Abstract: Objective To investigate the technical level of rat Sendai virus antibody detection for lab animal testing laboratories. To explore and solve the existing problems in Sendai virus detection,so as to promote quality management of laboratories and improve the detection level. Method To prepare rat serum samples after calibration and test their stability and homogeneity according to CNAS. The samples were numbered randomly and distributed to the participating laboratories. Each laboratory received three samples and must send results to us in a specified time. The results were expressed as positive or negative. If the test results were consistent with expected results,the laboratory was judged satisfactory. If the results were not entirely consistent or feedback was not got on schedule,the laboratory was judged unsatisfactory. Result Twenty-eight laboratories from 18 provinces enrolled in the program and sent the results in a specified time. The results of 25 laboratories were all satisfactory,which were 89. 3% of all laboratories. 3 laboratories were not satisfactory,one of which has one sample which result did not match and the other 2 laboratories have 2 samples which did not match. 27 of 28 laboratories adopted ELISA and one lab used IFA. The original records were authentic but individual labortories have some problems,such as reagent is not clear,operation process is not detailed and positive results were not retested. Conclusion The Sendai virus detection ability level of animal test laboratories is high. The implementation of proficiency testing can reflect the detection level of laboratories.

Abstract: Objective To study the effects of Curcumin on vital signs and related blood indexes of heatstroke rats in dry-heat environment of desert, so as to elucidated the possible mechanisms of the protective effects of Curcumin.Method The 200 male SPF grade SD rats,6 ~ 8 week-old,were randomly divided into control group,solvent group,low-dose Curcumin pretreated group,middle-dose Curcumin pretreated group and high-dose Curcumin pretreated group,40 rats for each group. Each group were intragastrically administrated for 7 consecutive days.Then all the rats were put into the dry heat environment ( temperature of 41℃,humidity 10%) in The Simulated Climate Cabin for Special Environment of Northwest of China. Each group were randomly selected 10 rats to measure core body temperature and weight at the time of 0 min,50 min,100 min,and 150 min. At each time point,the rats of were caught out of the cabin and then were be anesthezied,and arterial and venous blood were collected to detect blood gas, blood routines and biochemical indicators. Result The average core body temperature rising rate of each 10 min were significantly lower in the Curcumin high dose pretreatment group compared with the control group and the solvent group,was significantly different ( P < 0. 05) . The average weight falling rate of each 10 min were were significantly slower in the Curcumin high dose pretreatment group compared with the control group and the solvent group ( P < 0. 05) . The average weight falling rate of each 10 min in the high-dose Curcumin pretreated group was negatively correlated with pO2 ,pH,Glucose and Triglyceride ( r =- 0. 955、- 0. 355,- 0. 692,- 0. 733) ,and positively correlated with the number of white blood cells ( r = 0. 943) . Conclusion The average weight falling rate and the average core body temperature rising rate may be the useful indicator for the prognosis of the heatstroke rats; Curcumin may maintain higher level of blood pO2,blood glucose and triglyceride,slow down the falling speed of blood pH,and decrease the blood leucocyte numbers through reducing the average weight falling rate and the average core body temperature rising rate of heatstroke rats in dry heat environment of desert,hence maintain good internal environment,so as to exerted its protective effects on the heatstroke rats in the dry heat environment of desert.

Abstract: Objective To observe the effect of Curcumin pretreatment on serum and urine electrolytes in desert environment heat stroke rats. Method One hundred and forty male SD rats,200 ~ 250 g,were randomly divided into two groups: heat stroke group at 0h and heat stroke group after 24 h,each groups were made of seven subgroups,with 10 rats in each subgroup: normal sodium chloride control group,mild heat stroke sodium chloride group,moderate heat stroke sodium chloride group and severe heat stroke sodium chloride group,mild heat stroke Curcumin group,moderate heat stroke Curcumin group and severe heat stroke Curcumin group. The conditions of normal temperature group were at 25 ℃,humidity 35% RH,the condition of heat stroke group were 41 ℃,humidity 10%RH. A number of rats in the heat stroke zero hour were collected the inferior vena cava blood for serum electrolyte determination,another group of rats were collected the urine and inferior vena cava blood 24 hours after heat stroke. The serum and urine electrolyte was measured. Result With the deepening of the heat stroke,serum K + gradually decreased,Na + ,Cl - ,Ca2 + had an increasing trend,but did not reach a significant level,after Curcumin intervention,serum K + was retain to normal gradually ,while serum Cl - ,Ca2 + ,Na + were significantly increased. 24 h after heat stroke,serum K + in mild and moderate heat stroke groups returned to normal levels,while in severe heat stroke group,serum K + concentration was still very low. After Curcumin intervention,the serum K + level of the mild and moderate heat stroke rats increased significantly,and was significantly higher than that of the normal level,while the serum K + of the severe heat stroke rats recovered to normal level. And Na + ,Cl - ,Ca2 + ,24 h after heat stroke ,were restored to normal levels. As well as Curcumin was concerned,the concentrations of K+ ,Na + ,Cl-were lower in urine 24 h after heat stroke,and urine Cl-was increased significantly. Conclusion Curcumin can enhance serum K + concentration and promote urine Cldischarge to correct heat stroke rats electrolyte disturbances in heat-dry desert environment.

Abstract: Objective An inoculum of 1 × 107 cfu /mL non-pathogenic Escherichia coli-Mkate2 was injected directly into the bilateral renal cortex of the mouse to build the acute pyelonephritis model in normal environment.Method Check the result of model quality with imaging instrument,the method had improved the success of acute pyelonephritis model,and reduce the number of animals used in laboratory tests. Result It had built the acute pyelonephritis model,the E. coli could not survive in the kidney to reduce the biological risk for the people and environment.

Abstract: Objective The purpose of the present studies was to clone and sequence eukaryotic protein elongation factor 1a2 ( Eef1a2) gene of Mongolian gerbils,and analyze homology of Eef1a2 between four species. Method The Eef1a2 cDNA was cloned from skeletal muscle mRNA of Mongolian gerbils by homologous amplification and rapid amplification of cDNA ends ( RACE) . Then the homology of nucleotide and amino acid sequences of Eef1a2 were analyzed among gerbils,human,rats and mice by DANSTAR software. Result The cDNA length of Mongolian gerbils Eef1a2 was 1812 bp,coding sequence ( CDs) 1392 bp,coding 463 amino acids. It exhibited 88. 6% identity with human Eef1a2,94. 0% identity with rat Eef1a2,and 93. 8% identity with mouse Eef1a2.And Eef1a2 of Mongolian gerbils had 100%,99. 9% and 99. 9% amino acid sequence identity to human,rats and mice,respectively. Conclusion Both nucleotide and amino acid sequences of Eef1a2 from Mongolian gerbils are high identity with human Eef1a2. Therefore,Mongolian gerbils are expected to be one of the ideal models for studies of human Eef1a2.

Abstract: To investigate the influence of primary culture on physiochemical properties of normal hepatic tissues and hepatic tumor tissues of adult mice. The hepatic tumor tissues ( T) and tumor adjacent tissues ( P) of 9 month-aged H-ras12V transgenic mice and normal hepatic tissues ( Wt) of 9 month-aged C57 /BL6J mice were collected to perform the in vitro primary culture. The morphological changes of cultured hepatocytes were detected by hematoxylin-eosin staining during 12 days. The whole-protein variation tendency of cultured hepatic tissues were detected by bradford staining during 48 h. The protein expression levels of p-ERK,p-AKT,p-mTOR,Cyclin D1,and GAPDH of cultured hepatic tissues were detected by western blot during 48 h. The mRNA expression levels of Cyclin D1,GAPDH,UQCRB,and 18s RNA of cultured hepatic tissues were detected by RT-qPCR during 48 h.The survival condition of primary cultured hepatocytes of Wt,P,and T were fine during 6 days and the morphological changes were resemble. After 6 days,the cell number of hepatocytes of Wt and P decreased gradually. However,hepatocytes of T dead quickly and appeared a few clone-like cell masses. The whole-protein variation tendencies in Wt,P,and T were resemble. The abundance of protein bands in 130 ~ 170 kμ increased first and then decreased during 48 h. The abundance of protein bands in 40 ~ 130 kμ aincreased during 48 h. The protein expression levels of p-ERK,p-AKT,p-mTOR,Cyclin D1,and GAPDH in primary cultured hepatic tissues decreased gradually during 48 h. The mRNA expression levels of Cyclin D1 in hepatic tissues of Wt,P,T and GAPDH in Wt,P increased during 48 h. The mRNA expression levels of GAPDH in hepatic tissues of T and UQCRB in Wt,P,T decreased first and then increased during 48 h. There is no regularity variation tendency for the expression levels of 18s RNA. During the short-term primary culture for hepatic tissues and hepatic tumor tissues of adult mice,although morphology changes of hepatocytes are not obviously different,the mRNA and protein expression levels of many geneschange significantly. These changes will give great influences on the result of primary culture related experiments.

Abstract: Objective By sampling for cosmetics sold in some cities in Hebei,the situation of safety toxicological of the cosmetics was investigated. Method Using the animal experiments method,the 33 cosmetics were tested on toxicology. The cosmetic samples collected from markets,they were the beauty cream,sunscreen,facial cleanser and body wash and so on. Result Nineteen cosmetics ( 8 of body wash,8 of facial cleanser and 3 of beauty cream) in rabbits acute skin irritation test that resulted for non-irritating 10 cases ( 52. 6%) ,9 cases of mild irritant ( 47. 4%) ; 16 samples ( 8 of body wash and 8 of facial cleanser) in rabbits eye irritation test resulted for 0 cases of non-irritating reaction,micro stimulation 3 cases ( 18. 8%) ,13 cases of mild stimulation ( 81. 2%) ; The results of 17 samples ( 3 of beauty cream,9 of Sunscreens and 5 of Freckle cream) in the guinea pig skin allergy tests displayed that the weakly and mild allergenic reaction were total of 3 cases ( 17. 6%) ,and no allergic reactions in 14 cases ( 82. 4 %) . Conclusion Survey results show that all kinds of cosmetics have different proportions of light ( or micro) of the toxicological response. Although it can be determined that the products under investigation are acceptable for the human,but suggested that the regulatory authorities should attach importance to and strengthen risk monitoring,in order to protect the safety of consumers.

Abstract: Objective To analyze and compare the hematological and serum biochemical status between adult rhesus monkeys( Macaca mulatta) and aged rhesus monkeys ( Macaca mulatta) . Method Blood samples were obtained from 20 healthy adult rhesus monkeys and 12 aged healthy rhesus monkeys( half male and half female) .All the samples were tested in the automatic blood cellular analyzer,automatic blood biochemical analyzer and automatic blood gas analyzer. Result Compared with the adult rhesus monkeys,hematologic values of the aged monkeys such as MCHC( P < 0. 01) ,HCG,PLT,RDW-SD( P < 0. 05) ,were significantly higher,while other hematologic values such as LYMPH#,EO# ( P < 0. 01) ,WBC( P < 0. 05) ,were significantly lower. In serum biochemistry test,values of the aged rhesus monkeys such as MAO,TRIG( P < 0. 01) ,ALT,GLU,LDH( P < 0. 05) ,were significantly higher than the correspond value of the adult rhesus monkeys,while other values such as SOD( P < 0. 01) were significantly lower. In artery blood gas analysis,compared with the adult rhesus monkeys,the aged monkeys' Lac values were significantly higher( P < 0. 01) ,while values such as PaCO2,HCO3 + ,TCO2,BE were lower ( P < 0. 01) . Conclusion There are some significantly differences in hematological and serum biochemical values between adult and aged rhesus monkeys,and we should take this into full consideration during biochemical or medical research.

Abstract: Objective To study the Qingxuetongmai granule on improving microcirculation and antithrombus actions. Method The model of microcirculatory hindrance was made by dripping epinephrine in mesentery in mice,and examination indexes include capillaries diameter,micro-flow velocity and flow state were detected.Electrical stimulation proximal cephalic artery is liable to injury the internal wall of vessels,thereby leading to blood flow,and ultimately thrombosis. The effect of fighting clotting by Qingxuetongmai granule was measured.Result Qingxuetongmai granule can improve capillaries diameter,micro-flow velocity and flow state,decrease blood clot blocking rate in rats. Conclusion Qingxuetongmai granule has the effect of improving microcirculation and antithrombus in rats.

Abstract: Objective To study the influence on the weight of weaned mice by high fat food and forced sports,and the behavioral difference between dietary induced obesity ( DIO) and dietary induced obesity-resistant ( DIO-R) .Method Fifty weaned mice were randomly divided into control group ( n = 60) and high-fat group ( n = 50) ,and fed normal and D12492 high fat diet respectively for 75 d. D12492 group was screened into obesity ( DIO) and obesity-resistant mice ( DIO-R) according to body weight,and then DIO ( n = 20) mice were divided into forced sports and control group ( n = 10) . The mice in forced sports group were forced to have sports for an hour every day. The total cholesterol ( CHO) and triglyceride( TG) were measured by Hitachi automatic biochemical analyzer and activity were measured by roller recorder. Result There was significant difference for TC and TG between high fat food and normal food; After forced sports,the fat male mice reduce the fat more than female. Conclusion High fat food can induce the mice's obesity; forced sport can reduce the body weight in male animal,female mice with more voluntary wheel running were inclined to obesity resistant.

Abstract: The clear grade Sprague Dawley rat was purified by cesarean birth and a gnotobiotic population was established by breeding,it will meet requirement of scientific research units,the 35 offspring were obtained by cesarean birth in isolation environment. Two hours later,survival 20 offspring were feed the modified milk by intragastrical gavages and self-designed equipment. The feeding rate was 5 ~ 6 times a day. the 13 offspring were successful wean. These gnotobiotic animals were verified by detecting excrement padding and cotton swab,the gnotobiotic rat model was successful established by germ-free technology and modified milk,which could be conserved by breeding. It solves the problem of shortage of gnotobiotic rat.

Abstract: Acute lung injury ( ALI) and acute respiratory distress syndrome ( ARDS) are common clinical syndrome with high mortality rate,and with no exactly effective treatments,which have not been researched thoroughly at present. Endotoxin / Lipopolysaccharide( LPS) has been reported as an important risk factor of ALI /ARDS. The studies of ALI are mainly based on the animals,that its species and the responses to the inducer would always affect the trial.
