    Analysis of Main Biological Characteristics of Several Commercial Immunodeficient Mice in China

    Objective To clarify the biological characteristics of commercialized immunodeficient mice in China and provide a scientific basis for their standardized management and quality control.Methods Fourteen strains of immunodeficient mice from seven domestic enterprises were selected, which were classified into three categories, T cell-deficient, T and B cell-deficient, and T, B and NK cell-deficient. Their organ coefficients, hematological indicators, peripheral blood lymphocyte subsets, and histological characteristics of immune organs were detected and compared with those of age-matched BALB/c mice. Results All three types of immunodeficient mice exhibited phenotypic characteristics corresponding to the deficiency of target immune cells. The thymus coefficients were significantly lower than those of control group, lymphocyte-related indicators (LYMPH, LYMPH%, CD3+ T cells, etc.) were significantly downregulated, and indicators such as neutrophil percentage (NEUT%) were significantly upregulated. However, statistical differences were observed in some hematological and lymphocyte subset indicators among mice of the same strain from different enterprises (P<0.05, P<0.01).Conclusion The overall quality of commercialized immunodeficient mice in China is stable, and their biological characteristics are basically consistent with those of immunodeficient mice. However, significant variations in biological indicators within certain strains deviate greatly from the average, indicating that standardization needs to be further improved.
    Analysis of Gut Microbiota Characteristics in Retired Aged C57BL/6 Mice

    Objective To investigate the characteristics of gut microbiota in retired aged C57BL/6 mice. Methods Rectal content samples were collected from female C57BL/6 mice at 8 weeks of age (control group,n=10) and retired mice at 72 weeks of age (aged group,n=10). The V3-V4 hypervariable region of the 16S rRNA gene was amplified using universal primers and sequenced via high-throughput sequencing technology. Microbial community composition and diversity indices were analyzed using QIIME software, while linear discriminant analysis effect size (LEfSe) was employed to identify statistically significant microbial biomarkers between the two groups.Results A total of 1 618 804 high quality 16S rRNA sequences were obtained from the gut microbiota samples of 20 C57BL/6 mice. At the phylum level, Bacteroidetes and Firmicutes were the dominant taxa with relatively high abundance in both  groups. Alpha diversity analysis revealed no significant differences between the two groups in any of the five indices measured: Observed features, Chao1, Shannon, Simpson, and Good's coverage. However, beta diversity analysis indicated a statistically significant separation in microbial community structure between the aged group and the control group (P<0.05). LEfSe analysis revealed that bacteria from the class Coriobacteriia, order Coriobacteriales, family Eggerthellaceae, and genus Enterorhabdus were identified as significant biomarkers in the gut microbiota of the aged group mice. In contrast, bacteria belonging to the order of Clostridia-UCG-014, family Rikenellaceae, and genus Alistipes were significantly enriched in the gut microbiota of the control group mice.Conclusion This study characterized the composition and abundance of the gut microbiota in rectal contents of retired aged C57BL/6 mice using 16S rRNA high-throughput sequencing. These findings provide fundamental data for understanding the biological characteristics of retired aged C57BL/6 mice and offer valuable reference information for the selection of appropriate animal models in aging-related research.
    Test on the Sterilization Efficacy of a Terminal Sterilization Equipment for Biosafety Laboratories Based on Formaldehyde Generation

    Objective To test and evaluate the sterilization efficacy of a self-developed formaldehyde generating terminal sterilization equipment for biosafety laboratories, and to provide scientific support for its application and promotion.Methods Using Geobacillus stearothermophilus or Bacillus subtilis var. niger spores as biological indicators, the single-factor variable method was applied to systematically investigate the effects of three key parameters—paraformaldehyde dosage, sterilization duration, and ambient relative humidity—on the sterilization efficacy of isolators in biosafety laboratories. Results Complete sterilization was achieved with paraformaldehyde dosages of 3, 1.5, and 0.75 g/m3 for 6 h of sterilization. With a fixed dosage of 0.75 g/m3, valid sterilization was attained after 2, 4, and 6 h of exposure. Under the fixed dosage and duration, qualified sterilization efficacy was obtained at relative humidity levels of 70%, 75%, and 80%.Conclusion The self-developed formaldehyde generating terminal sterilization equipment exhibits excellent and reliable sterilization efficacy for isolators in biosafety laboratories under optimized parameters, and can be used for terminal disinfection and sterilization in biosafety laboratories. 

    Effect of Shenqi Shiyiwei Granules (SQS) on Cancer-Related Fatigue in CT26 Colorectal Cancer Mice

    Objective To investigate the effect of Shenqi Shiyiwei Granules (SQS) on cancer-related fatigue (CRF) in CT26 colorectal cancer mice.Methods Thirty BALB/c mice were randomly divided into a blank group, model group, and SQS group [1.56 g/(kg·d) ],10 mice per group. Except for the blank group, the other two groups were subcutaneously inoculated with CT26 cells in the right hind limb to establish a tumor model. After successful modeling, the treatment group received SQS by gavage for three weeks. Body weight and tumor volume were monitored weekly. Exercise endurance and spontaneous activity were assessed using the forced swimming test and open field test. Liver and gastrocnemius glycogen content, serum urea nitrogen (BUN), lactate (LA) levels, and lactate dehydrogenase (LDH) activity were measured by colorimetric assays. Complete blood counts were analyzed by using an automated hematology analyzer. Spleen and tumor weights were recorded. Pathological changes in gastrocnemius muscle and epididymal adipose tissue were observed by HE staining.Results Compared with the model group, mice in the SQS group showed significantly increased body weight, prolonged load bearing swimming time, and increased total distance traveled in the open field test, while tumor volume and weight were significantly reduced, the differences were statistically significant (P<0.05). Liver and gastrocnemius glycogen levels were higher, while serum BUN, LA, and LDH were significantly lower (P<0.05). Blood parameters, including red blood cells, hemoglobin, platelet count and white blood cells, were significantly improved, and the differences were statistically significant (P<0.05). Histopathological analysis revealed attenuated gastrocnemius muscle atrophy and enlarged epididymal adipocytes in the SQS group. Conclusion SQS alleviates CRF by improving energy metabolism, reducing tissue damage, and regulating hematological parameters.
    Validation of Sterilization Efficacy of 75% Ethanol Against Staphylococcus aureus within 30 d after Opening

    Objective To validate the time-dependent changes in the sterilization efficacy of 75% ethanol after opening and to determine its safe use period to ensure disinfection effectiveness.Methods After opening, 75% ethanol was transferred into a spray bottle and placed in a laboratory animal barrier facility at room temperature. A quantitative suspension bactericidal test was performed using Staphylococcus aureus as the test strain to evaluate the bactericidal efficacy of 75% ethanol at different time intervals (0 30 d) after opening.Results The quantitative suspension bactericidal test demonstrated no significant decline in the bactericidal efficacy of 75% ethanol against Staphylococcus aureus after 30 d of room temperature storage. The log reduction value (LRV) remained above 5.0 throughout the testing period. Conclusion After being transferred to spray bottle and stored at room temperature, 75% ethanol shows no significant decline in its bactericidal efficacy against Staphylococcus aureus within 30 d, which can serve as evidence for extending its service life.
     Flow Field Analysis in Aseptic Breeding Isolators Based on Computational Fluid Dynamics

    Objective To analyze the internal flow field of a sterile isolator using computational fluid dynamics (CFD) and investigate the influence of different air inlet and exhaust vent distributions on the isolator’s internal flow field.Methods The k-ω turbulence model was used to numerically simulate a sterile isolator for housing mice and to compare the flow field characteristics under two configurations: one with vents at the same height on both sides of the rear wall, and another with vents diagonally distributed on the left and right side walls.Results Regardless of the vent distribution pattern, the flow field in the isolator was non-uniform, and the complexity of the flow field increased significantly after the placement of mouse cages. When the inlet and exhaust vents were diagonally distributed on the left and right side walls and the inlet was located above, the local air velocity in the mouse cage below the inlet reached 0.227 m/s, which was notably higher than in the configuration with vents at the same height on the rear wall.Conclusion This study provides a theoretical basis for the design optimization of sterile isolators and animal husbandry management. It is recommended to avoid placing mouse cages in high-velocity areas in practical applications to ensure consistent animal housing conditions.

    Objective To discuss application of multimodal MRI combined with inflammatory factors in evaluating the morphological changes of the sciatic nerve of diabetic rabbits.Methods Sixty eight New Zealand white rabbits were divided into normal control group (n=8) and model group (n=60) by a random number table. The model included the diabetes (DM) group (n=20) and diabetes peripheral neuropathy (DPN) group (n=40). The multimodal MRI findings, microcirculation factors (VEGF), tumor necrosis factor-α (TNF-α) and C-reactive protein (CRP) of three groups were compared.Results The levels of VEGF, TNF-α, CRP of DM group and DPN group were significantly higher than that in the control group, while the levels of VEGF, TNF-α and CRP of the DPN group were significantly higher than that in the DM group (P<0.05). The cross-sectional area of nerves in both the DM and DPN groups were significantly higher than that in the control group, and the cross-sectional area of nerves in the DPN group was significantly higher than DM group (P<0.05). The conduction velocity of neural action potentials of DM group and DPN group were significantly lower than control group, and the conduction velocity of neural action potentials of DPN group was significantly lower than the DM group (P<0.05). The three-dimensional arterial spin labeling (3D-ASL), diffusion tensor imaging (DTI) and magnetic resonance functional imaging non-invasive quantitative evaluation indicators of cerebral blood flow (CBF) and anisotropy score (FA) of DM group and DPN group were significantly higher than those in the normal control group, with statistical significance (P<0.05). The non-invasive quantitative evaluation indicators CBF and FA values of 3D-ASL and DTI magnetic resonance functional imaging of DPN group were significantly higher than those in DM group, with statistical significance (P<0.05). The combined detection of multimodal MRI with VEGF, TNF-α and CRP had an area under the curve (AUC) value of 0.964, sensitivity of 95.00% and specificity of 95.00%, which were higher than the detection of CBF value, FA value, VEGF, TNF-α and CRP alone.Conclusion The dynamic changes of serum VEGF, TNF-α and CRP levels in diabetes rabbits, combined with the changes of 3D-ASL and DTI functional magnetic resonance imaging parameters, can effectively reflect the stage evolution of sciatic nerve cell morphology. This multimodal non-invasive quantitative evaluation strategy provides objective and reproducible imaging evidence for early identification and staging diagnosis of sciatic nerve lesions in DPN rabbit models.
     Experimental Study on the Role of Tetramethylpyrazine in Promoting Vascular Endothelial and Neural Repair Following Spinal Cord Injury in Rats via the miR-497-5p/EGFL7 Axis

    Objective To explore the protective effect of tetramethylpyrazine (TMP) on vascular endothelium and nerves in rats with spinal cord injury (SCI), and to analyze its mechanism of action in the treatment of SCI based on the miR-497-5p/EGFL7 axis.Methods A total of 78 healthy male SD rats were randomly grouped into 13 groups, with 6 rats in each group. The SCI rat model was constructed by the modified Allen method, and then TMP intervention was continuously given for 2 weeks. The expression levels of miR-497-5p and epidermal growth-like protein 7 (EGFL7) were detected by RT qPCR and Western blot. Meanwhile, Basso, Beattie and Bresnahan (BBB) scoring, footprint analysis,  HE staining, terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining, enzyme linked immunosorbent assay (ELISA), dual-luciferase reporter assay and in vivo gene regulation experiments were employed. A comprehensive evaluation was conducted on indicators such as motor function, spinal cord tissue injury, apoptosis, inflammatory response and angiogenesis in rats.Results After SCI,the expression of miR-497-5p in the spinal cord tissue of rats was significantly increased, and the expression of EGFL7 was significantly decreased. At the same time, it was accompanied by typical manifestations such as motor dysfunction, tissue structure destruction, increased apoptosis and elevated levels of inflammatory factors. After TMP intervention, the above-mentioned abnormal changes were significantly improved. Specifically, the expression of miR-497-5p was down-regulated, the expression of EGFL7 was up-regulated, the BBB score and footprint analysis indicators were improved, the degree of pathological damage and apoptosis was reduced, and the expression of platelet endothelial cell adhesion molecule-1 (CD31) was increased. The levels of tumor necrosis factor -α (TNF-α), transforming growth factor-β1 (TGF-β1), and interleukin-1 β (IL-1β) decreased.Conclusion TMP improves the local microenvironment following SCI by regulating the miR-497-5p/EGFL7 axis, thereby promoting angiogenesis and neural repair. Downregulation of miR-497-5p or upregulation of EGFL7 enhances its protective effects, whereas their opposite regulation attenuates these effects.

    Objective To investigate the prevalence of murine norovirus (MNV) in commercial laboratory mice and the genomic characteristics of circulating strains.Methods Reverse transcription polymerase chain reaction (RT-PCR) was employed to detect MNV infections in commercial Kunming mice from Beijing(10 samples), Shandong(10 samples), and Liaoning (15 samples). Positive samples were subjected to whole genome sequencing analysis.Results The Kunming mice from different regions were infected with varying degrees of MNV. The whole genome sequencing and analysis of the positive sample MNV 2024BJ strain revealed a genome length of 7 382 nt, and the nucleotide homology with the representative MNV strains ranging from 88.9% to 89.9%. Structural analyses of the ORF1 cleavage site, glycosylation site, and non coding region indicated similarities between this strain and the representative strains of MNV genotype 1. Phylogenetic analysis demonstrated that this strain is closely related to domestic Guangzhou/K162/09/CHN and BJ10-2062 strains, and belongs to the same evolutionary branch.Conclusion MNV infection is common in commercial Kunming mice, with certain  differences in nucleotide homology with reported strains at home and abroad, the potential risks caused by it should be further emphasized.

    The syndrome of yang deficiency of spleen and kidney is a common syndrome in traditional Chinese medicine, which is characterized by dull pain in the abdomen, diarrhea and loose stools, or even dawn diarrhea and so on. The replication of experimental animal model is an important means in basic research, but there is a lack of unified standard for the replication of animal model of spleen and kidney yang deficiency syndrome. This paper systematically summarizes the research on the animal model of spleen and kidney yang deficiency syndrome, and makes a visual analysis of the animal model of spleen and kidney yang deficiency syndrome based on CiteSpace software to reveal its research status, hot spots and trends. It is found that in recent years, the research on the animal model of yang deficiency of spleen and kidney has gradually increased, and rats are common model animals. The method of warming the kidney and invigorating the spleen (Sishen pill) is the key treatment of spleen and kidney yang deficiency syndrome,and thecombinationof diseaseandsyndrome is the focusof research.

    Objective To investigate the effect of interferon-induced protein with tetratricopeptide repeats 1 (IFIT1) on the progression of pancreatic cancer.Methods The expression of IFIT1 in pancreatic cancer samples was analyzed using The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) datasets. The mRNA levels of IFIT1 were detected by real-time quantitative PCR in pancreatic cancer cell lines (AsPC-1, MiaPaCa-2, BxPC-3, Patu8988, Panc-1, and CFPAC-1) and normal pancreatic cells. IFIT1 knockout cells were generated using the CRISPR/Cas9 system in AsPC-1 and BxPC-3 cells. AsPC-1 cells were divided into AsPC-1-ko-NC group (negative control group) and AsPC-1-ko-IFIT1 group (IFIT1 knockout group). Similarly, BxPC-3 cells were divided into BxPC-3-ko-NC group (negative control group) and BxPC-3-ko-IFIT1 group (IFIT1 knockout group). Cell viability was assessed using the cell counting kit-8 (CCK-8) assay in the four groups. Wound healing assay was performed to observe the closure status after 24 h and to evaluate cell migration ability in the four groups. Transwell assay was used to detect cell invasion ability in the four groups. The expression levels of IFIT1, β-catenin, and EMT-related proteins (E-cadherin, N-cadherin, Vimentin) in the four groups were detected by Western blot. Subsequently, the above four groups of cells were respectively injected subcutaneously into BALB/c nude mice, the volume and weight of the transplanted tumors were measured, and the expression of proliferation markers (Ki-67) in the transplanted tumors were measured. Results Compared with normal tissues, the expression level of IFIT1 was significantly elevated in pancreatic cancer tissues. Following IFIT1 knockout in AsPC-1 and BxPC-3 cells, compared with the respective negative control groups (AsPC-1-ko-NC and BxPC-3-ko-NC groups), the AsPC-1-ko IFIT1 and BxPC-3-ko-IFIT1 groups exhibited decreased cell proliferation activity, migration, and invasion ability. In these knockout groups, E-cadherin protein expression was increased, while the expression of IFIT1, N-cadherin, Vimentin, and β-catenin was decreased. The in vivo xenograft tumor assay showed that compared with the negative control groups, the AsPC-1-ko-IFIT1 and BxPC-3-ko-IFIT1 groups had reduced tumor volume and weight, as well as decreased Ki-67 expression in the tumor tissues. Conclusion IFIT1 promotes the proliferation, migration, invasion, and in vivo tumor growth of pancreatic cancer cells by activating the Wnt/β-catenin signaling pathway. EMT is regulated by IFIT1. IFIT1 may serve as a potential therapeutic target for pancreatic cancer.
    Establishment and Evaluation of Rat Rheumatoid Arthritis Model Induced by Adjuvant Combined with Collagen Tail Injection

    Objective To establish a rheumatoid arthritis (RA) model in Wistar rats using an adjuvant combined collagen tail injection method, and to evaluate the model through pharmacological intervention. Methods Thirty male Wistar rats aged 6-8 weeks with an average body weight of (170±10) g were included in this study. Six rats were assigned to the normal control group, and the remaining 24 rats were used to establish RA models via immunization with bovine type II collagen (CII) combined with Freund’s adjuvant. Primary and booster immunizations were performed by multi-site intradermal injection of collagen-adjuvant emulsion at the base of the tail. Twenty days after the primary immunization, paw volume, paw thickness, arthritis index and body weight were measured to verify model establishment. A total of 18 rats with successful modeling were selected and randomly divided into three groups: model group, methotrexate (MTX) group and etanercept group (n=6). All animals were treated once every three days for 9 consecutive weeks. MTX was administered via intraperitoneal injection, while etanercept was given by subcutaneous injection. Rats in the normal control and model groups received an equal volume of 0.9% sodium chloride solution. During the intervention, the arthritis index and joint swelling were assessed every three weeks, and serum levels of tumor necrosis factor-α (TNF-α) and anti-CII antibodies were detected. Upon completion of intervention, the pathological conditions of ankle and knee joints in the hind limbs of rats were observed through imaging and histopathological testing, as well as the expression changes of interleukin-6 (IL-6), interleukin-17A(IL-17A), and vascular endothelial growth factor (VEGF) in synovial tissue. The therapeutic effects of the two drugs were comprehensively evaluated.Results Compared with the normal group, the model group showed significantly increased paw thickness, paw volume and arthritis index in RA rats, as well as elevated expression of IL-6, IL-17A and VEGF in synovial tissues of the hind-limb ankle and knee joints (P<0.05, P<0.01). Severe destruction of articular cartilage and massive fibrous tissue hyperplasia were observed in the joint cavity. After intervention with MTX or etanercept, paw thickness, paw volume and arthritis index of RA rats were significantly reduced. Serum anti-CII antibody titers, TNF-α levels, and the expression of IL-6, IL 17A and VEGF in synovial tissues of the hind-limb ankle and knee joints were all significantly decreased (P<0.05, P<0.01). The severity of articular cartilage destruction was alleviated, fibrous tissue hyperplasia was reduced, and symptoms including joint space narrowing, joint ankylosis, inflammatory cell infiltration, erosion and destruction were improved.Conclusion The RA model in Wistar rats induced by the modified adjuvant-combined collagen method recapitulates the pathological features of human RA and is suitable for efficacy studies of therapeutic drugs for RA.

The cleaning unit removes dust from the pipes through high-speed rotation of brushes and a negative pressure suction device. The disinfection unit uses high-concentration ozone as the disinfectant, achieving sterilization and decontamination via a closed internal circulation mode, thereby enhancing the efficiency of IVC cleaning and disinfection. The effectiveness of the device was verified by testing cleaning, disinfection, ventilation, ozone concentration, and air cleanliness. Bacterial test strips were placed in different parts of the cage rack to assess bacterial kill efficacy. Results After cleaning, dust was removed from the interior and exterior of main pipes, vertical pipes, and air nozzles. All disinfection indicator cards reached the endpoint color, confirming the disinfection effect. Bacterial test strips in the experimental group yielded negative result, while those in the control group showed positive result, indicating a bacterial kill log value >3 (kill rate >99.9%). Post-cleaning, the air velocity at the cage rack outlets and air exchange rate of cage boxes were significantly higher than before cleaning (P< 0.001). After 5 min of operation, the ozone concentration inside the disinfection hood exceeded 2 140 mg/m3, while the indoor ozone concentration was 0, the air cleanliness reached Class 7. Conclusion This device can efficiently and conveniently clean and disinfect IVC for laboratory animals, with clear cleaning and disinfection effects.

    As a cornerstone discipline of modern medical research, the quality of development in laboratory animal science directly influences the pace of innovation in medicine and related fields. In response to the lack of established frameworks for evaluating scientific and technological performance in laboratory animal institutions, this study aimed to develop a tailored evaluation system. Adopting a disciplinary ontology perspective, the research systematically examined the unique characteristics and developmental patterns of laboratory animal science. Through literature analysis and reference to the Guidelines for the Evaluation of Scientific and Technological Work in Research Institutions issued by the Evaluation Center of the Ministry of Science and Technology, a multi-tiered evaluation framework was constructed. Grounded in the discipline’s intrinsic features and developmental laws, the proposed system centers on two core dimensions—scientific research achievements and technological talent, covering six primary indicators, sixteen secondary indicators and forty-three tertiary indicators. This framework not only reflects the distinct nature of the discipline but also enables precise measurement of disciplinary contributions through differentiated indicator design, thereby offering a valuable reference for relevant authorities in China.

    Objective Isolate and identify the virus from the feces of wild tree shrews, and explore the establishment of method for isolating and cultivating tree shrew rotaviruses.Methods Fecal samples from tree shrews collected in the wild were used to amplify pathogen RNA via reverse transcription polymerase chain reaction (RT-PCR) using specific primers. The amplified products were separated by agarose gel electrophoresis, and the target bands were purified through gel extraction for subsequent analysis.Results The target fragment of 365 bp was obtained via RT-PCR. After sequencing, the gene fragment showed a nucleic acid sequence coverage of 92% and a matching consistency of 99% when compared with the VP7 segment sequence of bovine rotavirus on the Basic Local Alignment Search Tool (BLAST) in the National Center for Biotechnology Information (NCBI) database. Significant cytopathic effects (CPE) were observed upon inoculation into normal MA104 cells.Conclusion The rotavirus strain carried by the tree shrew isolated in this study has extremely high sequence homology with the rotavirus strain carried by mammalian cattle (coverage 92%, base identity 99%), which has certain reference value for the isolation and identification of tree shrew rotavirus in the future.

    Objective To analyze the nutrient components of SPF-grade rodent growth and reproduction feeds.Methods Five types of SPF-grade rodent growth and reproduction feeds were sampled and submitted for testing of conventional nutritional components indicators, amino acids indicators, vitamins indicators, as well as macro-minerals and trace minerals.Results The conventional nutrient components indicators of the five feeds complied with GB 14924.3—2010 standards. However, when compared with the reference ranges, deficiencies were identified in amino acid and vitamin indicators across five feeds. Only one feed met the GB 14924.3—2010 requirements for macro-minerals and trace minerals, while the remaining four feeds exhibited non-compliant indicators.Conclusion The nutrient components of five SPF-grade rodent growth and reproduction feeds showed varying degrees of deviation from the standards, indicating the need for enhanced management and quality monitoring during production and use links to meet the nutritional requirements of laboratory animals.

    Objective To establish a rat model of benign prostatic hyperplasia (BPH) induced by testosterone propionate in olive oil and to optimize the dosage.Methods Twenty male SD rats were stratified and randomly assigned to groups based on body weight. Fifteen rats in the model group were divided into low-dose (5 mg/kg), medium-dose (7.5 mg/kg), and high-dose (10 mg/kg) groups (n= 5), receiving daily subcutaneous injections of testosterone propionate dissolved in olive oil. The control group (n=5) received an equal volume of olive oil (0.1 mL/100 g) subcutaneously. All treatments lasted for 29 consecutive days, and body weights were recorded weekly. 24 h after the final injection, the animals were sacrificed. The prostate, seminal vesicles, and coagulating glands were harvested to calculate organ indices (wet weight/body weight ×100%). Histomorphological changes (acinar wall thickness and papillary epithelial hyperplasia) were observed by hematoxylin-eosin (HE) staining. Lesion severity was evaluated using a semi-quantitative histopathological scoring system (grades 0-4) for the accessory sex glands and quantitative image analysis of the lumen-to-stroma ratio in prostate tissue. Data were analyzed using SPSS 20.0.Results All model groups successfully developed benign prostatic hyperplasia. The pathological score in each dose group was significantly higher than that in the control group (P<0.05). The gonadal index in all dose groups was significantly higher than that in the control group, and the prostate index in the high-dose group was significantly higher than that in the low-dose group (P<0.05). The proportion of glandular lumen area in each dose group was significantly lower than that in the control group (P<0.05), and interstitial fibrosis was aggravated in a dose-dependent manner. Conclusion Under the fixed administration period (29 d) and subcutaneous injection route, testosterone propionate at doses of 5-10 mg/kg can effectively establish a BPH model, with the high-dose group exhibiting the most pronounced effects. The medium-dose group showed intermediate values for all parameters, exhibiting a dose-dependent trend.

    Objective To establish an easy-to-operate and safe rat scald model with reproducible scald depth.Methods A total of 42 healthy SPF-grade SD rats were randomly assigned into seven groups, groups A, B, C, D, E, F and control group. Rats in groups A, B and C were subjected to scald injury using a thermostatic water bath, while rats in groups D, E and F were modeled with a thermostat scald instrument, the control group underwent sham scald treatment. The ideal parameters for model creation were determined based on factors such as scald area, whether the scald depth met expectations, and survival rate of experimental animals. Additionally, serum biochemical changes induced by different scald method were assessed.Results Rats in groups A and B exhibited second-degree scalds, while those in group C visually reached a preliminary third-degree scald level. However, the scald area did not meet expectations. Rats in groups D and E showed second-degree scald, and those in group F exhibited third degree scald with well-defined scald areas. Histological examination of skin in group F rats met the required standards. Compared with control group, the ALB levels of rats in water bath group and scald instrument group were significantly decreased. Meanwhile, the ALP and AST levels in these two groups were significantly elevated, and ALP level in the water bath group was statistically higher than that in the scald instrument group (P<0.05).Conclusion The constant temperature and pressure scald device allows for precise control of scald depth and demonstrates excellent performance in controlling the scald area,resulting in less damage to experimental animals.

    Ischemic brain injury is a leading cause of mortality in adults. Two main strategies are adopted for the treatment of ischemic brain injury with neural stem cells: activating endogenous neural stem cells and transplanting exogenous neural stem cells. Endogenous neural stem cells primarily reside in specific neurogenic niches of the brain and respond to cerebral injury. A large number of preclinical studies have demonstrated that signaling pathways including Wnt, PI3K-Akt, MAPK, Notch and Eph mediate the proliferation and differentiation of endogenous neural stem cells after brain damage. Exogenous neural stem cell transplantation facilitates functional recovery following brain injury via cell replacement, neuroprotection, immunomodulation and paracrine effects. This article reviews the research progress of various signaling pathways regulating the proliferation and differentiation of endogenous neural stem cells after ischemic brain injury, and summarizes the potential therapeutic mechanisms of neural stem cell transplantation for ischemic brain injury, aiming to provide new insights for the application of neural stem cell therapy in ischemic brain injury.

    This study quantifies behavioral traits of rodents in a standardized environment and establishes an assessment system based on four classic paradigms. The open-field test measures anxiety levels and exploratory motivation through parameters like time spent in the central zone. The elevated plus maze test is based on the approach-avoidance conflict between open and enclosed spaces, with the proportion of time spent and entries made in open arms serving as the key indicators. The forced swimming test and tail suspension test assess depression-like behaviors in rats by inducing a “behavioral despair” state, using immobility time as the evaluation metric. The experiments require rigorous standardization of environmental parameters, operational procedures, and testing sequences, and different animal strains must be selected based on strain-specific baselines to mitigate genetic background interference. These methods, through a multi-level analysis of locomotor activity-spatial exploration-behavioral despair, provide quantifiable and repeatable assessment tools for investigating emotional disorder mechanisms and advancing drug development.

    In hot environments, officers and soldiers undertaking military training and operations other than war (MOOTW) are at high risk of heat stroke (HS), a severe heat-related disorder that poses grave threats to their physical health and life safety.The rat HS model is of great significance for studying the pathogenesis, preventive measures, and treatment methods of this disease. At present, there are many studies on making rat HS models, involving various aspects such as the selection of rat species and sex, modeling standards, key evaluation indicators, and modeling methods. In this paper, the above aspects are reviewed, in order to provide a reference for relevant personnel to make rat HS models.

    As a treasure of traditional Chinese medicine, acupuncture has evolved over two thousand years. Its efficacy and neurobiological mechanisms have been verified by modern research, and it is widely applied in the diagnosis and treatment of human and animal diseases. As a modernized modification of acupuncture, electroacupuncture enhances acupoint effects via electrical stimulation, further expanding the application scope of acupuncture in veterinary medicine. This paper systematically reviews the research progress of acupuncture and electroacupuncture in diseases of dogs, cats, rabbits and other animals, focusing on four core conditions: analgesia, anti-inflammation, movement disorders and gastrointestinal diseases, and elaborates on their mechanisms of action and clinical applications. Meanwhile, this paper puts forward the prospect that future research still needs to further clarify the mechanisms of action of acupuncture, optimize its treatment parameters, and improve the evidence-based medicine system, so as to promote the precise and scientific application of acupuncture techniques in the field of veterinary medicine, and provide green and efficient traditional Chinese veterinary treatment plans for animal health protection.
