建立快速检测禽白血病病毒的液相芯片方法

Rapidly Detecting Avian Leucosis Virus through Establishing Luminex Liquid Chip Technology

摘要: 摘要: 目的 建立快速检测禽白血病病毒( Avian Leukosis Virus,ALV) 的液相芯片方法,为大量样品的检测提供技术支撑。方法 制备抗 ALV P27 单克隆抗体,生物素标记抗体,磁珠偶联捕获抗体,利用 Luminex200 分析系统验证该方法的特异性、敏感性及重复性。结果 阳性判定标准定为大于或等于空白对照的两倍。特异性检测结果显示与鸡的其他多种病原不存在交叉反应,特异性良好。同时多批次实验结果显示其变异系数在 7% 以内,说明该方法的稳定性好,可重复。结论 建立的检测 ALV 病原的液相芯片方法特异性好,稳定且可重复。

Abstract: Abstract: Objective To establish the liquid phase chipmethod of rapidly detecting avian leucosis virus,and provide technical basis for the detection of large quantities of samples. Method Prepare matching monoclonal antibody against P27 protein of avian leucosis virus,biotin labelling antibody,magnetic beads coupling capture antibody. this method is validated with Luminex200 analysis system to test specificity,sensitivity and repeatability.Result The positive criterion is more than twice as much as the blank control. The specificity test result showed that there was no cross-reaction between many other pathogens of the chicken,meaning good specificity. Simultaneously repetitive experiment shows the coefficient of variation is within 7% ,which indicates that the stability of the experiment is good and repeatable. Conclusion The Luminex liquid chip technology,which directly detects the ALV pathogen,is specific,stable and repeatable.
