Abstract: Abstract: Objective The effects of BAY and DMSO on acrosome reaction in vitro in mice spermatozoa were investigated. Method The influence of both chemicals on AR in vitro in mice by using 6 different concentrations of DMSO and 5 different concentrations of BAY for 5 duration time were invesgigated. The AR was evaluated by FITC-PSA and Coomassie brilliant blue stain. Result Both sperm AR rate and survival rate varied with could be affected by DMSO within a certain concentration and time,while the BAY had litter influence on them with all measured concentration ( P > 0. 05) . Conclusion The results showed that the BAY and DMSO had different pattern on sperms. DMSO has a direct effect on both the AR rate and survival rate,while the influence of BAY on the AR rate and the survival rate is neglectable. Thus a proper usage of DMSO in manipulation of sperm should be carefully reconsidered.

摘要: 摘要: 目的探究BAY( 激素敏感脂酶抑制剂) 及DMSO( 二甲基亚砜) 对小鼠精子顶体反应( AR) 的直接影响。方 法分别通过5 个和6 个不同的浓度梯度的BAY 及DMSO 在体外探究其对小鼠精子AR 的影响,在不同孵育时间 段采用FITC-PSA 和考马斯亮蓝染色法对小鼠精子的顶体反应进行鉴定。结果在所选浓度范围内BAY 未能直接 影响精子的顶体反应及存活率( P > 0. 05) ,而DMSO 在一定浓度和时间内可直接影响精子顶体反应及精子存活率。 结论DMSO 和BAY 对小鼠精子发生的作用是通过不同途径来实现的,前者可直接影响精子顶体反应和精子的存 活率,而后者则与顶体反应及存活率无关。提示在AR 的研究中使用DMSO 时需控制好作用浓度及作用时间。
