    Effects of BAY and DMSO on Acrosome Reaction in vitro in Mice Sperm

    Abstract: Objective The effects of BAY and DMSO on acrosome reaction in vitro in mice spermatozoa were investigated. Method The influence of both chemicals on AR in vitro in mice by using 6 different concentrations of DMSO and 5 different concentrations of BAY for 5 duration time were invesgigated. The AR was evaluated by FITC-PSA and Coomassie brilliant blue stain. Result Both sperm AR rate and survival rate varied with could be affected by DMSO within a certain concentration and time,while the BAY had litter influence on them with all measured concentration ( P > 0. 05) . Conclusion The results showed that the BAY and DMSO had different pattern on sperms. DMSO has a direct effect on both the AR rate and survival rate,while the influence of BAY on the AR rate and the survival rate is neglectable. Thus a proper usage of DMSO in manipulation of sperm should be carefully reconsidered.

    Abstract: Objective To study the inhibitory effects of Huaweishu granule on mice transplanted tumors. Method The S180-mice model was used to observe the tumor-inhibition rate of Huaweishu granule and the H22-mice model was used to test the survival time. To observe the effect of Huaweishu granule with combination of CTX on WBC and tumor weight of S180-mice model. Result Huaweishu granule had the inhibitory effects on tumor cell growth of S180-mice and the inhibitory ratio of Huaweishu granule in S180- mice was 30% above; Huaweishu granule combined with CTX could reduce the number of WBC in S180 tumor-bearing mice and improve the antitumor effects of CTX. Conclusion Huaweishu granule has potent inhibitory effect on the growth of implanted S180 and H22 in mice and has synergetic effect and attenuated toxic effect in combination with CTX.

    Abstract: Objective Standardized determination of immune function indicators and effect of cyclophosphamide on the H22 solid tumors in mice. Method H22 cells were implanted into the left axilla of 46 BALB /c mice. Tumors growth was observed 1 days later,The mice were randomly divided into tumor-bearing mice + cyclophosphamide group,tumor-bearing mice group,control mice + cyclophosphamide group,control mice group. Cyclophosphamide group intraperitoneal injection of cyclophosphamide 50mg /kg,2 times a week. Observe the general clinical manifestations,tumorigenicity,body weight,food intake,tumor volume,tumor weight,immune organ weight, blood values,blood biochemistry values,cytokines,lymphocyte transformation,NK cell activity,lymphocyte subsets and histopathology. Result Compared with normal mice,H22 tumor-bearing mice reduced food consumption,RBC,HGB,HCT significantly decreased ( P < 0. 01) ,WBC,PLT,RDW significantly increased ( P < 0. 01) ,GOT,GPT significantlyincreased ( P < 0. 01) ,CD4 - CD8 - significantly decreased ( P < 0. 01) . The average food intake reduces the tumor-bearing mice given cyclophosphamide,D9 to D14 body weight was significantly lower ( P < 0. 05) ,D5 to D14 tumor volume was significantly lower ( P < 0. 01) ,tumor weight,spleen weight,thymus weight was significantly sexual lowering ( P < 0. 01) ,the WBC,MCV significantly with lowering ( P < 0. 01 ) ,HGB,the MCH,MCHC,the PLT significantly with increased ( P < 0. 05 ) ,the GOT,GPT significantly ( P < 0. 01) lower of IL - 2 significantly with reducing ( P < 0. 01) ,IFN - γ significantly increased ( P < 0. 01 ) ,CD4 + CD8 - ( %) ,CD4 + CD8 + ( %) ,CD4 - CD8 + ( %) ,CD3 + ( %) lymphocytes significantly with increased ( P < 0. 01) ,CD4 - CD8 - ( %) lymphocytes was significantly lower ( P < 0. 01) ,spleen lymphocytes transformed absorbance difference significantly lower ( P < 0. 05) ,NK cell activity significantly reduce ( P < 0. 05) . Conclusion Blood parameters and immune function indicators were measured in H22 tumorbearing mice. We provide the basic data for biological response modifiers anticancer drug screening.

    Abstract: Objective To observe the effect of chloradurat,propofol,ketamine on hematology in mice. Method Mice were anaesthetized by intraperitoneal injection of chloradurat,propofol,and ketamine respectively. Result Serum biochemical parameters and blood cell indexes of anesthetized mice showed significant difference in comparison with normal mice. Conclusion Anaesthesia can affect the hematology considerably.

    Abstract: Objective To observe the effect of Kv1. 2、Kv3. 4 channels in rat pulmonary arterial smooth cells on the process of pulmonary vasoconstriction induced by 15 - KETE. Method Western blot and PCR were used to identify the effect of 15 - KETE on expression of Kv1. 2、Kv3. 4 channels in cultured rat pulmonary arterial smooth muscle cells ( PASMCs) . Result ( 1 ) The expressions of Kv1. 2 mRNA and protein in rat PASMCs were significantly down-regulated by 15 - KETE. ( 2) The expressions of Kv3. 4 mRNA and protein in rat PASMCs were significantly down-regulated by 15 - KETE. Conclusion Kv1. 2、Kv3. 4 channels are involved in vasoconstriction induced by 15 - KETE in rat PASMCs.

    Abstract: The length and diameter of underfur and guard hair of Wistar-Imamachi rat population of 2 to 16 week-old were measured respectively. The 4 measurement indexes increase rapidly form 2 to 3 week-old and increase slowly from 3 to 4 week-old,the length and diameter of underfur and the length of guard hair increase slow remarkably after 5 week-old,but the diameter of guard hair increases rapidly again form 6 to 7 week-old, the growth rate slows down after 7 week-old and lasts to the old age.

    Abstract: Objective To study the efficacy of Xiaozhangmingmu capsules on the preventing and treating of cataracts induced by D-galactose. Method The model of D-galactose-induced cataract in rats was established by means of intraperitoneal injection and oral administration of D-galactose. The opacities of eye lens were compared and analyzed with the slit lamp. Simultaneously,the contents of SOD,MDA and GSH-Px in lenses were assayed. Result Xiaozhangmingmu capsules could delay the advance of cataract,decrease significantly the content of MDA,Increase the activity of SOD and GSH-Px. Conclusion Xiaozhangmingmu capsules can delay the appearance and lessen the extent of cataracts induced by D-galactose.

    Abstract: Objective To study the preparation and identification of the model of rabbit knee osteoarthritis with the improved operational method of Hulth. Method According to the improved operational method of Hulth,rabbits were cut off knee medial collateral ligament,anterior cruciate ligament,and medial meniscus. After 8 weeks, cartilage and synovium were observed by X ray photograph,naked eye and HE staining. The expression of IL - 1、 TNF - α in serum and joint fluids were detected by Enzyme-linked Immunosorbent Assay ( ELRSA) . Result X ray showed that obvious arthrostenosis and spur were developed. Cartilage defect and synovial inflammation were observed by naked eye. HE staining showed that cartilage defected,chondrocyte reduced and apoptosis. Compared with normal group,There were higher expression of IL - 1、TNF - αin serum and joint fluids in model group( P < 0. 05) . Conclusion After 8 weeks,the model of rabbit knee osteoarthritis with the improved operational method of Hulth was successfully made. And the model can accurately reflect the characteristic of osteoarthritis through the image analysis and biochemistry detection. It will be the theoretical basis and research groundwork for clinical treatment.

    Abstract: Objective To explore the feasibility of disinfection using an ozone generator in the animal lab equipped with individual ventilated cages ( IVC) . Method Use ozone testing tube to test the ozone concentration in the animal lab and the working IVC,observe health condition,biochemical index,histology changes of the windpipe and lungs in rats. Result The low,medium and high filters on the IVC cage have a certain barrier function to prevent ozone,and can reduce 80% of ozone. Under no special protection,the rats have a high resistance to the ozone. In the 35 m3 IVC animal lab,using an ozone generator with 3 g /h ozone production for 35 minutes exposure,there are no detectable physiological and pathological changes. Conclusion Ozone generator can be used for environment disinfection in IVC animal laboratory.

    Abstract: Objective Establish mouse superovulation and embryo transfer technology,in order to further use the technology to control their offspring,and gradually will these technologies applied to other animals,so as to speed up its breed improvement,improve the utilization rate of good varieties and individual. Method Of 4-week-old C57BL /6 female mouse by intraperitoneal injection 7. 5 IU PMSG ( pregnant mare serum gonadotropin) 48 hours after intraperitoneal injection of superovulation 7. 5 IU HCG ( human chorionic gonadotropin) and C57BL /6 male rats mated,see bolt 0. 5 days,1. 5 days and 3. 5 days,remove the corresponding embryo,the resulting embryoscreening,elect better quality embryos and embryonic stages ( single cells,2 cells capsule fallopian tube or uterus KM pseudopregnant mice mouse embryo) into estrus synchronization see bolt after KM ligation male rats mating stimulus be in production pregnancy after transplantation in mouse. Result Superovulation 88 C57BL /6 mice,see bolt 62,a total of 510 single-cell embryos,400 2-cell stage embryos,blastocysts 35,the ministry of tubal transplantation,tubal notches,uterine transplant transplant pseudopregnant female mice 43,pregnancy producing 39 farrowing 361. Conclusion Tubal fimbria transplantation compared with tubal Jiankou transplant was no significant difference tubal transplantation hurt small animals,but for patients who require a higher; fallopian tube the Jiankou transplant operation is relatively quick,but the larger animal injury; uterine transplant the pregnancy best effect at this stage of embryo easily implantation.

    Abstract: Objective Study the advantage of venous indwelling needle in applications of clinical animal experiment. Method Use indwelling needle in clinical animal experiment and evaluate the puncture effects and results. Result In animal experiment,venous puncture with indwelling needle have high efficiency,alleviate animal pain,reduce animal dysphoria and inconvenient of experiment caused by punctures repeatedly. Meantime,it can accelerate experiment progress,reduce the work of experimenter. Venous puncture with indwelling needle is in conformity with the ethical requirement of experiment animal and animal welfare law. With the development of medical and health services and the exploration of staff in clinical practice,venous puncture with indwelling needle can be used extensively in practical work of health care.
