    Establishment and Application of RT-PCR Method for Detection Pneumonia Virus of Mice in Infection Laboratory Animals and Interna- tional Comparison Samples

    Objective To establish a RT-PCR method for detection of Pneumonia virus of mice ( PVM ) in laboratory animals and related samples. Method The primers were designed and synthesized by selecting the conserved G gene sequence of PVM, and the RT-PCR method was established. And the specificity, sensitivity,repeatability and stability of the established method were verified. The established method was used to detect the lung tissue samples of 27 mice,9 rats, 5 gerbils submitted for routine examination and 19 PVM infected mice, and 8 ICLAS international comparison samples. Result The established method had no cross reaction with Sendai virus( SV) , Reovirus type 3 ( Reo3) , ( HV) and Lymphocytic Choriomeningitis Virus ( LCMV) ; The method could detect the minimum template concentration of PVM DNA was 8. 77×102 copies / μL, and could detect the minimum titer of virus was 10- 4. 0 / mL. The primers and PVM plasmids stored at -30 ℃ refrigerator for 12 months were used for PCR detection, and the target fragment of about 249 bp could still be amplified. The lung tissue samples test result of 27 mice, 9 rats and 5 gerbils submitted for routine examination were all negative. The result showed that 7 lung tissue samples of the 19 mice infected with PVM were positive. Among the 8 ICLAS international comparison samples tested, 1 sample was positive for PVM nucleic acid, which was in line with the expected result.Conclusion The established PVM RT-PCR method is good in specificity, sensitivity, repeatability and stability,and can be used for the monitoring of mice, rats, gerbils and other experimental animals and the detection of related samples.

    Establishment and Application of Cardiomyocyte-specific Snhg5 Transgenic Mice

    Objective To establish a transgenic mouse with cardiomyocyte-specific overexpression of Snhg5.Method A construct that directed the expression of Snhg5 to cardiomyocytes was made. The linearized constructs were introduced into the fertilized eggs by microinjection to obtain the transgenic mice. The integration of Snhg5 gene was detected by PCR. The tissue-specific expression of Snhg5 was confirmed by Real-time PCR. Cardiac phenotypic analysis in control and transgenic mice were performed at the gross morphological, histological and molecular levels. Result A transgenic vector that specifically overexpressing Snhg5 in cardiomyocytes was successfully constructed, and 4 Snhg5 transgenic founder mice were successfully obtained by microinjection of fertilized eggs. Detecting the expression level of Snhg5 in different tissues showed that Snhg5 was upregulated specifically in cardiac tissue. Further phenotypic analysis demonstrated that cardiomyocyte-specific Snhg5 transgenic mice were more prone to pathological cardiac remodeling under isoproterenol ( ISO ) stimulation compared to control mice. Conclusion A cardiomyocyte-specific Snhg5 transgenic mouse was generated successfully, providing an ideal mouse model for studying the function of Snhg5 in cardiac tissue and the mechanism underlying the pathological cardiac remodeling.

    Abstract:Objective To investigate the effects of eEF1A2 deletion on contents and composition in mice skeletal muscle. Method Twelve-month-old eef1a2fl / fl;CreERT2+ mice ( iHBKO) and their control eef1a2fl / fl;CreERT2- mice ( Control) were intraperitoneally injected with 30 mg / kg tamoxifen for three consecutive days, respectively, and the efficiency of eEF1A2 knockout in skeletal muscle was verified using real-time qPCR and Western blot, the skeletal muscle morphology was detected by HE staining, and the expression of the fiber type markers were measured by real-time qPCR. Result eEF1A2 was knocked out in skeletal muscle in iHBKO mice. The gastrocnemius muscle organ coefficient decreased significantly in knockout mice. However, the organ coefficients in other skeletal muscle and the gastrocnemius muscle morphology did not alter. eEF1A2 knockout also resulted in a significant increase of the slow muscle marker myosin heavy chain 7 ( Myh7) expression and a significant decrease of the fast muscle marker myosin heavy chain 4 ( Myh4) expression in the gastrocnemius and extensor digitorum longus muscle, while Myh4 and Myh7 both decreased in the slow muscle soleus and tibialis anterior muscle. Conclusion eEF1A2 deletion reduced the gastrocnemius composition, and converted fast to slow muscle fibers in mice.

    Abstract: Objective Whether skin presensitization is necessary in experimental colitis model induced by trinitrobenzenesulfonic acid ( TNBS ) has not been determined, and the effect of skin sensitization on it is investigated. Method BALB / c mice were randomly divided into normal control group ( NC group ) , skin presensitization group ( SP group) , and non-skin presensitization group ( NP group) , 5 per group. The success of the two groups of models and the Luminex multifunctional liquid chip analysis system was used to evaluate the differential expression levels of BAFF / BLys / TNFSF13BTNF-α、 ICAM-1 / CD54 and TIMP-1 inflammatory cytokines in serum. Result Compared with the NC group, there was no significant difference in body weight between SP group and NP group from skin presensitization to before enema; After enema, the body weight of SP group was significantly lower than NP group ( P<0. 05 ) . Compared with the NC group, the spleen weight ( P<0. 01) and spleen index ( P<0. 05) of the NP group were significantly reduced; the scores of disease activity index in SP group and NP group were significantly higher than the NC group ( P <0. 05) ; the colon macroscopic damage index ( CMDI) in the SP group increased significantly ( P<0. 05) ; the SP group had a significant increase in histopathological score ( P<0. 05) . Serological detection of inflammatory cytokines, the result of BAFF / BLys /TNFSF13B showed that compared with the NC group, the SP group was increased significantly ( P<0. 01) , but there was no significant difference between the SP group and the NP group. Conclusion Both the SP group and the NP group can successfully establish model of acute experimental colitis, but the benefit of acute experimental colitis established by skin presensitization has a better modeling effect.

    Effect of Hypobaric Hypoxic Exposure on Intestinal Microbiome Composition and Community Structure in Mice

    Abstract: Objective To analyze the intestinal microbiome composition and community structure of normal and hypobaric hypoxic environment in mice. Method The 6-week old SPF C57BL / 6 N male mice were randomly divided into 2 groups with 6 mice in each group: the normal atmospheric pressure group ( alt. 20 m to 60 m, oxygen concentration 21%) as the control test group ( the CT Group) , the hypobaric hypoxic group ( alt. 5 000 m, oxygen concentration 10%, the HYP group ) . After 4 weeks, the fresh feces of the mice were collected to investigated by 16 s sequencing analysis. Result The relative abundance of Bacteroidetes and Verrucomicrobia in the feces of mice in the HYP group was higher than that in the CT group, the relative abundance of Firmicutes and Proteobacteria was lower than the CT group. The heat map result show an increase in the OTUs of the Ruminococcus, Enterococcus, Akkermansia, Prevotellaceae UCG-001, Bacteroides, Alistipes, Parabacteroides and a decrease in Lactobacillus, Allobaculum, Ruminiclostridium and Roseburia in the HYP group. Compared with the CT group, the dominant flora is Akkermansia ( 20. 60%) of Verrucomicrobia. Conclusion The composition and community structure of intestinal microbiome is affected by the simulated conditions of altitude environment which may be related to the hypobaric hypoxic.

    Abstract: Objecive To observe the effects of XiaoChaihu KeLi combined with Levofloxacin on Escherichia coli ( E. coli) result on antibacterial and antidiarrhea in vivo. Method The seventy Kunming mice (weighing (20±2) g, sex in half) were divided into seven groups, and different concentrations of E. coli bacterial were intraperitoneal injection, which to determine the concentration of bacterial suspension of minimum lethal dose ( MLD) . The fifty Kunming mice were divided into five groups, which were model group, normal saline group, levofloxacin group, Xiaochaihu granule group and combined treatment group ( levofloxacin and Xiaochaihu granule ) . The bacterial suspension concentration of minimum lethal dose ( MLD) in mice was determined. In the in vivo antibacterial test, the bacterial diarrhea model of mice was established by intraperitoneal injection of E. coli suspension ( MLD concentration) . After 15 days of intragastric administration of corresponding drugs, the protective effect of combined drugs on lethal E. coli infection in mice was explored. The effects of combined administration on diarrhea index and organs of mice infected with E. coli and on intestinal propulsion rate of mice were explored after 7 days of gavage of corresponding drugs. Result The combined drug had a good protective effect on diarrhea induced by E. coli infection in mice, and the survival rate was 90%, which was statistically significant compared with the levofloxacin group and the Xiaochaihu granule group ( P<0. 05) . Combined drug group significantly improved the diarrhea symptoms of mice, compared with model group and Xiaochaihu granules group, diarrhea index of combined drug group had significant statistical significance ( P<0. 01) . Compared with the levofloxacin group, the diarrhea index of the combined treatment group was statistically significant ( P <0. 05) . The combined drug group protected the organs of mice and reduced the effect of diarrhea caused by E. coli infection on the toxicity of the organs, it could also improve the immune organ coefficient and improve the body immunity. Compared with model group, blank group, levofloxacin group and Xiaochaihu granule group, the thymus organ coefficient and spleen organ coefficient of combined medication were statistically significant ( P<0. 05) . Compared with levofloxacin group and model group, the liver organ coefficient and kidney organ coefficient in the combined treatment group were statistically significant ( P < 0. 05 ) , compared with blank group, were nonstatistically significant ( P > 0. 05 ) . Combined drug improved intestinal motor function of diarrhea mice caused by E. coli infection. Compared with model group and blank group, small intestinal propulsion rate had significant statistical significance ( P < 0. 01) , Compared with levofloxacin group and Xiaochaihu granule group, there were statistical significance ( P < 0. 05) . Conclusion The combination of Chinese and Western medicine have a synergistic effect, improve the antibacterial and anti-inflammatory effect, reduce the use of antibiotics, at the same time, it can play the immunity-enhancing effect of Xiaochaihu KeLi, improve the body’ s resistance to foreign evil, improve the survival rate. The result of this study provide experimental basis for the clinical combination of bacterial diarrhea.

    Abstract: Objective To study the protective effect of miR-208b on myocardial infarction mice. Method Construction of a mouse model of myocardial infarction. The mice were randomly divided into the model group ( MI group) , the empty body group ( Ad group without miR-208b antagomir ) , the experimental group ( miR-208b antagomir group with miR-208b antagomir) . The mice without ligation of anterior descending branch were selected as the sham operation group ( sham group) . Echocardiography was used to detect cardiac function, the expression of miR-208b was measured by qRT-PCR, the area of myocardial infarction was measured by TTC staining, the apoptosis of myocardial cells in the periphery of infarction was detected by TUNEL, and the expression of Bcl-2 and Bax in myocardial cells was detected by Western-blot. Result After 7 days of myocardial infarction, the expression of miR-208b in the MI group and the Ad group was significantly increased compared with the Sham group ( P<0. 05) ; The infarct area of the miR-208b antagomir group was significantly smaller than that of the MI group ( P<0. 05) ; The levels of LVEDd and LVESD in the MI group, the Ad group and the miR-208b antagomir group were significantly higher than those in the Sham group, while LVEF and FS were significantly lower ( P<0. 05) ; The levels of LVEDd and LVESD in the miR-208b antagomir group were significantly lower than those in the MI group, while LVEF and FS were significantly higher ( P<0. 05) ; The apoptosis rate of myocardial cells in the MI group, the Ad group and the miR-208b antigomir group was significantly higher than that in the Sham group ( P<0. 05) ; The apoptosis rate of myocardial cells in the miR-208b antigomir group of was significantly lower than that in the MI group( P<0. 05) ; The result of Western blot showed that the heart cells in the group of MI and Ad were significantly lower than those in the group of MI ( P<0. 05) . The expression of Bcl-2 and Bax in myocardial infarction mice was significantly down regulated ( P < 0. 05) . The expression of Bcl-2 in the miR-208b antiagor group was significantly up regulated ( P < 0. 05 ) and Bax was significantly down regulated ( P < 0. 05 ) . Conclusion miR-208b is highly expressed in myocardial infarction mice. Inhibiting the expression of miR-208b can reduce the area of myocardial infarction, inhibit the apoptosis of myocardial cells, and improve the cardiac function of mice, so as to play the role of heart protection.

    Comparative Study on Hyperlipidemia Model of SD Rats and KM Mice Induced by Fat Emulsion

    Abstract:Objective A comparative study of hyperlipidemia models induced by fat emulsion in SD rats and KM mice was conducted in order to provide reference for the study of the pathogenesis of hyperlipidemia and the basis for the selection of animal disease models for the development of lipid-lowering drugs. Method The hyperlipidemia model was induced by gavage of fatty emulsion for 2 weeks and 4 weeks, and the four blood lipids ( TG, TC, LDLC, HDL-C) , liver fat ( TG, TC) and liver tissue lesions were detected. Result Compared with the normal group, the fat emulsion of KM mice was intragastrically administered for 2 weeks, and there was no obvious change in blood lipids. After 4 weeks of intragastric administration, the liver tissues showed mild pathological damage, but blood lipids and liver lipids remained unchanged. SD rat fat emulsion was given to the stomach for 2 weeks, serum TC and LDL-C were significantly increased (P<0. 001, P<0. 01) , HDL-C was significantly reduced (P<0. 001) , and TG had no significant change; fat emulsion was given to the stomach 4 In week, rat serum TG, TC, LDL-C were significantly increased (P<0. 001, P<0. 01) , HDL-C was significantly reduced ( P < 0. 001) , and the liver was obviously enlarged, white in color, and liver tissue lesions. The degree is heavier, with obvious fat vacuoles. Conclusion The hyperlipidemia model of SD rats induced by fat emulsion gavage for 4 weeks was better.

    Abstract:Objective Through the verification of the detection ability of Yersinia enterocolitica in laboratory animals, we can understand the testing ability of laboratory animal testing institutions and improve the testing level of laboratory animals. Method According to the proficiency testing scheme approved by China National Accreditation Service for Conformity Assessment ( CNAS ) , the samples were prepared by freeze-drying at low temperature; after passing the stability and homogeneity test, they were randomly grouped and numbered through the CNAS platform and issued to participating units. A work instruction is attached. The inspection report and original records are required to be submitted within the allotted time. Those whose result are consistent with the sample setting shall be regarded as satisfactory result, and those inconsistent or not submitted on time shall be regarded as unsatisfactory result. Result There were 24 laboratories which joined to this test plan. Among them 22 laboratories (91.7%) achieved satisfactory test result, and two laboratories (8.3%) with unsatisfactory test result. Conclusion Most of experimental animal quality testing laboratories in China have reliable detection capability for Y.enterocolitica.

    Study on the Amount of Corncob Bedding in IVC System of SPF Rats and Mice

    Abstract:Objective Explore the reasonable usage of corncob bedding for SPF rats and mice in standard IVC system. Method Under the upper limit condition of feeding density required by national standard, the corncob bedding for mouse and rat cages was used in different amounts according to the low, medium and high groups. The weight, feed intake, and fecal weight of rats and mice were recorded every 3 d in a period of 21 d, and the cleanliness of the bedding was scored. Result There were no statistically significant differences in body weight, feed intake, and fecal weight among the three groups of rats and mice within 21 days. Mouse litter cleanliness was significantly different from the high litter group after the 12 days ( P < 0. 001) , and the middle litter group was significantly different from the high litter group after the 9th day ( P < 0. 05 ) . There was not any statistically significant difference in the cleanliness of bedding materials between the three groups in the rat. Conclusion Under the standard IVC system, when the cage change cycle was 7 days, the amount of corncob bedding used for mouse cages is 100 g, and the amount of corncob bedding used for rat cages is 400 g. As the cage change cycle prolongs, the amount of corncob bedding used needs to be increased accordingly.

    Abstract:Objective In order to provide a basis for the selection and application of the model of post-stroke depression in rats, the differences between the two most commonly used method for the preparation of the model of post-stroke depression in rats were compared by systematic evaluation. Method We searched Chinese and English databases including VIP, Wanfang, CNKI, Sinomed, PubMed, WOS, EMBASE on line, and collected all literatures about the post-stroke depression model in rats. Time limit was set since the establishment of the database to August 22nd, 2020. Literature screening and data extraction were conducted by two researchers independently. The quality of literatures was evaluated using the Systematic Review Centre for Laboratory animal Experimentation ( SYRCLE) animal experiment bias risk assessment tool, and reticulation meta-analysis was performed by Stata10. 0 software. Result A total of 998 rats were included in 51 studies. Among them, 809 rats received chronic unpredictable stress ( CUMS) in 42 studies after cerebral ischemia. While the chronic binding stress ( CRS) were carried out in of the rest of 9 studies including 199 rats. In terms of body weight, the availability of the two models was followed as CRS and CUMS, both of them showed significant weight loss since post-stress 14 d to 28 d ( P <0. 05) . However, for the sucrose preference rate, CUMS was superior to CRS, the former displayed significant decrease from post-stress 7 d ( P < 0. 05) , while the latter began at post-stress 14 d. Furthermore, CUMS model showed a significant difference from post-stress 7 d in the open field test ( P < 0. 05) , while CRS model appeared after 14 d of intervention ( P < 0. 05 ) . Conclusion The present study showed that rats received CUMS after cerebral ischemia was easier to produce depression-like behavior than the intervention of CRS.

Animal welfare not only affects the behavior of laboratory animals, but is also related with the accuracy of animal experiment. The article mainly expounds the behaviors of laboratory animals such as foraging behavior, territorial behavior, aggressive behavior, defensive behavior, reproductive behavior, nursing behavior, rhythm behavior, social behavior, communication behavior and corresponding welfare demand. And we analyze the essential points of guaranteeing animal welfare as well as impacting to the behavior of laboratory animals of environment change and animal welfare change. In this paper, we investigate the question of animal welfare from the behavior of laboratory animals point,hoping to provide some reference for the guarantee of animal welfare.

    Summary and Analysis of Histopathological Surveillance of Rodent Experimental Animals in Beijing in 2020

    Abstract:Objective Two times a year, through random inspection the rodents the pathological changes of experimental animals in Beijing, on the one hand, the overall quality of Beijing rodent experimental animals levels to make scientific evaluation, discover the problems existing in the quality of experimental animals, thus provide a reference for the production management and improvement of the experimental animals, on the other hand, enhance the consciousness of the experimental animal quality management and safety, security and maintenance of laboratory animal production stability. Method The heart, liver, spleen, lung, kidney, rectal and intestine of rodents were collected and fixed with formaldehyde calcium solution. The tissues and organs of rodents were evaluated by HE staining, oil-red O staining and PAS staining. Result Hepatocyte swelling was found in a few cases in rats, mice, guinea pigs and hamster. In mice and rats, the white marrow of spleen decreased and the extramedullary hematopoiesis increased. Very few mice showed epicarditis, epicardial calcification, rectal parasitic infection, enteritis and nephritis. Guinea pigs presented with pneumonia, hepatic ductal epithelial hyperplasia and intestinal bleeding. Conclusion Most of the experimental animals submitted for testing are generally healthy; SPF mice and rats need to be screened for causes of decreased white pulp in the spleen in order to improve and ensure animal quality. The pathological changes of guinea pigs mainly occurred in the lungs of ordinary grade guinea pigs, so it was speculated that the breeding environment of ordinary grade guinea pigs should be improved. Detection of pneumonia in SPF grade guinea pigs also requires identification of the cause.

    Abstract:Objective To explore an optimized method for making paraffin sections of whole eyeballs in rats.Method 18 SD rats were divided into three groups, with 6 in each group. Eyeballs from all three groups were enucleated and fixed in 4% paraformaldehyde, 10% neutral formalin, and a mixed fixative solution, respectively. 2 hours later, a small hole was punctured with a syringe at the cornea of the left rat eye of all groups. Meanwhile a slice of about 0. 5 cm in length was cut longitudinally at the sclera of the right eye with a sharp blade. The rat eyes were then put back into the corresponding fixatives for 24 hours. The eyeballs were subjected for the whole eyeball slicing and the hematoxylin-eosin staining. Observation of the effect of eyeball tissue section was taken under microscope. Result The eyeballs fixed with 4% paraformaldehyde and 10% neutral formalin were shrank and deformed, with decreased structure of lens. The sign of the retinal detachment was found and the continuity of each nuclear layer in the retina was poor. The overall structure of the eyeball fixed in the mixed solution was relatively complete, with no shrinkage. No obvious retinal detachment occurred, and the continuity of each nuclear layer was better. Compared with the left eye, the lens structure of the right eye was more complete. Conclusion The paraffin slice of the whole rat eyeball with a longitudinal scleral section fixed in the mixed solution was relatively complete in structure. The method was simple, and could be applied in a certain range.

    Abstract:human enterovirus 71 ( EV71 ) is the causative agent associated with hand foot and mouth disease ( HFMD) , and be found to be neurotropic. The worldwide spread of EV71 has drawn increasing attention from researchers and medical professionals. Animal model is an important means to study infectious diseases, at present many result have been obtained on EV71 susceptible animal models. In this study we summarized the current research on EV71 animal models and focus on the future research direction.

Gut microbiota and its metabolites have become an important target for the prevention and treatment of atherosclerosis. Animal models play an important role in studying gut microbiota and its effects on host physiology and disease. This paper reviewed the latest progress of gut microbiota in animal models of atherosclerosis and clinical patients.
