    Objective To set up the animal model of type 2 mellitus diabetes in SD rats.MethodsSD rats were fed with a diet enriched in fat and fructose for 4 weeks to induce insulin resistance,and the rats were injected intraperitoneally with a low_dose streptozocin(STZ) once(30mg/kg) to induce hyperglycemia after 2 weeks.ResultsInsulin sensitivity index decreased significantly of experimental group and hypertension was seen after rats fed with a diet enriched in fat and fructose.There were hyperglycemia,increased blood insulin and lower insulin sensitivity in rat models after STZ injection.ConclusionThe experiment provides an ideal model of type 2 mellitus diabetes with SD rats.

    ObjectiveThe aim of this study was to investigate the purgative function of green tea.MethodsThe mice stomach were fed with the green tea at the doses of 835\,1670 and 3340mg/kg,respectively, once a day for 8 days.And then mice bowels propulsion,the time of the first soil and the soil's quantity and weight were measured.ResultsThe results showed that green tea could accelerate the freated mice bowels propulsive movement,shorten the time of first soil and increase the quantity and weight of the soil in treated mice.Conclusion The results demonstrate that green tea could relax the bowels.

    Using the technology of immunohistochemical staining and color image analysis to investigate the spatiotemporal pattern and age_related changes of type Ⅰ collagen expression in the corneal development of Bufo raddei larvae during metamorphosis.Results showed that the inner and outer cornea combined together into mature cornea during metamorphosis.Both corneal epithelium and corneal stroma become thickened with progressively increase of the age.The analysis of immunohistochemical staining by color image analysis found that type Ⅰcollagen expressed widely during the period,existing in Bowman's membrane,Descemet's membrane,corneal stroma and endothelium.Espeacially,a progressively change in corneal stroma was observed.ConclusionThe defect of qualitative index may be remedied by color index in the quantitative immunohistochemical image analysis of type Ⅰ collagen.The inner and outer cornea combine together gradually with type Ⅰ collagen increasing in the corneal development of Bufo raddei larvae during metamorphosis.Type Ⅰcollagen,which is the main component in the increased collagens,expresses progressively with age.Type Ⅰcollagen may be the important component in corneal stroma,and it plays an important role in the construction and maintaining of the corneal stroma.

    ObjectiveTo prepare rabbit serum and test its usefulness.MethodsRabbit serum was prepared from blood obtained from femoral artery and compared the efficiency of rabbit serum by inspecting character of cell growth and cryoprotecting ability with foetal bovine serum.ResultsCell division index was 1%~2% and cryopreservation survive rate was 90% in 10% rabbit serum medium,compared with 0.5% and 85% in 10% foetal bovine serum medium.ConclusionThe method of preparation is simply and rabbit serum is preferable to foetal bovie serum for rabbit tissue culture.

    ObjectiveTo study anatomic characteristics of swine cerebral arteries and investigate applications in human cerebral arteriovenous malformations.MethodsHead and neck angiography and anatomization were observed in 6 domestic swine,and anatomic characteristics were observed then.ResultsIn swine,bilateral common carotid arteries originated from the brachiocephalic trunk,then issued the ascending pharyngeal and external arteries.The left vertebral artery originated from the left subclavian artery,while the right vertebral artery originated from costocervical trunk.The brain was mainly supplied by bilateral pharyngeal arteries.The pharyngeal arteries formed rete mirabile(RMB) inside sphenotic foramen between sphenoid saddle,then RMB issued bilateral internal carotid arteries supplying the brain and there was inter_communicant between bilateral RMB.The external arterial system formed communication with RMB,further with intracranial internal carotid arteries,by the means of ramus anastomoticus(RA) and arteria anastomotica(AA).There were also anastomoses between vertebral and occipital arteries and among intracranial arteries.ConclusionBased on cerebral arterial anatomic characteristics,the AVM model could be constructed in swine for experimental studies in hemodynamics,pathophysiology and embolic materials,and for interventional neurosurgeon trainings.Nevertheless,there are some insufficiencies in swine AVM model in terms of morphological and hemodynamic simulations of AVMs.

    ObjectiveTo establish PCR detect method of the laboratory monkeys and biological products.MethodsKidneys,lungs and spleens of 56 monkeys and 10 poliovirus vaccines were examined by PCR using designed three pairs of primers on SV40-776 sequence.Results Results indicated three pairs of primers obtained exactly expectable results.Neither viscera of 56 macaque monkeys nor 10 poliovirus vaccines were positive by PCR.Conclusion The three pairs of designed primers are available and can be used for quality control.

    The canine Hepcidin gene was amplified by RT-PCR from canine liver and cloned to pGEM-T-Easy vector.The gene was sequenced,and the result indicated that the gene was canine Hepcidin sequence,it was 258bp.After comparing it with the genes released in Genbank(AY773532),a nucleotide mutation was found.

    Objective To identify a strain of P.mirabilis isolated from laboratory mice which may provide an useful reference for bacterial detecting and identifying in laboratory animals.Methods The suspicious strain of Salmonella spp isolated from SPF mice was identified by cultural characteristics,colony morphology,staining,motility and biochemical testing,and was further identified with the slide agglutination test.Results The results of cultural characteristics,colony morphology,staining,biochemical testing,and slide agglutination test showed that the isolates was a strain of P.mirabilis but not Salmonella spp.Conclusion Although SPF mice are allowed to be P.mirabilis carriers in our country,it is worthy to state that P.mirabilis is an conditional pathogen,which may cause some potential harm to researchers and laboratory animals,and also can cause certain adverse effect to scientific experiments.It is still to be considered as an important affair because of the special use of the carriers.

    The experimental adult KM mice was pta into the same cage(male:female 1:2)divided into four goup in order to get the embryo on different stage.Group 1 is controle group fed with the standard feeding,group 2 fed with melon seeds,group 3 fed with the makt,group 4 fed with SUBU on d4/d8 and dl(the first pregnant day is dl).The embyo was observed respectively.There was a obviously improvment in the quantity and quality of the embyo on different stage from group 4.Compared with the traditional superovulation technique,the feeding method for getting more and fine embyo is safe and convenient in KM mouse.
