Objective\ To investigate the difference of antigenicity between human and pig skin.Methods\ Frozen se ction of full thickencss skin of human and pig were stained with immunohistochemical method,using the following anti\|human antibodies:HLA\|ABC,HLA\|DR,vimentin,desmin,actin,FⅧRA,cytokeratin,hyaluronic acid,fibronectin,laminin,type Ⅰ/Ⅲ/Ⅳ and Ⅶ collagen,estradiol\|17β,and testosterone.Results\ The antigens of collagen of types Ⅰ,Ⅲ,Ⅳ,Ⅶ,hyaluronic acid,fibronectin,laminin,FⅧRA,cytokeratin,IL\|6,IL\|8,estradiol\|17β,testosteron,and ICAM\|1 were detected in pig skin.The positive rate was higher with polyclonal antibodies than with in monoclonal antibodies.There are relatively good cross reactivity of extracellular matrix(ECM),cytokins with smaller molecules,and proteins with primitive functions,such as IL\|6,IL\|8,ICAM\|1,estradiol\|17β and testosterone in skin of human and pig. However,the cross reactivity of recipents of cytokins and CD(cluster of differentiation)molecule in skin were relatively poor.The results showed the specificity and species variation of antigen and antibodies.Conclusion\ Antigens are different between human skin and pig skin.This may be the possible reason for rapid rejection following skin xeno grafting.
