肥胖、高脂血症性脂肪性肝炎模型的建立

Abstract: Objective To establish a Non\|alcohlic steato\|hepatitis animal model. METHODS Male SD rats were fed with a high\|fat diet for 12 weeks which consisting of the standard diet, 10% lard and 2% cholesterol. After that then the serum lipid, aminotransferase values were detected and with histologically examination. RESULT All model group rats developed NASH which is characterized by significant obesity, elevated serum total cholesterol, free fatty acid and aminotransferase activities with histologically showed hepatocellular macrovesicular steatosis, lobular and portal inflammatory cell infiltration and necrosis. The incidence of NASH in rats with high\|fat diet is 100%. CONCLUSION A new NASH animal model was successfully developed in rats chronically fed with high\|fat diet in 3 months.

摘要: 为建立大鼠肥胖、高脂血症性脂肪性肝炎 (NASH)的动物模型。取雄性SD大鼠 1 9只 ,随机分为模型组 (n =1 0 )和正常组 (n =9)。模型组喂高脂饮食 ,即普通饲料基础上加 1 0 %猪油 ,2 %胆固醇。正常组喂普通饲料。观察一般情况及血清肝功能、血脂 ,并行病理学检查。模型组体重、肝指数、血清总胆固醇、游离脂肪酸及转氨酶均显著高于正常组 ;病理学均出现弥漫性肝细胞脂肪变性 ,且 1 0 0 %存在小叶内炎症 ,80 %出现汇管区炎症 ,并有 2例出现碎屑样坏死。电镜示肝细胞索排列紊乱 ,胞浆脂滴大量出现、聚积 ,线粒体嵴肿胀 ,肝星状细胞和Kupffer细胞增生。通过 3个月的高脂肪、高胆固醇饮食成功地复制了大鼠NASH模型
