    Tests of Coagulation Time of Four Kinds of Coagulation Factors in ICR Mice,SD Rats,Wistar Rats,and Mongolian Gerbils

    Objective To test coagulation time of four kinds of coagulation factors in ICR mice,SD rats,Wistar rats,and Mongolian gerbils and compare difference in coagulation time of four kinds of coagulation factors among the animals.Methods Prothrombin time(PT),activated partial thromboplastin time(APPT),fibrinogen time(FIB),and thrombin time(TT) were tested by HemoClot MC-2000plus in ICR mice,SD rats,Wistar rats,and Mongolian gerbils.Results TT in SD rats,Wistar rats,ICR mice,and Mongolian gerbils was respectively 31.72 s,47.35 s,106.34 s,and 35.96 s;APPT was respectively 17.66 s,20.49 s,20.74 s and 16.27 s;FIB was respectively 20.45 s, 18.20 s,22.81 s,and 21.35 s;PT was respectively 21.00 s,21.76 s,16.74 s,and 25.16 s.The differences in TT between ICR mice and other 3 kind of animals,and between SD rats and Wistar rats were significant((P<0.05));the difference in PT between ICR mice and Mongolian gerbils was significant(P<0.05);the differences in APTT between ICR mice and Wistar rats,between ICR mice and SD rats,between Wistar rats and SD rats,and between Mongolian gerbils and Wistar rats were significant(P<0.05);and the differences in FIB between ICR mice and SD rats,between ICR mice and Mongolian gerbils,between SD rats and Wistar rats,and between Wistar rats and Mongolian gerbils were significant ((P<0.05)).Conclusion There were significant differences in PT,APPT,FIB,and TT among SD rats,Wistar rats,ICR mice,and Mongolian gerbils.

    Objective To determine blood physiological indexes of p21~(HBsAg/HBsAg) and p21~(HBX/HBX) transgenic mice of different ages so as to delve further into the effect of HBsAg/HBsAg and HBX/HBX transferred into p21 locus of mice on their blood physiological indexes. Methods The blood physiological indexes of p21~(HBsAg/HBsAg) and p21~(HBX/HBX) transgenic mice of different ages were determined and the results were compared with those of wild mice.Results The differences in many blood physiological indexes between male and female mice were very significant,and the differences in blood physiological indexes between p21~(HBsAg/HBsAg),p21~(HBX/HBX) transgenic mice and wild mice were very significant.Conclusion The transferring of HBsAg/HBsAg and HBX/HBX gene into p21 locus of mice could exert the effect on their blood physiological indexes.

    Objective To understand the effect and the degree on various organs of rat by stress.Methods The rats were treated by stress of hungry,chill and tiring up,and then the pathological changes of different tissues and viscera in rats were observed grossly and microscopically.Results After stress above administration,there were the most significant lesions in digestive system,more obvious lesions in respiratory and urogenital system,slightly changes in nervous and endocrine system,and no any changes in circulatory and locomotory system.Conclusion Animal growth and health are affected by stress,and also there are certain effects on animal productive practice and economic losses by stress.
    A Comparison of OPS and SSV Vitrification Freezing Preservation of KM Mouse's Immature Oocytes

    Objective To search the new way of freezing preservation of KM mouse's immature oocytes.Methods The immature oocytes from KM mice of 5~6 weeks old were equalized for 5 minutes in 10% EG,or a mixture of 10% EG and 10% DMSO,next were equalized for 30s in different freezing liquids,and then were freezingly preserved by OPS(open pulled straw) and SSV(solid-surface vitrification).Results In mouse's immature oocytes vitrified by OPS,over 84.4% oocytes preserved in EFS were morphologically normal and the mature rate was less than 16.7% after thawing;the 90.5% oocytes preserved in EDFS30 were morphologically normal and the mature rate was 22.5% after thawing;91.9% oocytes preserved in EDFS40,were morphologically normal and the mature rate was 40.5% after thawing,with a very significant difference as compared with the control group(P<0.01).In mouse's immature oocytes vitrified by SSV,86.0% oocytes preserved in EFS were morphologically normal with a mature rate of 46.5% after thawing;92.3% oocytes preserved in EDFS30 were morphologically normal with a mature rate of 57.2% after thawing;90.1% oocytes preserved in EDFS40,were morphologically normal and the mature rate was 48.4% after thawing,with a very significant difference as compared with the control group(P<0.01).In mouse's immature oocytes vitrified by OPS and SSV,morphologically normal rate of the oocytes was respectively 91.6% and 91.4%,the differences were significant as compared with the control group(P<0.01);mature rate of the oocytes was respectively 42.9% and 59.1%,with significant difference as compared with the control group(P<0.01).Conclusion The effect of combination of multiple cryoprotective agents is better than that of single cryoprotective agent,and the effect of EDFS freezing liquid,particularly EDFS30,is better than that of EFS in cryopreservation of mouse oocytes.SSV method is superior in cryopreservation of mouse oocytes to OPS method.

    Objective To observe effects of dietary with different protein contents on growth and reproduction of KM mice.Methods 120 KM mice(weighting 25 g,male and female being half and half) were randomly divided into the control group,the experimental groupⅠand experimental groupⅡ;and the mice in the control group were fed with dietary containing 20% protein,the mice in the experimental groupⅠwith dietary containing 18% protein and adding with definite number of lysine and methionine,and the mice in the experimental groupⅡwith dietary containing 16% non-animal protein and adding with a large number of lysine and methionine.The growth and reproduction of KM mice were observed.Results As compared with the control group,feeding with dietary containing 18% protein and adding with definite number of lysine and methionine did not impair significantly growth and reproduction of KM mice,and feeding with dietary containing 16% non-animal protein and adding with a large number of lysine and methionine could significantly impair growth and reproduction of KM mice.Conclusion The feeding with dietary that protein content was decreased by 4% and was absent from animal protein(fish powder) could significantly impair growth and reproduction of KM mice.

    Objective To compare the difference in the temperature range of conventional and SPF rabbits.Methods The temperatures in 166 conventional rabbits and 196 SPF rabbits were taken and recorded at once every ten minutes for four hours.Results The standard deviation of mean temperature was higher in conventional rabbits than in SPF rabbits.Conclusion The temperature of SPF rabbit is more stable than that of conventional rabbits.
    A Comparison of Blood Biochemical Parameters in The Minipigs of Three Different Strains

    Objective To determine and compare blood biochemical parameters in the minipigs of three different strains.Methods Blood biochemical parameters were determined in experimental Gunagxi Bama and Zhong Guo Nong Da minipigs of four months old and Wuzhishan minipigs of five months old by Hitachi 7600 fully-automatic biochemical analyzer.Blood biochemical parameters were compared between female and male minipigs,and 13 parameters were compared among the minipigs of three different strains.Results Similar blood biochemical parameters were found in different strains of minipigs.In 13 parameters,ALT and BUN were significantly lower in Guangxi Bama minipigs than in other two strains(P<0.05).TG was significantly higher and ALP was significantly lower in Wuzhishan minipigs than in other two strains(P<0.01).The differences in most of blood biochemical parameters(except BUN) between female and male minipigs were not significant(P>0.05).Conclusion Most of blood biochemical parameters in same strain are not affected by sex,and three strains of minipigs have approximate blood biochemical parameters.

    Objective To confirm diagnosis of feline infectious peritonitis by the method of molecular biology and pathology.Methods A pair of primers was designed according to 3′UTR conservative domain of coronavirus genome.A reverse transcriptase PCR(RT-PCR) was carried out in some tissues from 4 cats died because of feline infectious peritonitis and ascitic fluid of 10 cats with feline infectious peritonitis.PCR product was sequenced.Histopathological examinations of the tissues from the died cats were performed.Results 97~98 percent of the sequence of the PCR product was identity with the sequence of feline infectious peritonitis virus gene in GenBank.The disease was pathologically characterized by disseminated inflammatory granuloma,focal necrosis,and infiltration of mixed inflammatory cells in small bowel serosa,liver,spleen,etc.Conclusion Diagnosis of feline infectious peritonitis has been confirmed in the four cats,and 10 ascitic fluids contained nucleic acid of feline infectious peritonitis virus.

    Objective To investigate relevant characteristics of experimental dogs and provide the data for selection of experimental dogs in safety evaluation for drugs.Methods 28 Beagles from Guangdong,20 Beagles from Shanghai,and 24 hybrid dogs from Wuhan suburb were used in this series.Their electrocardiogram and blood routine examination were done,and their blood biochemical indexes were determined before and after oral administration of H_2O(2 mL/kg).Results Heart rate was faster and QRS and QT intervals were more extended in Wuhan hybrid than in Guangdong Beagles before the experiment.Haemoglobin level,erythrocyte and neutrocyte count were higher in Wuhan hybrid than in Shanghai Beagles before oral administration of H_2O and leucocyte count was lower in Wuhan hybrid than in Shanghai Beagles after oral administration of H_2O for 4 weeks.Haemoglobin level and erythrocyte count were higher in Wuhan hybrid than in Guangdong beagles before oral administration of H_2O,and erythrocyte count was higher in Wuhan hybrid than in Guangdong beagles after oral administration of H_2O for 4 weeks.ALB,Cre,GLU,ALT,and AST were significantly higher and ALP,BUN,and CHOL were lower in Wuhan hybrid than in Shanghai Beagles.AST,TP,ALB,and GLU were significantly higher and ALP was lower in Wuhan hybrid than in Guangdong Beagles before oral administration of H_2O.Conclusion Beagle should be popularly used in safety evaluation for drugs.
    Application of Ozone Disinfection in Running of Barrier System for Laboratory Animal

    Objective The correct use of ozone research on experimental animals barrier environment for disinfection methods and concentration,to ensure that both disinfection effectiveness and environmental barriers that the experimental animals and staff security.Methods A check experiment on sterilization effects is conducted between two barriers in which the concentrations of ammonia gas are 0.32 mg/m~3and 0.32 mg/m~3.Clean ICR mice are selected for normal reproduction.The next step is to observe what influences the ozone sterilizations with different concentrations have on the birth rate and pulmonary functions of the mice as well as on the concentration of ammonia gas in the air.Results It is found that ozone sterilization does take place both when its concentration is 0.21 mg/m~3 and 0.32 mg/m~3.When the concentration of ozone is 0.32 mg/m~3,the rate of dead fetus and the case number of lung collapse of laboratory animals is obviously different from those of the control group and the group in which the concentration of ozone is 0.21 mg/m~3,and the concentration of ozone dropped 85%.When the concentration of ozone is 0.21 mg/m~3,the concentration of ozone dropped 75%,and there is no distinctive difference of the rate of dead fetus and the case number of lung collapse of animals between this group and the control group.Conclusion When the barrier environment of laboratory animals is running dynamically,the most appropriate dose of ozone disinfection is 0.21 mg/m~3.

    Objective To investigate sterilizing effect of fumigating of formaldehyde by electrically heating on barrier system.Methods Formaldehyde solution was heated in the electric boiler without potassium hypermanganate to fumigate the barrier system.The culture plate was exposed to the barrier system for 30 minutes,then was incubated for a week in 37℃ incubator.The fallen bacteria count was carried out.Results The fallen bacterium in the barrier system was 0.42 per plate after being fumigated for 4 hours,which was in keeping with the demands of barrier system.Conclusion The method of fumigating of formaldehyde by electrically heating could be applied to sterilize the barrier system for laboratory animal.
