Pathological Evaluation of Skin Injury and Healing by Different Modeling Methods

摘要: 目的 探讨不同造模方法对皮肤损伤的影响及不同时间段创面皮肤愈合的病理学评价。 方法 实验选用SD 大鼠 78 只,雌雄各半,分为正常对照组(14 只) 、烧伤模型组(24 只) 、烫伤模型组(24 只)和全切皮肤模型组( 16只) 。 其中烫伤模型组采用台式超级控温烫伤仪烫伤皮肤组织,并于创面滴入混合菌,建立浅 II 度烫伤合并伤口感染模型。 烧伤模型组采用自制烧伤锥形瓶烧烫伤皮肤组织,并于创面滴入混合菌,建立浅 II 度烧伤合并伤口感染模型。 全切皮肤模型组采用无菌手术刀对皮肤进行面积为 3 cm×3 cm 切除,切除不伤及肌肉,建立皮肤全切模型。正常对照组分别于 D1、3、4、5、7、10、14 取 2 只动物进行解剖取材,烧伤和烫伤+细菌感染模型组分别于 D1、3、5、7、10、14 各取 4 只动物进行解剖取材,全切皮肤模型于 D4、7、10、14 各取 4 只动物进行解剖取材,对创面皮肤组织进行病理学检查。 结果 大体观察发现,不同模型造模初期皮肤均出现不同程度创面损伤;中期皮肤损伤部位周围均出现红肿、水泡等局部反应;末期皮肤愈合较为完整。 显微镜观察发现,各模型初中期皮肤主要表现为表皮变性坏死,角化不全,颗粒层、棘细胞层及基底细胞层缺乏,真皮不同程度炎性细胞浸润;末期皮肤表皮各层结构较为完整,真皮层可见大量新生血管,成纤维细胞聚集及少量炎症细胞浸润。 结论 通过对造模不同阶段皮肤组织的病理学评价,较为全面的分析了不同造模方法对皮肤的损伤与愈合情况,为后续药物药效学研究奠定了基础。

Abstract: Objective To investigate the effects of different method of modeling on skin injury and thepathological evaluation of wound healing in different time periods. Method Seventy-eight SD rats, halfmale and half female, were divided into normal control group ( 14 rats) , burn model group ( 24 rats) ,scald model group (24 rats) and total skin resection group (16 rats) . In the scald model group, the skintissue was scalded by desktop super temperature control scald instrument, and the mixed bacteria weredripped into the wound surface to establish the model of superficial II degree scald combined with woundinfection. Burn model group was established by using self-made burn conical flask to burn skin tissue anddripping mixed bacteria into the wound surface to establish the model of superficial II degree burncombined with wound infection. In the total skin resection model group, the skin was resected with asterile scalpel in an area of 3 cm × 3 cm without muscle injury to establish the total skin resection model.第 3 期 肖 洒等:不同造模方法对皮肤损伤及愈合情况的病理学评价Two animals of the normal control group were dissected and the samples were taken on the day 1, 3, 4,5, 7, 10 and 14 respectively. Four animals of the burn and scald + bacterial infection model group weredissected and the samples were taken on the day 1, 3, 5, 7, 10 and 14 respectively. Four animals of thetotal skin resection model were dissected respectively on the day 4, 7, 10 and 14. The skin tissues of thewound were examined pathologically. Result The gross observation showed that the skin of differentmodels had different degrees of wound damage at the initial stage of modeling; in the middle stage, localreactions such as redness, swelling and blisters appeared around the skin injury site; the final skinhealing is relatively complete. Microscopic observation showed that the skin of each model in juniormiddle school was mainly characterized by epidermal degeneration and necrosis, incomplete keratosis,lack of granular layer, acanthocyte layer and basal cell layer, and different degrees of inflammatory cellinfiltration in the dermis. At the end of the period, the structure of each layer of skin epidermis isrelatively complete, and a large number of new blood vessels, fibroblasts and a small amount ofinflammatory cells can be seen in the dermis. Conclusion Through the pathological evaluation of skintissue at different stages of modeling, the injury and healing of skin caused by different modeling methodwere comprehensively analyzed, which laid the foundation for the follow-up pharmacodynamic study.
