大黄牡丹汤组方对急性胰腺炎模型大鼠炎症反应及氧化应激水平的影响

The Effect of Dahuang Mudan Decoction on Inflammatory Response and Oxidative Stress in Acute Pancreatitis Model Rats

摘要: 摘要: 目的 研究大黄牡丹汤组方( Rhei radix et rhizome peony Decoction Prescription,RPDP) 对急 性胰腺炎( AP) 模 型大鼠炎症反应及氧化应激水平的影响。方法 SD 大鼠分为假手术组、模型组和治疗组。以 0. 1 mL/min 的速度, 经十二指肠壁浆肌层穿刺距离胆胰管开口 0. 3 mm 处的胆管,加压注射 3. 5% 牛磺胆酸钠 0. 1 mL/100 g,建立大鼠 AP 模型。治疗组造模后立即给予 RPDP 40 g /kg 灌胃 1 次,假手术组、模型组给予等量 的生理盐水灌胃 1 次。各 组分别于造模后 4h、8h、12h 处死 10 只大鼠并取血,检测血清中 IL-6、IL-β 和 TNF-α 含量; 同时切取各时段大鼠胰 腺组织,检测 MDA、MPO 和 SOD 的含量。结果 与模型组相比,治疗组各时段血 清 IL-6、IL-β 和 TNF-α 含量均显 著减少( P < 0. 05) ; 各时段胰腺组织中 MDA、MPO 含量显著降低( P < 0. 05) ,SOD 含量显著升高( P < 0. 05) 。结论 RPDP 有抑制 TNF-α、IL-6、IL-β 的释放,减少 MDA 产生,有抗过氧 化损伤作用,提高机体对 OFR 的清除能力。

Abstract: Abstract: Objective To study the effect of Dahuang Mudan Decoction ( RPDP) on inflammatory response and oxidative stress in acute pancreatitis( AP) model rats. Method Divided 90 SD rats randomly into three groups: the sham operation( SO) group ,the AP group,and the RPDP group. The models of AP were made by pressurize inject 3% sodium taurocholate into the bile duct where was 0.3mm away from the pancreatic duct with the speed of 0. 1ml /min. The rats in the RPDP group were lavaged with RPDP ( 40 g /kg) after made AP model. The rats in the SO group and the AP group were lavaged with partes aequales Sodium Chloride. Sacrifice the rats 4 h,8 h,12 h hours after establish the animal model,separated the serum and detected the content of IL-6,IL-β and TNF-α in the serum,cut pancreatic tissue in every time quantum and detected the content of MDA,MPO and SOD. Result Compared with the AP group,the serum content of IL-6,IL-β and TNF-α were significantly decrease in RPDP group( P < 0. 05) ,the content of MDA and MPO in pancreatic tissue in every time quantum were also decrease in RPDP group( P < 0. 05) ,while the content of SOD were significantly improve( P < 0. 05) . Conclusion Dahuang Mudan Decoction could repress the release of TNF-α,IL-6 and IL-β,could decrease the product of MDA,and had the effect of anti-oxidative damage,cleaning oxygen free radicals.
