Analysis on Changes of Related Indexes in Different Periods of Hyperlipidemia of Rat Model

Objective To explore the level of blood lipid and oxidative stress in different periods of hyperlipemia of rat model, and to scientifically analyze its changing regularity. Method SD rats were randomly divided into control group ( normal diet and drinking water ) and treatment group ( normal drinking water, high-fat feed: basic feed +15% lard +20% sucrose +1. 2% cholesterol +0. 2% sodium cholate) . Each group was divided into five sub-groups: 0, 15, 30, 60 and 90 d. The general state of animals was observed and recorded. Animals were treated at different time, serum was taken, blood lipid level and oxidative stress level were detected, liver was weighed, liver coefficient was calculated, some livers were placed in 4% paraformaldehyde, HE histopathological staining was performed, some livers were homogenized, and blood lipid level was detected. Result In the treatment group, the fur of the animals was dull, and there were depilation and slow movement. The weight increased significantly from 15 d, and the liver coefficient increased significantly from 30 d. Compared with the control group, the difference was statistically significant ( P < 0. 05 ) . TG and CHO in serum of treatment group were increased significantly on 30 d and 60 d, and the difference was statistically significant (P<0. 05) , but the difference of TG on 90 d was not statistically significant compared with that of control group. From the 15 d of treatment group, MDA in serum increased and SOD decreased significantly, and the difference was statistically significant ( P < 0. 05 ) . T-AOC decreased significantly from the 30th day, and the difference was statistically significant (P<0. 05) . From the 15 d, lymphocytes were gathered, liver cells were swelled and degenerated, nuclei were squeezed to one side. Until the 90 d, liver cords were disordered, liver cells were swelled, fatty degeneration occurred, and vacuoles with different sizes in cytoplasm were compressed and deformed. Conclusion The modeling rate of hyperlipemia animal model was high, but in the research and analysis of drug prevention and treatment, the change of metabolism and the length of time should be considered. Besides serum indexes, it was suggested to increase the detection indexes of tissue and other metabolic enzyme activities, so as to analyze the effects of drugs more scientifically.
