Abstract: Abstract: Objective To establish transgenic mice brain-specifically expressing insulin-like growth factor -1 ( Insulin-Like Growth Factors,IGF-1 ) . Method Sperm was used for gene transduction as a carrier,postnatal mice were detected by PCR assay to establish transgenic mouse lines. Western blot and immunofluorescence were applied to detect the IGF-1 expression level in transgenic mouse subgranular zone( SGZ) and the amount of reporter gene EGFP positive cells. Result Three positive mice were obtained,2 of them could be inherited stably,and established transgenic mouse lines. The IGF-1expression level in transgenic mouse SGZ was higher than that in the normal mice,meanwhile,EGFP could be expressed in the same area. Conclusion IGF-1 transgenic mice were prepared successfully.

摘要: 摘要: 目的制备脑组织特异性表达胰岛素样生长因子-1( Insulin-Like Growth Factors 1,IGF-1) 转基因小鼠。方法 采用精子为载体法进行基因转导,出生后小鼠PCR 检测,建立转基因小鼠系,用Western blot 和免疫荧光法分别 检测转基因小鼠海马齿状回亚粒状区( subgranular zone ,SGZ) IGF-1 表达量和报告基因EGFP 阳性细胞数。结果 获得3 只阳性小鼠,2只外源基因能稳定遗传,并建立了转基因小鼠系,转基因鼠SGZ 区域IGF-1 表达量显著高 于正常鼠,EGFP 也在此区域表达。结论成功制备IGF-1 转基因小鼠。
