    Comparison of Ocular Changes after Simulated Long-term Microgravity Exposure between Albino and Non-albino Rats

    Objective To compare the changes of ocular structure and function in albino rats and nonalbino rats after simulated long-term microgravity exposure, and to improve and supplement the animal model of ocular injury caused by simulated long-term microgravity exposure. Method Male SD and BN rats, each 12, were randomly divided into control group (NS and NB ) and model group ( MS and MB ) . 30 ° headed-down tail suspension for 8 weeks was used to simulate microgravity for model group. At the end of the model, electroretinogram (ERG) and visual evoked potential (VEP) were used to detect the function of retina. The morphology of retina was observed by HE staining, and the changes of blood vessels in fundus were observed by small animal fundus imaging system. Result After 8 weeks of simulated weightless suspension, the structure and function of the retina of Sprague-Dawley rats ( SD, albino rats) changed significantly, and the amplitude of ERG dark adaptation 3. 0 response decreased significantly (P< 0. 01) , the peak time of P1 wave of VEP was significantly prolonged (P<0. 01) , HE staining showed thinning of the outer nuclear layer (P<0. 01) , increased blood flow of choroidal vessels was observed in the fundus, while there were no significant changes in retinal structure and function in Brown-Norway rats (BN, non-albino rats) after suspension. Conclusion Albino rats exhibit heightened vulnerability to ocular damage when exposed to prolonged simulated weightlessness, thus warranting investigation into the role of melanin synthesis-associated proteins within the retinal pigment epithelium as a potential means of mitigating retinal degeneration induced by prolonged simulated weightlessness.

    Objective The expression level of peripheral hemoglobin aGal in the offspring of the Wuzhishan inbred min-Pig (WZSP) and its aGal knock gene pig offspring was carried out. Method The expression levels of peripheral erythrocyte aGal in the offspring of different generations of WZSP inbred lines and their GGTA1 / b4GalNT2 knockout pigs were measured by using the separation technology of fluorescein isothiocyanate ( FITC) . Result The average percentage of immunofluorescence response of aGal on the 26 heads red blood cells in the inbred line F27- 28 was 56. 17% ( 16. 2% - 86. 2%) . Among them, the average value of F24 8 heads red blood cells of aGal cells was 65. 39% ( 35. 5% -99. 2%) ,the average value of F1 9 - 21 21 heads is 69. 67%(38. 8% -89. 4%) . The average values of the group group 1 and group 3 were significantly different ( P< 0. 05) . The further analysis showed that the percentage of immunofluorescence response of inbred red blood cells of aGal cells in inbred F27- 28 and F24, F24 and F20. individual red blood cells of aGal cells decreased by 3. 07%, 4. 19% per generation; with the advancement of inbreeding lines, the immunofluorescence response of red blood cells of aGal cells decreased significantly. The immunofluorescence response of 8 offspring red blood cells of the knock gene modified pigs was 0. 1% which was consistent with the control group and could be regarded as homozygotes. It was with extremely low aGal expression was found, indicating that its immune rejection ability for xenotransplantation was low also. The study also found that the aGal expression of two siblings at F27 WZSP was lower ( 16. 2% and 19. 5%) than that ( 31. 03%) of knock gene heterozygous pigs. Conclusion With the advancement of inbred lines, the expression of aGal in individual red blood cells was decreased gradually. Eight GGTA1 / b4GalNT2 knockout pigs were homozygous. The inbred line F27 was an individual with low expression of aGal.

    Objective The guide dogs’ working status of Labrador retrievers was evaluated by complex environment test, to explored the relationship between exploratory and working status of Labrador retrievers. Method Using the improved DMA test method of Swedish Working Dog Association, 41 Labrador dogs from Dalian training base of Chinese guide dogs were evaluated, and the dogs were divided into groups according to the score of exploratory behavior. Design a test method for complex environment, score the working state of guide dogs under different environmental stimulation conditions, analyze whether there are differences in the working state scores of different exploratory guide dogs in complex environment, and explore the relationship between exploratory and guide dogs’ working state. Result There were significant differences in the working state scores of the three groups of seeing-eye dogs under the stimulation of novelty objects (P<0. 05) , and there were significant differences in the working state scores of seeing-eye dogs with different genders under the stimulation of lighting and sound in complex environment (P < 0. 05) . Conclusion The working state of dogs with high exploratory ability is less affected when facing novelty objects. Under the condition of light and sound stimulation, the working state of male dogs is less affected. Exploratory males are more adaptable to complex environments.

    Objective To explore the mechanism of miR-494 on the pathogenesis of osteoporosis in rats by targeting TLR-4 pathway. Method Thirty 8-week-old female Sprague Dawley rats were selected to construct the rat model of osteoporosis. Bone mineral density was measured by dual energy X-ray absorptiometry. The contents of osteocalcin (BGP) and total alkaline phosphatase ( TALP) in rat serum were detected by ELISA. Rat bone marrow mesenchymal stem cells (BMSCs) were cultured. qRT-PCR was used to detect the expression of mir-494 and TLR4 mRNA. The expression of TLR4 protein was detected by Western blot. Double luciferase reporter gene verified the targeting relationship between miR494 and TLR-4. The proliferation ability of BMSCs in each group was detected by CCK8 experiment. Alizarin red S staining was used to detect the osteogenic differentiation of BMSCs in each group. The adipogenic differentiation of BMSCs in each group was detected by oil red O staining. Result Compared with NC group, bone mineral density, BGP content, cell proliferation ability and the number of mineralized nodules in OP group were significantly decreased, while the content of TALP, the expression of miR-494 and TLR4 and the number of adipocytes were significantly increased ( P<0. 05) . Compared with OP group, the bone mineral density, BGP content, cell proliferation ability and the number of mineralized nodules in OP + miR-494 inhibitor group were significantly increased, while the content of TALP, the expression of miR-494 and TLR4 and the number of adipocytes were significantly decreased (P<0. 05) . Compared with OP + miR-494 inhibitor group, the bone mineral density, BGP content, cell proliferation ability and the number of mineralized nodules in OP + miR-494 inhibitor + TLR-4 vector group were significantly decreased, while the content of TALP, the expression of miR-494 and TLR4 and the number of adipocytes were significantly increased; In OP + miR-494 inhibitor + sh TLR-4 group, bone mineral density, BGP content, cell proliferation ability and the number of mineralized nodules increased significantly, while the content of TALP, the expression of mir-494 and TLR4 and the number of adipocytes decreased significantly ( P < 0. 05) . Conclusion MiR-494 can accelerate the progress of osteoporosis in rats by promoting the expression of TLR-4 pathway. To provide a new theoretical basis for the pathogenesis of osteoporosis.

    Objective To evaluate the reliability of establishing a young rat model of bronchial asthma with depression behavior by solitary cage combined with ovalbumin. Method Young SD rats were randomly divided into control group, solitary control group, asthma group and solitary asthma group. The rat model of asthma with depression behavior was established by solitary cage and gregarious cage feeding combined with ovalbumin-aluminum hydroxide suspension intraperitoneal injection sensitization and ovalbumin normal saline atomization stimulation. The rat’ s weight, physical signs, lung pathology, white blood cell count and classification in venous blood, and serum content of IL-4 BDNF were observed and recorded. The behavior of rats was evaluated by sugar consumption test and forced swimming test. Result Compared with the two control groups, the weight growth of gregarious asthma group was slow ( P < 0. 05) , and the weight growth of solitary asthma group was significantly slow (P<0. 05) ; the gregarious asthma group and solitary asthma group had typical asthma like symptoms such as polypnea, scratching ear nose, the solitary asthma group had depression symptoms such as less activity and less water and food intake; the lung tissue had asthma pathological changes; the white blood cell count and lymphocyte count were increased (P<0. 05) ,compared with the two control groups and gregarious asthma group, IL-4 in serum increased ( P< 0. 05) , BDNF in serum decreased ( P< 0. 05) , sugar preference index decreased (P<0. 05) , immobility time prolonged in forced swimming test (P<0. 05) . Conclusion It is reliable to establish a young rat model of bronchial asthma with depression behavior by solitary cage combined with ovalbumin.
    Effects of Different Feeding Methods on Growth Performance and Blood Indexes of British-shorthair Cats

    Objective In order to study the feeding method suitable for experimental cats. Method 18 female British short haired cats aged 7 months were randomly divided into three groups, with 6 cats in each group. The experiment was conducted in three ways, namely, cage breeding, regular release of active breeding and captive breeding. The total duration of the experiment was 8 weeks. The body weight, average daily gain, average daily food intake, average daily water intake and blood physiological and biochemical indexes of experimental cats in each group were measured and analyzed. Result The weight of 7-9 month old experimental cats was not significantly affected by different feeding method;in the first week of the experiment, the average daily increase of the regular release of active breeding group was significantly higher than that of the captive group ( P < 0. 05 ) . From the third week, there was no significant difference in growth performance indicators between the captive group and the regular release of active breeding group;The weight growth rate of the experimental cats in the cage group was slower than that of the other two groups, and there was a significant difference between the cage group and the other two groups at the 4th, 6th and 8th weeks ( P < 0. 05) ,In terms of daily average food intake, the regular release of active breeding group in the first week was significantly higher than the cage group (P< 0. 05) , the regular release of active breeding group in the second week was significantly higher than the cage group and the cage group (P<0. 01) , and in terms of daily average water consumption, the regular release of active breeding group in the fourth week was significantly higher than the cage group ( P < 0. 05) ;the lymphocyte ratio ( LY%) test result showed that the captive group was significantly lower than the regular release of active breeding group and the cage group ( P<0. 01) , the number of neutral cells (NE) test result showed that the captive group was significantly higher than the cage group (P<0. 05) , and the platelet distribution width ( PDW) test result showed that the captive group was significantly lower than the regular release of active breeding group and the cage group ( P < 0. 05 ) ; the total cholesterol ( T-CH) test result showed that the caged group was significantly lower than the cage group with regular release of active breeding combination ( P < 0. 05) . Conclusion Regular release of active breeding is suitable for long-term rearing of experimental cats.

    Objective To investigate the role of three imaging modalities including MRI, in vivo imaging and ultrasound in evaluating chemotherapy in lymphoma-bearing mice. Method 10 BALB / c female mice were subcutaneously injected with lymphoma cells on the right neck and shoulder to build a tumor-bearing model, and were randomly divided into a CTX intervention group and a 0. 9% NS intervention group. Before and after the intervention, MRI, in vivo imaging, and Ultrasound were used to detect tumor-related indicators. After the detection, the mice were sacrificed, and the tumor was taken for volume measurement and pathological examination. Result MRI accurately demonstrated a decrease in tumor size after intervention, which was comparable to gross measurements. In vivo imaging can indicate that the tumor size was reduced and the fluorescence intensity of the tumor decreased after the intervention. Combined with the analysis of the pathological section, the CEUS demonstrated that the perfusion of the tumor-related blood vessels decreased after the intervention, and the differences were statistically significant. Conclusion The three imaging modalities have the advantages of good imaging effect, high imaging sensitivity, non-invasive and reproducible, and the combination of multiple imaging modalities has a better synergistic evaluation effect on the intervention indicators of tumor mouse models.
    Sulforaphane Reduces Chronic Mechanical Pain in a Rat Model of Endometriosis

    Objective Based on the HMGB1 / TLR4 / MyD88 signaling pathway, to explore the effect of sulforaphane on chronic mechanical pain in rats with endometriosis ( EMs) . Method The EMs model was constructed by autologous endometrial tissue transplantation in rats. The rats were divided into sham operation group ( sham group) , EMs group, sulforaphane low-dose group (5 mg / kg) , sulforaphane high-dose group ( 15 mg / kg) , HMGB1 inhibitor group ( EP group) , and TLR4 inhibitor group ( TAK-242 group) . The pain threshold of rats was detected by an electronic pain meter; the levels of HMGB1, TNFα, IL-6 and IL-10 in rat serum were detected by ELISA; the protein levels of HMGB1, TLR4, MyD88 and TRPV1 in ectopic endometrial tissue were detected by Western blot. Result Compared with the Sham group, the paw withdrawal threshold ( PWT) of the EMs group was lower (P<0. 05) ; the levels of serum HMGB1, TNF-α, IL-6 and IL-10 were higher ( P < 0. 05) ; the expression of HMGB1, TLR4, MyD88 and TRPV1 proteins in ectopic endometrial tissue was increased (P<0. 05) . Compared with the EMs group, low and high doses of sulforaphane, HMGB1 inhibitor, and TLR4 inhibitor could increase PWT of rat and reduce levels of serum HMGB1, TNF-α, IL-6 and IL-10, and reduce expression of HMGB1, TLR4, MyD88 and TRPV1 proteins in ectopic endometrial tissue ( P < 0. 05) . Conclusion Sulforaphane can reduce the mechanical pain caused by EMs by inhibiting the HMGB1 / TLR4 / MyD88 signaling pathway.
    Comparison of Bactericidal Effects of 5 Disinfectants on 4 Kinds of Bacteria

    Objective To verify the bactericidal effect of 5 kinds of disinfectants on 4 kinds of bacteria which must be excluded in SPF experiment animals, this study was performed. Method D / E neutralizing broth was used as neutralizing agent to investigated its neutralizing effect on 5 disinfectants. The bactericidal effects of the 5 disinfectants on Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumoniae and Salmonella typhimurium were investigated by suspension quantitative bactericidal test. Result D / E neutralization broth had good neutralization effect on bromogeramine solution, Jianzhisu disinfectant, Didecyl dimethyl ammonium bromide solution, BIOCID-30 and 75% alcohol solution. The bromogeramine solution, Jianzhisu disinfectant, BIOCID-30 and 75% alcohol solution had good killing effect after 1. 5 min respectively with four kinds of bacteria. The 600 times diluted Didecyl dimethyl ammonium bromide solution could effectively kill Staphylococcus aureus,  Klebsiella pneumoniae and Salmonella typhimurium in 5 - 10 min, while its bactericidal effect on Pseudomonas aeruginosa was not ideal. The 200 times diluted Didecyl dimethyl ammonium bromide solution could kill Pseudomonas aeruginosa effectively for 1. 5 min. Conclusion The bromogeramine solution, Jianzhisu disinfectant, BIOCID-30 and 75% alcohol solution could produce good bactericidal effect on four kinds of bacteria within a short time. A higher concentration of Didecyl dimethyl ammonium bromide solution could effectively kill Pseudomonas aeruginosa.
    Establishment of Real-time PCR Method for Diagnosis of Tyzzer’ s Organism in Rabbits

    Objective To establish a real-time PCR method for diagnosis of Tyzzer’s organism in rabbits. Method One sets of primers and probes were designed according to the 16S rRNA sequence of Tyzzer’ s organism. The concentration of primers and probes was optimized, and the sensitivity, specificity and repeatability of this real-time PCR method were evaluated. At the same time, clinical samples were tested using this method and compared with nested PCR method reported before. Result A real-time PCR was developed to specifically detect Tyzzer ’ s Organism, detection limit of the positive plasmids were 8 copies/ μL. This method did not cross-react with various pathogens that should not be detected in laboratory animals. The coefficient of variation of intra- and inter-batch repetition was less than 3%, indicating that the repeatability of this method was good. According to the testing result of 70 clinical samples, it was found that the positive sample detection rate of this method was higher than that of the nested PCR method. Conclusion The real-time PCR method established in this study has good specificity, high sensitivity and good reproducibility, and was suitable for the detection of Tyzzer ’ s organism in rabbit fecal samples.
    Establishment of Adult Cardiomyocyte-specific Oga Knockout Mouse Model using CRISPR / Cas9 System

    Objective Constructing an adult cardiomyocyte-specific Oga knockout mouse model to study the function of endogenous OGA in the maintenance of adult cardiac homeostasis. Method Firstly, Screening the effective sgRNAs targeting the Oga coding region and and constructed them in AAV9 virus. Then AAV9 was injected into cardiomyocyte specific SpCas9 knockin mice ( α-MHCCas9) by intraperitoneal injection. qPCR and Western blot were performed to confirm whether Oga was successfully knockout. Finally, to study the function of Oga in cardiac homeostasis maintenance, the cardiac structure was analyzed by histology and the cardiac function was analyzed by transthoracic echocardiography. Result Two effective sgRNAs targeting Oga were identified and delivered by AAV9 to disrupt Oga gene successfully in cardiomyocytes. Consistently, O-GlcNAcylation level was significantly increased by Oga deletion. There were no significant differences in overall structure and function indicated by the histological and echocardiographic analyses. Conclusion By using the AAV9-mediated delivery of CRISPR / Cas9 system, we successfully constructed the adult cardiomyocyte- specific Oga knockout mouse model, which would be provided as an effective mouse model for studying the function and mechanism of OGA-mediated O-GlcNAcylation clearance in the maintenance of cardiac homeostasis.
    Analysis on Changes of Related Indexes in Different Periods of Hyperlipidemia of Rat Model

    Objective To explore the level of blood lipid and oxidative stress in different periods of hyperlipemia of rat model, and to scientifically analyze its changing regularity. Method SD rats were randomly divided into control group ( normal diet and drinking water ) and treatment group ( normal drinking water, high-fat feed: basic feed +15% lard +20% sucrose +1. 2% cholesterol +0. 2% sodium cholate) . Each group was divided into five sub-groups: 0, 15, 30, 60 and 90 d. The general state of animals was observed and recorded. Animals were treated at different time, serum was taken, blood lipid level and oxidative stress level were detected, liver was weighed, liver coefficient was calculated, some livers were placed in 4% paraformaldehyde, HE histopathological staining was performed, some livers were homogenized, and blood lipid level was detected. Result In the treatment group, the fur of the animals was dull, and there were depilation and slow movement. The weight increased significantly from 15 d, and the liver coefficient increased significantly from 30 d. Compared with the control group, the difference was statistically significant ( P < 0. 05 ) . TG and CHO in serum of treatment group were increased significantly on 30 d and 60 d, and the difference was statistically significant (P<0. 05) , but the difference of TG on 90 d was not statistically significant compared with that of control group. From the 15 d of treatment group, MDA in serum increased and SOD decreased significantly, and the difference was statistically significant ( P < 0. 05 ) . T-AOC decreased significantly from the 30th day, and the difference was statistically significant (P<0. 05) . From the 15 d, lymphocytes were gathered, liver cells were swelled and degenerated, nuclei were squeezed to one side. Until the 90 d, liver cords were disordered, liver cells were swelled, fatty degeneration occurred, and vacuoles with different sizes in cytoplasm were compressed and deformed. Conclusion The modeling rate of hyperlipemia animal model was high, but in the research and analysis of drug prevention and treatment, the change of metabolism and the length of time should be considered. Besides serum indexes, it was suggested to increase the detection indexes of tissue and other metabolic enzyme activities, so as to analyze the effects of drugs more scientifically.
    Experimental Study on the Repair of Bone Defect with a New Type of Allogeneic Bone

    Objective To explore the effect of new type of allograft bone in repairing bone defect. Method Male New Zealand rabbits were used to construct the animal model of bone injury, and 60 rabbits were randomly divided into experimental group, control group and blank control group. At the outer edge of proximal femur and 1 cm lower edge of greater trochanter, the bone defect with a depth of about 1 mm was prepared by precision grinding mechanism with a diameter of 4 mm ball head. Do the same for the contralateral femur. In the experimental group and control group, the left femoral defect was filled with cortical bone particles, and the right femoral defect was filled with cancellous bone fragments, and the material was filled with light pressure to make the material close to the hole as much as possible. Layered suture. Animals in the blank group underwent the same operation as described above without any material filling. Imaging ( X-ray) and histopathological examination were performed at 1, 4, 8 and 12 weeks after operation. Result One week after operation, there was no difference in bone defect repair among the three groups. At 4 weeks, 8 weeks and 12 weeks after operation, the bone tissue repair speed of the tested group was significantly better than that of the control group. Conclusion This type of allograft bone can stimulate the differentiation of mesenchymal cells into osteoblasts, promote the formation of osteocytes and the maturation of callus, and is an ideal bone graft material.

    The number of laboratory animals used in teaching and scientific research in medical colleges is increasing, and the waste generated is also increasing, which poses a certain hidden danger to public health safety. This article first expounded the sources, hazards and safe disposal method of laboratory animal waste, including gas, water, litter, animal carcasses and other wastes, and then analyzed the problems and deficiencies in the disposal and management in medical colleges. Finally, suggestions for the management of laboratory animal waste have been put forward in terms of perfecting laws, regulations and supervision system, strengthening publicity and training, improving disposal technology, increasing funding investment, improving working conditions, and implementing safety protection measures.

    Objective The traditional method of collecting arterial blood from mouse heart puncture has many problems, such as high difficulty and low success rate. An improved method is proposed under the existing conditions to ameliorate the above problems. Method Forty male Balb / c mice aged 8 weeks were randomly divided into the traditional method group and modified method group. Arterial blood collection and blood gas analysis were performed by percutaneous left ventricular puncture and left ventricular puncture through muscle layer after skin stripping, respectively. The puncture success rate, maximum blood volume and blood gas analysis of the two groups were counted, and the result were compared and analyzed. Result The puncture success rate of the traditional method was 40%, and the maximum blood collection volume was ( 0. 41 ± 0. 13) mL. The puncture success rate of the modified method was 90%, and the maximum blood collection volume was (0. 51±0. 10) mL. The result showed that the success rate and maximum blood volume of the modified method were significantly higher than those of the traditional method, and the differences were statistically significant (P<0. 01, P<0. 05) . In terms of blood gas analysis, the pH of the traditional method group was 7. 08 ± 0. 12, and that of the modified method group was 7. 09 ± 0. 13 ( t = - 0. 17, P = 0. 87) . The PCO2 of the traditional method group was 35. 60±6. 02, and the PCO2 of the improved method group was 32. 33±4. 09 ( t = 1. 27, P = 0. 22) . The oxygen partial pressure ( PO2 ) of the traditional method group was 90. 13±10. 23, and that of the modified method group was 97. 13 ± 14. 76 ( t = - 1. 10, P = 0. 29) . The blood oxygen saturation ( SO2 ) of the traditional method group was 87. 00 ± 5. 29, and that of the modified method group was 91. 63 ± 4. 41 ( t = - 1. 90, P = 0. 78 ) . The above blood gas analysis result showed no statistical difference. Conclusion The modified method of collecting arterial blood from mouse heart puncture is simple and visualized, with high success rate and good repeatability.

    Rodent animal is an important model for the study of mechanism and drug of depression because it can better simulate a series of biochemical changes of human depression. Behavioral experiments are widely used to evaluate animal models and drug efficacy of neuropsychiatric diseases such as depression. However, the animal behavior evaluation method related to depression are complex, each has its own advantages and disadvantages, and there is no unified operation standard or evaluation standard. Therefore, this paper aims to summarize the experimental principles and operational method of depression-related behavioral evaluation in rodents, and review the advantages and disadvantages as well as the improved method, so as to provide a basis for establishing stable and reliable behavioral evaluation standards for depression in animals.
