Objective\ To explore the methods of inducing the animal model of heart\|\%qi\% deficiency syndrome.Methods\ The animal model was established by giving basic amount of food for 24 days,compelling to swim with a burden of 5% of their weights until they exhausted,and feeding a great quantity of Propranolol (2.4mg/100g) in 40 SD rats.In order to prove the model was indeed the heart\| qi deficiency syndrome model,the authors divided the animal into ginseng group and normal control group.Results\ Compared with the normal group,the cardiac functions of contraction and relaxation in the model group decreased;the quantity of myocardial SOD decreased which reflects the deficiency syndrome,and the quantity of MDA increased.All the changes of the fixed quantity indexes were coincident with heart\| qi deficieny syndrome.Conclusion\ The multiple methods, giving basic amount of food,compelling to swim with burden and feeding Propanolol to establish the model of heart\| qi dificiency syndrome,are feasible.

Objective\ To explore the factors that affect the hemagglutination and hemagglutination\|inhibition test. Method\ Hemagglutination and hemagglutination\|inhibition test in different dilution and different serum treatments and 7 kind of reactive plates were compared. Result\ The test condition of clear image, regular and stable result was obtained. Conclusion\ The results provided some experimental data for fine instructions of the national standard.

Objective\ To investigate the difference of antigenicity between human and pig skin.Methods\ Frozen se ction of full thickencss skin of human and pig were stained with immunohistochemical method,using the following anti\|human antibodies:HLA\|ABC,HLA\|DR,vimentin,desmin,actin,FⅧRA,cytokeratin,hyaluronic acid,fibronectin,laminin,type Ⅰ/Ⅲ/Ⅳ and Ⅶ collagen,estradiol\|17β,and testosterone.Results\ The antigens of collagen of types Ⅰ,Ⅲ,Ⅳ,Ⅶ,hyaluronic acid,fibronectin,laminin,FⅧRA,cytokeratin,IL\|6,IL\|8,estradiol\|17β,testosteron,and ICAM\|1 were detected in pig skin.The positive rate was higher with polyclonal antibodies than with in monoclonal antibodies.There are relatively good cross reactivity of extracellular matrix(ECM),cytokins with smaller molecules,and proteins with primitive functions,such as IL\|6,IL\|8,ICAM\|1,estradiol\|17β and testosterone in skin of human and pig. However,the cross reactivity of recipents of cytokins and CD(cluster of differentiation)molecule in skin were relatively poor.The results showed the specificity and species variation of antigen and antibodies.Conclusion\ Antigens are different between human skin and pig skin.This may be the possible reason for rapid rejection following skin xeno grafting.

Objective\ To study immunologenic characteristics of recombinant hepatitis E vaccine.Methods\ Mice and rabbits were immunized with recombinant hepatitis E vaccine by multipoint intradermal injections and muscle injections,then the serum antibody titer was measured.Resaults\ The serum antibody was detected 7 days after mice immunized.The positive rate was 80% after 2 weeks and 100% after 4 weeks. In rabbits,antibody was detected after 2 weeks and reached the peak after 3 weeks,the serum antibody positive rate was 40% on 1∶6400 dilution.Conclusion\ The results indicated that the recombinant hepatitis E vaccine had good immunogenic and can be used for hepatitis E vaccine research.

Objective\ To investigate the dose of pentobarbital sodium in anesthesia of rats. Method Pentobarbital sodium was diluted in 0 9% germfree sodium chloride to get 0 3% pentobarbital sodium solution and then injected the anesthesia solution into peritoneal cavities of rats at the dose of 1mL/100g. Results and Conclusion The process of anesthesia was stable and was kept long enough. The respiration and heart rate are normal. The method is easy to perform and control.

The studies were based on testing the following parementers:percentages of CD4 +, CD3 +, CD8 +, CD19 + cells and tilles of IgG by FCM or ELISA. The results showed significant difference in the percentages of CD8 +, CD3 + and CD19 + cells between mice keeping in the barrier system and conventional condition. The ratio of CD4 +/CD8 + cells was lower in mice of barrier system than those in conventional condition. The studies contribute a base parameters for the mice maintained in barrier system.

In order to decrease the amount of sudden death and increase the rate of laying, we trided to limit the feed uptak for specific pathogens free(SPF) layers, which was in their medium or late term of laying. One layer's feed was reduced from 125g to 107g per day. The result shown that there's no sudden death in test group and the laying rate is about 5 percents more than the control group on average.

Using 3Rs principles, which includes reduction, replacement and refinement in the animal experiment is an important application. Propagation of avian adenovirus type Ⅰ and Ⅲ with SPF chicken liver cell culture could replace chicken kidney cell culture. By observing cell pathogenic effect and detecting antigen titers, the resuts showed the former method can completely replace the later, which is a practice applying 3Rs principle. Utilizing possible replacement instead of laboratory animals. The methods of both cell culture and tissue culture can replace live animals to carry out experiments in order to minimize animal suffering.
