微量细胞毒法在近交系小鼠遗传检测中的应用

Abstract: In order to establish a method of microcytotoxicity test and to examine the genetic characteristics of the inbred strain mouse, the inbred strain mouse's spleen was taken out,and put in the foreign monoclonal antibodies against H_2 antigens of inbred strain mouse,the death rate of the spleen cells was observed by inverted microscope.If the percentage of the obituary cells was more than 60%,then you could be sure of acquiring the genotype of the inbred strain mouse.So we can using microcytotoxicity test to identify H_2 haploid.The results showed that microcytotoxicity test is efficient,simple and acurrate.

摘要: 建立微量细胞毒实验检测技术 ,对近交系实验动物免疫遗传特性进行常规检测和质量监测。引进小鼠主要组织相容性复合体 (MHC)国际标准单克隆抗体系列 ,制取近交系小鼠脾细胞 ,加入特异的单克隆抗体 ,通过倒置显微镜观察最终反应结果 ,根据死亡细胞占总细胞的百分比来判定实验结果。死亡细胞超过细胞总数的 6 0 %即为阳性。可依照加入的单克隆抗体基因型来确定近交系小鼠的基因型 ,因此可利用微量细胞毒法识别近交系小鼠H- 2单倍型 ,区别不同的品系。此方法检测速度快、操作简单、结果准确 ,故便于在国内推广使用
