significantly difference ( t=2 35,P <0 05),but the level of serum lipid was not significantly different (t=0 83,P>0 05). Conclusion The extracted liquid of alcohol in cortex mori is effective on lowering the glucose of diabetic rats, but not effective on serum lipid.

    Objective To determine the acute toxicity (LD 50 ) of SJAMP(stichopus japonicus acid mucopolysaccharide) via intramuscular injection on rats and mice. Methods LD 50 were tested by simplified probit method. Results The mice were injected with SJAMP intramuscularly, LD 50 was(598 81±60 81)mg/kg. LD 50 of male and female mice were(652 42±85 31)and (578 16±88 68) mg/kg,respectively ( P >0 05). The rats were injected with SJAMP intramuscularly, LD 50 was (566 09±85 17) mg/kg. Conclusion The acute toxicity of SJAMP im was significantly lower than intravenous or intraperitoneal injection. The main fatal cause of SJAMP im on animals was hemorrhage.

    Objective To know the quality of laboratory animals,to publicize and carry out the new GB, the pathogen of conventional mice and rats in Shandong province from 2000 to 2002 was checked. Method Conventional mice and rats were choosed from different unit of different regions in Shandong province,then we tested whether they carried the pathogenic bacteria and virus according to the standards of clean laboratory animals. Results The sera positive rates of Mouse Hepatitis Virus and Sendai Virus were 11% and 19% respectively. The sera positive rate of other virus is zero in mouse.The sera positive rates of rat Hanta virus and Sendai virus are zero. The positive rate of Bordetella browchiseptica in rats is 8 3%.But all the other pathogenic bacteria,except Mycoplasma spp. and Tyzzer's organism unchecked,are negative. Conclusion Shandong province had improved obviousely in the quality of laboratory animals,attained the conventional standards.But the existence of MHV and SV in mouse and the hidden infection of Bordetella bronchiseptica in rats suggested that was needed to spread clean mice and rats in Shandong province.

    Adopting one step and two\|step method,we use Lats knock\|out mice embryo to optimize the condition of vitrified with straw and cell freezing tube.The developing percentage of thawed embryo reached 90%,90.1%,89.2% and 91.4%,respectively.The implantation ratio reached 50%,47.1%,43.4% and 46.4%,respectively.The difference is not remarkable compared to the control and each other.So the Lats knock\|out mice can be preserved by vitrification.

    Objective To investigate the perioperative nursing of orthotopic heart transplantation in pig.Methods\ Thirty orthotopic heart transplantation in pigs were performed,and perioperative nursing were patiently observated.Results\ The successful rate of the operations was 70%.Meantime,the complications of all cases were found in time and treated appropriately.Conclusions\ Positive preoperative preparation,familiarity with operative procedure,actively cooperation during operation,and intensively post\|operation nursing and observating,orthotopic heart transplantation in pigs can be performed smoothly.
    Experience of Breeding Nude Mice in Laminar Flow Cabinet

    Aim To summarize the Experience of breeding nude mice in laminar flow cabinet.Methods\ 2 sets of JD\|25 laminar flow cabinet were used and maintained according to the instruction manual provided by the producer.560 nude mice in 18 groups were bred in the laminar flow cabinets in a separated clean room for the total time of 6 years.The room was disinfected every day and the lights were on for 10 hours each day.Fallout bacteria were detected every month.Results\ The room temperature was kept 22℃—28℃,humidity 40%—60%,bacteria colony number was 3/dish on average.The ammonia concentration in the air of the room and cabinets was lower than 10ppm,positive pressure was kept in the cabinets,the noise in the room was below 55dB.All the nude mice bred in the cabinets were well enough for the experimental studies.Conclusion\ Laminar flow cabinet used in a clean room is safe and easy way to breed nude mice.
