Abstract: Objective In order to provide basic data for the application of MERS-CoV studies, biochemical parameters of transgenic mice expressing human dipeptidyl peptidase 4 were examined. Method A transgenic mouse model that expressing hDPP4, named C57BL / 6J-TgN ( UBC-hDPP4- GFP) CLARS was constructed, which was referred to Tg^hDPP4. Ten blood biochemical indexes, 18 blood physiological indexes, growth and development indexes from 4 to 16 weeks of age and 8 organ indexes of Tg^hDPP4 mice were measured, and compared with wild-type C57BL / 6J mice. Result The alanine aminotransferase and alkaline phosphatase of female Tg^hDPP4 F3 mice was higher than wild-type C57BL / 6J mice. The alanine aminotransferase and alkaline phosphatase of male transgenic mice were lower than the wild-type C57BL / 6J mice. Elevated blood glucose and blood urea ammonia were found in male Tg^hDPP4 F3 mice. Conclusion Through the analysis of the blood biochemical parameters of Tg^hDPP4 F3 mice, it is found that the exogenous hDPP4 gene can affect the blood sugar, blood urea ammonia, alanine aminotransferase and alkaline phosphatase in transgenic mice and may affect the average body weight of Tg^hDPP4 mice.

摘要： 目的 分析表达人二肽基肽酶 4( hDPP4)的转基因小鼠的生物学特性指标,为将其应用于中东呼吸综合征的相关研究提供基础数据。 方法 本实验室构建了表达 hDPP4 的转基因小鼠 C57BL / 6J-TgN ( UBC-hDPP4-GFP )CLARS,简称 Tg^hDPP4。 对不同性别(每组 5 只)的 Tg^hDPP4 小鼠的 10 项血液生化指标、18 项血液生理指标、4 ~ 16 周龄生长发育指标和 8 项脏器指数进行测定,并与不同性别(每组 5 只)的野生型 C57BL / 6J 小鼠进行比较,用统计学方法进行分析。 结果 与野生型 C57BL / 6J 小鼠相比,Tg^hDPP4F3 雄鼠的丙氨酸转氨酶和碱性磷酸酶均降低,血糖和血尿氨素均升高;与野生型 C57BL / 6J 小鼠相比,Tg^hDPP4F3 雌鼠的丙氨酸转氨酶和碱性磷酸酶均升高;Tg^hDPP4F3 雄鼠的体质量自 5 周龄开始均高于野生型雄鼠。 结论 hDPP4 的转入对 Tg^hDPP4 小鼠的血糖、血尿氨素、丙氨酸转氨酶、碱性磷酸酶等部分血液生化指标和体质量均产生影响。
