    Experimental Observation On the Estrous Cycle of Female Tyrpan Meriones meri)ianus in Xin Jiang

    Objective To observe the estrous cycles of Meridian gerbils and understand the estrous regularity of Meridian gerbils. Method Twenty-four female M. meridianus vaginal epithelial cell smears were collected at intervals of 4 hours and 20 days. The morphology and quantity of conified cells of vaginal exfoliated epithelial cells were observed by hematoxylin-eosin staining. The estrous cycles of meridian gerbils were determined. Result The estrus cycle of meridian gerbils can be divided into four stages : proestrus ( 22. 0 ± 12. 0) h, estrus( 26. 8 ± 8. 1) h, metoestrus ( 18. 2 ± 9. 1 ) h and Dioestrus ( 50. 0 ± 38. 2 ) h, and eestrus cycle ( 4.7±1.2 ) d. The proportions of nucleated cellsin proestruswere ( 80.20 ±1.83 ) %.The proportions of conified cells in estrus were ( 88.00 ± 1.86 ) %.The proportions of conified cells in metoestrus were ( 24.80 ±1.43 ) %.The proportions of lymphocytes metoestrus were ( 48.20 ±2.05 ) %.The proportions of conified cells were ( 24.80 ±1.43 ) %.The proportions of lymphocytes in dioestrus were ( 91. 70±1. 66 ) %. Conclusion It provides the basis for the study of estrous cycle of female Meriones meridianus.
    The Evaluation of Proficiency Testing for Minute Virus of Mice Antibody in Laboratory Mice Detection

    Objective Through the capability verification of Minute virus of mice (MMV ) antibody in laboratory mice detection, to investigate the capacity of experimental animal quality control laboratories, so as to promote the improvement of laboratory animal quality testing level. Method According to the capability verification scheme approved by CNAS, the serums were calibrated, and passed the stability and homogeneity test. Then the serums were used as the proficiency testing samples. The samples were randomly numbered, and then sent to the units participating in the capacity verification, and attach the work instruction. This capability verification was a qualitative test, and the test method was ELISA. The participating units are required to submit the test report and copy of original record within the prescribed time limit, The satisfactory result is that the test result of the sample is consistent with the pre-test result, the unsatisfactory result is that the sample test result is inconsistent with the pre- test result or the participating units did not feedback the result. The participants must submit the test reports and original records in time. The feedback result were judged by the rate of concordance with the anticipated result. Result The result of uniformity test and stability test of samples all met the requirements of capability verification for samples. There were 33 animal testing institutions from 19 provinces and cities took part in this capability verification. The results ubmitted by 33 testing institutions are all satisfactory, and the satisfaction of this capability verification is 100%. Conclusion It can be reflected the testing ability and level of China ‘s testing laboratories for the quality control of laboratory animal by this capability verification plan , and the ability for detection of minute virus of mice antibody in laboratory mice was high .
    Pathological Diagnosis and Analysis of Bladder Tumor in Canine

    Objective Pathological diagnosis and analysis of 32 cases of canine bladder tumor were performed to investigate the pattern and pathological features of canine bladder tumor. Method Thirty-two tumors were collected from canine bladder surgery and sectioned for H.E staining. The diagnosis was based on pathological observation, and the tissue origin, age and location were analyzed. Result The statistical result showed that the most common type of bladder tumor was transitional cell carcinoma, accounting for 41%. In all cases , 59% of tumor are stem from epithelium which are the most common , with transitional cell carcinoma accounting for 68%, followed by papilloma accounting for 16%, and mesenchymal tumor accounting for 41%, the most common being hemangioma and leiomyosarcoma. The incidence of males is 78% , much higher than females. The incidence of tumors increases with age, and the probability of tumor malignancy increases significantly after 7 years of age. Conclusion The histological features of canine bladder tumors are summarized , which provides a reference for the diagnosis and differential diagnosis of tumors in this area.
    Effect of Acellular Dermis Matrix on Pigs Burning Wound Healing

    Objective To study the effect of acellular dermis matrix on pigs with deep partial thickness burn. Method Twenty-four male and female miniature pigs, weighing ( 15 ± 3 ) kg, were included. They were used to prepare the deep partial thickness burn model with the size of 4 cmX6cm by copper ingots, then randomly divided them into two groups: experimental group using a cellular dermis matrix and blank control group using absorbent cotton gauze. Detected the rate of wound healing, healing time, clinical and histopathological observation, hydroxyproline contents of wound in postoperative 1,2,3 and 4 weeks. Result During the experiment, all the animals survived , and no wound infection occurred. The wound surface of the experimental group was dried after operation, 2 weeks incrustation, 3 weeks decrustation and hair growth. In the blank control group, local exudation occurred on the wound surface at 1 weeks after operation, and the rest of the wound wasdry, incrustation at 2 weeks, decrustation and covered with hair at 4 weeks. The time of wound healing was ( 20. 83 ± 0.75 ) d in the experimental group and ( 27. 33 ± 0. 82 ) d in the blank control group. There was significant difference in wound healing rate between the two groups at 2 weeks and 3 weeks. The skin of the experimental group had clear structure at 3 weeks after surgery and the burn wound was basically healed, and after 4 weeks basic skin structure was intact. The formation of basic skin structure and append ages in the blank control group lagged by 6 days compared with the experimental group .There were significant differences in inflammatory cell infiltration and hydroxyproline contents of wound between the two groups at 1 weeks, 2 weeks and 3 weeks. Conclusion Acellular dermis matrix could effectively promote the healing of burn wound, and shorten the treatment cycle.
    Effects of Red and Blue Light on Premature Vision Failure and Neural Network in Rats

    Objective To investigate the effects of red and blue light on retinal senescence and neural network in rats. Method Fifty male Wistar rats were randomly divided into 5 groups : blue light irradiation 1 h、blue light irradiation 8 h、red light irradiation 1 h,red light irradiation 8 h and blank control group. A compulsory swimming experiment ( FSTs) was performed after 12 weeks of intervention to assess mental state. The primary culture of fresh retinal pigment epithelial cells ( RPE) of the eyeball was dissected and senescence-associated-identification of β- galactosidase ( SA-β-Gal ) activity, analysis of the association between illumination time and replicative senescence, and use of reagents box to detect nitric oxide ( NO) and nitric oxide synthase ( NOS) content in brain tissue and analyze the stress response of light stimulation to rats. Result Compared with red light irradiation , blue light irradiation 8 h significantly up-regulated the proliferation of RPE cells (P <0. 05 ) and the content of ( P< 0.01 ) . At the same time, blue light irradiation 8 h more easily caused rats to fall into a nervous and desperate mental state. Conclusion The effect of blue light on retinal senescence and neural network is greater than that of red light.
    Comparison and Improvement of Intrathecal Administration Methods in Rats

    Objective Based on experiments, an intrathecal administration method which is stable , reliable, easy, continuous and less damage was presented. Method Rats were randomly underwent catheter implantation either through atlanto-occipital membrane or foramen intervertebrale , the out erend of the catheters were either linked toa Alzet Osmotic Pump or just fixed to skin. According to the different treatment pattern , the animals were divided into 6 groups: 4 operation groups (atlanto-occipital catheterization group , atlanto-occipital catheterization with Osmotic Pump group , foramen intervertebrale catheterization group , foramen intervertebrale catheterization with Osmotic Pump group ) and 2 sham groups ( atlanto-occipital catheterization sham group , foramen intervertebrale catheterization sham group ). Death , paralysis , and sighs of developing motordys function and ill health were abserved after surgery. Rotarod motor function , paw with drawal threshold ( PWT ) and paw withdrawallatency (PWL) were also tested before and after surgery. Result Only half of the rats underwent atlanto-occipital catheterization could beused for further study , the others developed motordysfunction , ill health ( see page of eyes and nose , weight loss ) orweredead. Because of dislodging of the catheter , one-third softhe animals underwent for amen intervertebrale catheterization were excluded from further study , the other two-thirds of the rats were qualified. While all of the rats underwent foramen intervertebrale catheterization connecting to Osmotic Pump could beused for further study. Atlanto-occipital catheterization could resultin hyperalgesia and decrease of rotarod motor function.The duration of running times on rotarod , PWT and PWLwere all significantly lower than that of sham group and before-operation state. Foramen intervertebrale catheterization had no significante fecton PWT , PWL and the running time son rotarod. Conclusion Iintervertebrale catheterization combining with Osmotic Pump was a stable , reliable intrathecal adminidtration method with less damage.
    Occurrence and Analysis of Spontaneous Tumors in Sprague-Dawley Rat Background Data Acquisition Test

    Objective Sprague-Dawley rat is one of the important species in rats. To supply the reference data of spontaneous tumors of Sprague-Dawley rat for the drug preclinical safety evaluation, the types of spontaneous tumors of Sprague-Dawley rat were observed and their incidence of statistics was analyzed. Above all, the data would be so important to the long-term toxicity test, the carcinogenic test and the reproductive toxicity test. Method In the background data of acquisition test, 260 SD rats were randomly divide into eight groups ( A group, B group, C group, D group, E group, F group, G group and H group) according to weight. The animals of the above groups were dissected respectively in the planning anatomy at 2 weeks, 4 weeks, 8 weeks, 13 weeks, 26 weeks, 52 weeks, 78 weeks and 104 weeks. Result During the test, no tumor was found in the first five groups, but the tumor of the other three groups appeared notonly in the planning anatomy butalso in the urgent anatomy.The tumor was firstly discovered at 24th week of the test ( the age of the animal was 30 weeks ) among the animal necropsy and was found in total of 94 rats among which had 59 female rats and 35 male rats. A total of 185 tumors were detected, of which 81. 1% were benign tumors and 18.9% were malignant tumors. In the benign tumors, the breast fibroadenoma and the anterior pituitary adenoma were the most common. The major malignant tumors were as follows: breast cancer, pituitary cell adenocarcinoma, leukemia, deputy lachrymal cancer. Meanwhile, the malignant tumor such as pituitary cell adenocarcinoma, histiocytic sarcoma, leukemia, bronchial adenocarcinoma, breast cancer transferred to the surrounding or the whole visceralorgan. In addition to the obvious spontaneous tumors, the atypical hyperplasia and altered hepatocyte foci were visible. Conclusion According to the result derived from the background data acquisition test of SD rats of our center , the occurrence probability of spontaneous tumors had increased after 30 weeks old and the sites of predilection for the tumor were pituitarium , breast , skin and subcutaneous tissue.Such asthepituitaryadenoma , breast fibroadenoma are the most common spontaneous tumors.The occurrence rate of the tumors in females was higher than males.At the same time , compared with the result from the different research institutions athome and abroad , the data had some differences.The different occurrence rates of the spontaneous tumors were considered to have a tight correlation of the individual difference and the feeding conditions.
    Isolation, Purification and Identification of Pathogenic Leptospirosis from Rodents in Jiangxi Province

    Objective Separation, purification and identification of Leptospira in Jiangxi Province will provide a source of bacteria for the later establishment of the pathogenesis model of Leptospira in Jiangxi. Method The strain was obtained by aseptically removing the kidney from the storage host of the Leptospira source, the kidney, and inoculating the kidney into the EMJH medium to culture and purify the strain.The purified Leptospira was tested by PCM with theoutermembranelipoprotein LipL32 gene primer to identify the strain , and the pathogenicity of the isolateto golden hamster was studied .Result A strain of Leptospira was isolated and purified by PCM amplification. The nucleotide sequence analysis confirmed that the sequence was 100% homologous to the Leptospira FDAARGOS _ 203 gene sequence ( GenBank accession number: CP020414. 2 ) . The isolate has certain pathogenicity to the golden hamster. Conclusion We successfully isolated a pathogenic strain of Leptospira from the kidney of the storage rat brown rat.This study has accumulated valuable experience for the separation and purification of Leptospira in the future.By collecting the popular Leptospira type in Jiangxi Province , it provides a source of bacteria for the pathogenesis model of Leptospira , and based on this , developed reliable clinical early diagnosis products , in order to achieve effective control of leptospirosis in Jiangxi Province , and reduce public health hazards.
    Effects of Yinchenhao Decoction on Caspase-12 Pathway and TIMP-1 and Smad2 in Rats with Hepatic Fibrosis

    Objective To investigate the effects of Yinchenhao Decoction on Caspase-12 Pathway and TIMP-1 and Smad2 in Rats with Hepatic Fibrosis. Method Forty-five male Sprague-Dawley rats were randomly divided into three groups, namely, control group, model group and Yinchenhaotang group. The LF model was established by intraperitoneal injection of CCl(. The Yinchenhao decoction group was intragastrically administered with Yinchenhao soup for 8 weeks. The general situation of rats in each group was compared after 8 weeks. Liver tissue morphology and fibrosis were observed by HE staining and Masson staining, respectively. Protein and mRNA levels were determined using Western blot and RT-qPCR, respectively. Result Rats in the model group developed significant liverdamageand fibrosis.Theresultofblood biochemistryshowed thatthelevelsofALTand ASTin themodel group were significantly higher than those in the control group ( P<0. 05 ) . The levels of ALT and AST after the intervention of Yinchenhaotang, decoction were significantly lower than those in the model group (P<0. 05 ) . Liver injury and fibrosis in the Yinchenhaotang group were significantly alleviated. GRP78, eIF2& and Caspase-12 were significantly increased in the liver tissue of the model group at 8 weeks after modeling (P<0. 01 ) . GRP78, eIF2a and Caspase-12 in Yinchenhaotang group were significantly lower than those in the model group (P<0.01 ) . The mRNA and protein levels of Collagen I, Collagen III, TIMP-1 and Smad2 in the model group were significantly higher than those in the control group (P<0. 01 ) , but the mRNA and protein levels of Collagen I, Collagen III, TIMP-1 and Smad2 in Yinchenhao Decoction group were significantly lowerthan those in the modelgroup (P<0.01) . Conclusion Yinchenhao Decoction can significantly alleviate the fibrosis of rat liver tissue, which maybe related to the inhibition ofCaspase-12 pathway and the expression ofSmad2 gene in anti-fibrosis and anti-apoptosis.
    Effect of High Fat Diet on Glycolipid Metabolism and Hepatic Steatosis in Farnesoid X Receptor (FXR) Knockout Mice

    Objective To investigate the effects of high fat diet on glycolipid metabolism and hepatic steatosis in farnesoid X receptor ( FXR) knockout mice. Method Normal diet ( ND ) group : C57BL/6 ( wild type, WT) mice (n = 6 ) and FXR 7 mice ( n = 6) were given normal diet for 12 weeks. High fat diet ( HFD ) group : C57BL/6mice (n = 6) and FXR / mice ( n = 6 ) were given 45% high fat diet for 12 weeks. After the mice were sacrificed, serum total cholesterol ( TC ) , triglyceride ( TG) , low density lipoprotein cholesterol ( LDL-C ) , high density lipoprotein cholesterol ( HDL-C ) , alanine aminotransferase ( ALT) , aspartate aminotransferase ( AST) and total bile acid (TBA) indicators were detected by automatic biochemical analyzer. The expression levels of liver inflammatory genes TNF-a, TLR4 and the downstream genes of FXR small heterodimer partner ( SHP ) , cholesterol 7 &-hydroxylase ( CYP7A1 ) were determined by RT-PCR. Liver steatosis was observed by HE staining. Result There was no difference in body weight between C57BL/6 mice and FXR / mice under high fat diet, but FXR / mice showed more severe glucose tolerance than C57BL/6 mice ( P<0. 01) , lipid metabolism disorder ( P<0. 01 ), serum bile acid increase (1<0. 01) , liver inflammation (1<0. 01) and hepatic steatosis. Conclusion Deletion of FXR caused disordersofglucoseand lipid metabolism , abnormalbileacid metabolism and hepaticsteatosisin mice , but that required induction of high fat diet.

And there is no mouse-specific enema equipment on the market, so it is urgently needed to prepare suitable mouse enema equipment. The need of study to prepare inflammatory bowel disease model, making anal appliance and anal plug by myself for enema in mice through exploring and summarizing during the experiment. The device has the advantages of simple production , economic cost, the obvious effect and low fatality rate, it can be widely used in the experimental study of mouse enema.
    Progress in Research of Diabetic Nephropathy Animal Model

    Diabetic nephropathy ( DN) is the most common complication of diabetes mellitus, and the leading cause of death in patients with end-stage renal disease and diabetes mellitus. At present, DN development is prevented mainly by controlling blood glucose and blood lipids and lowering blood pressure. Because of the complicated etiology of DN , thecurrenttreatmentmethod failed toreach thedesired goal.Therefore , new interventions or treatments are urgently needed to delay the progression of diabeticnephropathy. Animal models are of great value in analyzing the pathogenesis of diseases and testing new therapies, and ideal animal models are capable of investigating the pathogenesis of diabetic nephropathy , determining drug targetsand verifying new therapeutic strategies. However, there is a lack of animal models that can simulate the main characteristics of human DN in the field of DN research at present. In this paper , we discussed the progress , limitations and improvement of DN animal models in order to replicate ideal animal models similar to the main clinical and molecular phenotypes of human DN in the future.

    The intestinal flora is a large and complex micro-ecological system, and it is also a key factor in maintaining the dynamic balance of the body. This system is now becoming a new target for disease prevention , diagnosisand treatment. Sterile animals provide an ideal animal model for the study of interactions between host and microorganisms and between microorganisms.By inoculating sterile animals with specific single ormultiple microorganisms , we can see their significance in the focused study of intestinal flora. In this paper , the characteristics and corresponding research applications of several commonly used sterile animals as experimental models were reviewed , and the application value of aseptic animals as model animals in the study of intestinal flora was analyzed.
